Abstract

This invention relates generally to the THAP1 gene and mutations in this gene, as well as the THAP1 protein and mutations in this protein, that are associated with dystonia. The invention relates to the identification, isolation, cloning and characterization of the DNA sequence corresponding to the wild type and mutant THAP1 genes, as well as isolation and characterization of their transcripts and gene products. The invention further relates to methods and kits useful for detecting mutations in THAP1 that are associated with dystonia, as well as to methods and kits useful for diagnosing dystonia. The present invention also relates to therapies for treating dystonia, including gene therapeutics and protein/antibody based therapeutics.

Claims (12)

1. A method for detecting the presence of a THAP1 mutation in a biological sample from a human subject, comprising:

(a) obtaining a biological sample from a human subject;

(b) contacting a THAP1 nucleic acid coding sequence in a biological sample with an oligonucleotide that specifically hybridizes to a human THAP1 nucleic acid sequence comprising a c.86G>C mutation of SED ID NO:4. and

(c) detecting hybridization of the THAP1 nucleic acid with the oligonucleotide, wherein detecting hybridization is indicative of a mutation of THAP1 nucleic acid in a biological sample from a human subject and wherein the mutation is c86G>C of SEQ ID NO:4.

The method according to claim 1 , wherein the method further comprises contacting genomic DNA obtained from the biological sample from the subject or cDNA produced from RNA obtained from the biological sample from the subject with a primer pair that amplifies all or a part of the THAP1 coding sequence.

3. The method of claim 2 , wherein the primer pair is SEQ ID NO: 22 and SEQ ID NO: 23.

4. The method of claim 2 , wherein the amplifying is performed by PCR or real-time PCR.

5. The method of claim 2 , further comprising digesting the amplified genomic DNA or cDNA with at least one restriction enzyme and comparing the restriction fragments of the amplified product from the subject with restriction fragments obtained from an amplification of a normal control sample.

6. The method of claim 1 , wherein the oligonucleotide is detectably labeled.

7. The method of claim 6 , wherein the oligonucleotide is detectably labeled with a radioisotope, fluorescent compound, enzyme, and/or enzyme co-factor.

The method of claim 1 , further comprising contacting a THAP1 nucleic acid with an oligonucleotide that specifically hybridizes to a human THAP1 nucleic acid sequence comprising a mutation selected from the group consisting of c.241T>C, c.266A>G, c.115G>A, c.460delC, c.134_135insGGGTT;137_139delAAC mutation, c.161G>A, c.1A>G, c.61T>A, c.67C>T, c.36C>A, c.2delT, c.65T>C, c.140C>T, c.392-394delTTT, c.11C>T, c.580T>C, c.424A>G, c.250-251delAC, and c.505C>T of SEQ ID NO: 4.

9. The method of claim 1 , wherein the oligonucleotide is attached to a solid support.
