Abstract

The present invention relates to a composition for controlling viability of a tissue including a potassium channel opener or adenosine receptor agonist, a compound for inducing local anaesthesia and compound for reducing the uptake of water by a cell in the tissue. The present invention also relates to the use of the composition according to the invention for controlling viability of a tissue.

30 . A method of controlling viability of an explanted tissue or organ including the step of contacting the tissue or organ with a composition including: a potassium channel opener or adenosine receptor agonist; a local anesthetic; and a sodium hydrogen exchange inhibitor.

31 . The method of claim 30 , wherein the sodium hydrogen exchange inhibitor is selected from the group consisting of: amiloride, EIPA, cariporide, eniporide, triamterene, EMD 84021, EMD 94309, EMD 96785, EMD 85131, HOE 694, BII B-513 and T-162559.

32 . The method of claim 30 , wherein the sodium hydrogen exchange inhibitor is amiloride.

33 . The method of claim 30 , wherein the concentration of the sodium hydrogen exchange inhibitor is between 1 nM to 1 mM.

The method of claim 30 , wherein the composition further includes at least one compound selected from the group consisting of: a compound for reducing the uptake of water by a cell in the tissue; diazoxide; a source of magnesium in a concentration of between 0.5 mM to 20 mM; and a source of calcium in a concentration of between 0.1 mM to 2.5 mM.

A method for controlling viability of an explanted tissue or organ including the step of contacting the tissue or organ with a composition including: a first compound comprising a potassium channel opener or adenosine receptor agonist; a second compound comprising a local anesthetic; and a third compound comprising a sodium hydrogen exchange inhibitor, wherein each of the three compounds is different.

36 . The method of claim 35 , wherein the sodium hydrogen exchange inhibitor is selected from the group consisting of amiloride, EIPA, cariporide, eniporide, triamterene, EMD 84021, EMD 94309, EMD 96785, EMD 85131, HOE 694, BII B-513 and T-162559.

37 . The method of claim 35 , wherein the sodium hydrogen exchange inhibitor is amiloride.

38 . The method of claim 35 , wherein the concentration of the compound is between 1 nM to 1 mM.

The method of claim 35 , wherein the composition further includes at least one compound selected from the group consisting of: a compound for reducing the uptake of water by a cell in the tissue; diazoxide; a source of magnesium in a concentration of between 0.5 mM to 20 mM; and a source of calcium in a concentration of between 0.1 mM to 2.5 mM.

40 . The method of claim 30 , wherein the composition further includes melatonin.

41 . The method of claim 30 , wherein the composition further includes insulin.
