Abstract

A container closure delivery system that is suitable for lyophilized pharmaceutical injectable powder products is disclosed. The system comprises storage stable powder formulations and a container closure assembly design wherein the formulation can be filled and lyophilized with a standard fill finish equipment, and the formulations and lyophilization processes are optimized to produce a powder that readily dissolves upon contact with a diluent, thereby facilitating the direct injection of the lyophilized product without the need for a separate reconstitution/mixing/priming step.

Claims (25)

1. A method of dispensing of a lyophilized pharmaceutical product, the method comprising the step of forcing a diluent through a container closure delivery system comprising the lyophilized pharmaceutical product in order to contact the diluent with the lyophilized pharmaceutical product in a manner that causes rapid dissolving of the lyophilized pharmaceutical product,

wherein the dissolved pharmaceutical product exits the container closure delivery system in an essentially constant gradient with more highly concentrated dissolved pharmaceutical product exiting first, and

wherein the method does not require a separate reconstitution, mixing, and/or priming step to dissolve the lyophilized pharmaceutical product with the diluent.

The method according to claim 1 , wherein the lyophilized pharmaceutical product comprises a small molecule, a vaccine, an oligonucleotide, a peptide, a protein, or a natural, synthetic, semi-synthetic, recombinant, or derivative thereof, useful for prophylactic, therapeutic or diagnostic application.

3. The method according to claim 2 , wherein the protein is an antibody, a hormone, a cytokine, a hematopoietic factor, a growth factor, an anti-obesity factor, a trophic factor, an anti-inflammatory factor, or an enzyme.

4. The method according to claim 1 , wherein the lyophilized pharmaceutical product further comprises a stabilizer, a bulking agent, a surfactant and/or a buffer.

5. The method according to claim 1 , wherein the lyophilized pharmaceutical product has a final solid content of 95% of the pharmaceutical product and 5% of the stabilizer.

6. The method according to claim 1 , wherein the diluent comprises water, a buffering agent, a surfactant, a stabilizer, a tonicity modifier, a metal ion, a bacteriostatic agent, an anesthetic agent, and/or a hydrogel.

7. The method according to claim 1 , wherein the diluent comprises water for injection (WFI), bacteriostatic water for injection (BWFI), or phosphate buffered saline (PBS).

8. The method according to claim 1 , wherein the diluent is delivered into the container closure delivery system by attaching a syringe filled with the diluent to a top end of the container closure delivery system configured to attach a syringe.

9. The method according to claim 1 , wherein a detachable tab seal on top end of the container closure delivery system is removed to define a fluid transfer channel that facilitates flow of the diluent through into the container closure delivery system.

10. The method according to claim 9 , wherein removal of the detachable tab seal forms an attachment for a syringe.

11. The method according to claim 1 , wherein the dissolved pharmaceutical product exits the container closure delivery system through an ejection port.

12. The method according to claim 11 , wherein the ejection port comprises a luer-lock needle, a luer-slip needle, a staked needle, a nozzle spread tip, or a blunt tip.

13. The method according to claim 1 , wherein the dissolved pharmaceutical product exits in an essentially linear concentration gradient with more highly concentrated dissolved pharmaceutical product exiting first.

14. The method according to claim 1 , wherein prior to forcing a diluent through the container closure delivery system, a detachable base is removed from the container closure delivery system to expose the ejection port.

15. The method according to claim 1 , wherein the container closure delivery system comprises:

a) a plunger assembly; and

b) a product container component defining a holding volume and comprising an upper base, a lower base and a mixing channel, wherein the upper base comprises an opening and the lower base comprises an ejection port; and wherein the mixing channel is integrated into the product container component at the lower base;

wherein the product container component is designed to accept the plunger assembly via the opening of the upper base.

16. The method according to claim 15 , wherein the plunger assembly is a one-piece plunger assembly.

17. The method according to claim 15 , wherein the plunger assembly comprises a top cup component and a plug component, wherein the top component is designed to engage the plug component.

The method according to claim 17 , wherein the top cup component is designed to comprise a top end configured to attach a syringe, a bottom end designed to comprise an opening, and one or more fluid transfer channels extending axially from the top end to the bottom end, wherein the top component engages the plug component via the opening of the bottom end.

The method according to claim 17 , wherein the plug component is designed to comprise a proximal end comprising an opening, a distal end comprising a flat surface, and one or more fluid transfer channels extending axially from the proximal end to the distal end, wherein the proximal end of the plug component engages the top component.

20. The method according to claim 17 , wherein the mixing channel is a spiral mixing channel.
