Pertussis antibodies and uses thereof
View Patent ↗Compositions and methods are provided that are useful to treat respiratory diseases such as whooping cough. Further, compositions and methods of immunizing are provided.
1. A humanized mouse 1B7 antibody capable of binding a pertussis toxin protein, the humanized antibody comprising a humanized heavy chain and a humanized light chain, wherein said humanized light chain comprises a phenylalanine at a position corresponding to Kabat position 65, a mouse CDR L1 as set forth in SEQ ID NO:3, a mouse CDR L2 as set forth in SEQ ID NO:4, and a mouse CDR L3 as set forth in SEQ ID NO:5; and
wherein said humanized heavy chain comprises a mouse CDR H1 as set forth in SEQ ID NO:6, a mouse CDR H2 as set forth in SEQ ID NO:7 and a mouse CDR H3 as set forth in SEQ ID NO:8.
2. The humanized mouse 1B7 antibody of claim 1 , wherein the antibody is a Fab′ fragment.
3. The humanized mouse 1B7 antibody of claim 1 , wherein the antibody comprises a human constant region.
4. The humanized mouse 1B7 antibody of claim 1 , wherein the antibody is an IgG.
5. The humanized mouse 1B7 antibody of claim 1 , wherein the antibody is a single chain antibody (scFv).
6. The humanized mouse 1B7 antibody of claim 1 , wherein the antibody is capable of binding said pertussis toxin protein with an equilibrium dissociation constant (K D ) less than 10 nM.
7. The humanized mouse 1B7 antibody of claim 6 , wherein said pertussis toxin protein is a truncated pertussis toxin protein.
8. The humanized mouse 1B7 antibody of claim 7 , wherein said truncated pertussis toxin protein comprises the amino acid sequence corresponding to amino acid residues 1 to 220 of SEQ ID NO:9.
9. The humanized mouse 1B7 antibody of claim 6 , wherein the humanized antibody is an antibody which competes with an antibody that is capable of binding a pertussis toxin protein comprising the amino acid sequence corresponding to amino acid residues 1 to 220 of SEQ ID NO:9.
10. A pharmaceutical composition comprising a therapeutically effective amount of a humanized mouse 1B7 antibody as set forth in claim 1 .
11. A method of treating whooping cough in a subject in need thereof, comprising administering to the subject a therapeutically effective amount of a humanized mouse 1B7 antibody as set forth in claim 1 .
