Abstract

The present invention relates to a method for preparing a library of template polynucleotides and use thereof in methods of solid-phase nucleic acid amplification. More specifically, the invention relates to a method for preparing a library of template polynucleotides that have common sequences at their 5′ ends and at their 3′ ends.

Claims (7)

1. A library of different target nucleic acids comprising a plurality of different polynucleotide duplexes having polynucleotide adapters at each end;

wherein polynucleotide adapters are forked adapters formed by annealing of partially complementary first and second polynucleotide strands;

wherein the first polynucleotide strand comprises the sequence set forth in SEQ ID NO: 1; and

wherein the second polynucleotide strand comprises the sequence set forth in SEQ ID NO: 2.

The library of claim 1 , wherein a sequence of at least 10 consecutive nucleotides at the 5′ end of the first strand of the forked adapters and a sequence of at least 10 consecutive nucleotides at the 3′ end of the second strand of the forked adapters are not complementary such that an unmatched region of at least 10 consecutive nucleotides on each strand remains in single stranded form when the double-stranded region is annealed.

3. The library of claim 1 , wherein the library of different template polynucleotide molecules comprises sequences of a whole complex polynucleotide sample.

4. The library of claim 3 , wherein the different target polynucleotide duplexes are fragments of genomic DNA.
