Abstract

The invention relates to the treatment and prevention of cancer by administering agents that inhibit the activity of microRNAs that modulate tumor suppressor genes, which can include PTEN, p53, and INPP4B, among others. Inhibitors can include oligonucleotides that are at least partially complementary to these miRNAs. In some embodiments, these inhibitors are chemically modified oliognucleotides, including locked nucleic acids (LNAs).

Claims (7)

1. A method of treating melanoma in a subject in need thereof, comprising administering to the subject an inhibitor of an miRNA,

wherein the miRNA is selected from miR-23a (SEQ ID NO: 13) and miR-23* (SEQ ID NO: 67), and the miRNA regulates and reduces the protein levels of both PTEN and p53, thereby reducing tumor growth.

2. The method of claim 1 , wherein the inhibitor is an antisense oligonucleotide.

3. The method of claim 1 , wherein the inhibitor is at least partially complementary to an miRNA selected from miR-23a (SEQ ID NO: 13), or miR-23*(SEQ ID NO: 67).

4. The method of claim 1 , wherein the inhibitor is chemically modified, and the chemical modification is selected from LNA, phosphorothioate, 2′-O-Methyl, and 2′-O-Methoxyethyl.

5. The method of claim 4 , wherein the LNA comprises 16 or fewer nucleotides.

6. The method of claim 1 , wherein the melanoma is metastatic or non-metastatic.
