Polypeptides having xylanase activity and polynucleotides encoding same
The present invention relates to isolated polypeptides having xylanase activity and isolated polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides.
1. A process for degrading a xylan-containing material, the process comprising:
(i) treating a xylan-containing material with a composition comprising a polypeptide having xylanase activity, and
(ii) recovering the degraded xylan-containing material;
wherein the polypeptide having xylanase activity is selected from the group consisting of:
(a) a polypeptide having at least 95% sequence identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4;
(b) a polypeptide is encoded by a polynucleotide that hybridizes under very high stringency conditions with (i) nucleotides 58 to 802 of SEQ ID NO: 3, (ii) a cDNA sequence thereof, or (iii) the full-length complement of (i) or (ii), wherein very high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 μg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 70° C.; and
(c) a fragment of the sequence of amino acids 20 to 248 of SEQ ID NO: 4 which has xylanase activity.
2. A process for degrading a cellulosic material, the process comprising:
(i) treating a cellulosic material with a composition comprising a polypeptide having xylanase activity, and
(ii) recovering the degraded cellulosic material;
wherein the polypeptide having xylanase activity is selected from the group consisting of:
(a) a polypeptide having at least 95% sequence identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4;
(b) a polypeptide is encoded by a polynucleotide that hybridizes under very high stringency conditions with (i) nucleotides 58 to 802 of SEQ ID NO: 3, (ii) a cDNA sequence thereof, or (iii) the full-length complement of (i) or (ii), wherein very high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 μg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 70° C.; and
(c) a fragment of the sequence of amino acids 20 to 248 of SEQ ID NO: 4 which has xylanase activity.
3. A process for producing a fermentation product, said process comprising:
(i) saccharifying a cellulosic material or xylan-containing material with a composition comprising a polypeptide having xylanase activity,
(ii) fermenting the saccharified cellulosic material or xylan-containing material with one or more fermenting microorganisms to produce the fermentation product; and
(iii) recovering the fermentation product from the fermentation;
wherein the polypeptide having xylanase activity is selected from the group consisting of:
(a) a polypeptide having at least 95% sequence identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4;
(b) a polypeptide is encoded by a polynucleotide that hybridizes under very high stringency conditions with (i) nucleotides 58 to 802 of SEQ ID NO: 3, (ii) a cDNA sequence thereof, or (iii) the full-length complement of (i) or (ii), wherein very high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 μg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 70° C.; and
(c) a fragment of the sequence of amino acids 20 to 248 of SEQ ID NO: 4 which has xylanase activity.
4. The process of claim 3 , wherein the polypeptide having xylanase activity has at least 95% identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4.
5. The process of claim 3 , wherein the polypeptide having xylanase activity has at least 96% identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4.
6. The process of claim 3 , wherein the polypeptide having xylanase activity has at least 97% identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4.
7. The process of claim 3 , wherein the polypeptide having xylanase activity has at least 98% identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4.
8. The process of claim 3 , wherein the polypeptide having xylanase activity has at least 99% identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4.

The process of claim 3 , wherein the polypeptide having xylanase activity is encoded by a polynucleotide that hybridizes under very high stringency conditions with (i) nucleotides 58 to 802 of SEQ ID NO: 3, (ii) a cDNA sequence thereof, or (iii) the full-length complement of (i) or (ii), wherein very high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 μg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 70° C.
10. The process of claim 3 , wherein the polypeptide having xylanase activity is encoded by a polynucleotide having at least 95% sequence identity to the sequence of nucleotides 58 to 802 of SEQ ID NO: 3 or to the cDNA sequence thereof.
11. The process of claim 3 , wherein the polypeptide having xylanase activity comprises the sequence of amino acids 20 to 248 of SEQ ID NO: 4.
12. The process of claim 3 , wherein the polypeptide having xylanase activity is a fragment of the sequence of SEQ ID NO: 4 and wherein the fragment has xylanase activity.
13. A recombinant host cell comprising a nucleic acid construct or recombinant expression vector comprising a polynucleotide encoding a polypeptide having xylanase activity, wherein the polynucleotide is operably linked to one or more control sequences that direct the production of the polypeptide in an expression host and wherein the polypeptide having xylanase activity is selected from the group consisting of:
(a) a polypeptide having at least 95% sequence identity to the sequence of amino acids 20 to 248 of SEQ ID NO: 4;
(b) a polypeptide is encoded by a polynucleotide that hybridizes under very high stringency conditions with (i) nucleotides 58 to 802 of SEQ ID NO: 3, (ii) a cDNA sequence thereof, or (iii) the full-length complement of (i) or (ii), wherein very high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 μg/ml sheared and denatured salmon sperm DNA, and 50% formamide and washing three times each for 15 minutes using 2×SSC, 0.2% SDS at 70° C.; and
(c) a fragment of the sequence of amino acids 20 to 248 of SEQ ID NO: 4 which has xylanase activity.
14. A method of producing a polypeptide having xylanase activity, the method comprising:
(a) cultivating the recombinant host cell of claim 13 under conditions conducive for production of the polypeptide; and
(b) recovering the polypeptide.
15. A nucleic acid construct or recombinant expression vector comprising a heterologous nucleic acid encoding a protein operably linked to an isolated polynucleotide encoding a signal peptide comprising amino acids 1 to 19 of SEQ ID NO: 4.
16. A recombinant host cell comprising the nucleic acid construct or recombinant expression vector of claim 15 .
17. A process of producing a protein, said process comprising:
(a) cultivating the recombinant host cell of claim 16 under conditions conducive for production of the protein; and
(b) recovering the protein.
