Abstract

The invention features a series of heterocyclic derivatives that inhibit tumor necrosis factor alpha (TNF-α) induced necroptosis. The heterocyclic compounds of the invention are described by Formulas (I) and (Ia)-(Ie) and are shown to inhibit TNF-α induced necroptosis in FADD-deficient variant of human Jurkat T cells. The invention further features pharmaceutical compositions featuring the compounds of the invention. The compounds and compositions of the invention may also be used to treat disorders where necroptosis is likely to play a substantial role.

3. A method of treating necrosis or necroptosis in a subject having a condition selected from stroke, myocardial infarction, organ ischemia, traumatic brain injury, liver disease, cancer chemo/radiation therapy-induced necrosis, acute necrotizing pancreatitis, and a neurodegenerative disease,

said method comprising the step of administering the compound of claim 1 , or any pharmaceutically acceptable salt thereof, or stereoisomer thereof, to said subject in a dosage sufficient to decrease said necroptosis or said necrosis.

4. The method of claim 3 , wherein said condition is a neurodegenerative disease.

5. The method of claim 3 , wherein said condition is stroke, liver disease, myocardial infarction, or retinal ischemia.

6. A method of treating necrosis or necroptosis in a subject having a condition selected from stroke, myocardial infarction, organ ischemia, traumatic brain injury, liver disease, cancer chemo/radiation therapy-induced necrosis, acute necrotizing pancreatitis, and a neurodegenerative disease,

said method comprising the step of administering a compound having the structure:

R 1 is isopropyl, cyclopropyl, or cyclobutyl;

R 7 is selected from hydrogen and an alkyl group;

each R 8 and R 12 is, independently, halogen;

R 9 is hydrogen or halogen;

or any pharmaceutically acceptable salt thereof, or stereoisomer thereof, wherein said compound is not

to said subject in a dosage sufficient to decrease said necroptosis or said necrosis.

7. The method of claim 1 , wherein said condition is a neurodegenerative disease.

8. The method of claim 1 , wherein said condition is stroke, liver disease, myocardial infarction, or retinal ischemia.

9. A method of treating necrosis or necroptosis in a subject having a condition selected from stroke, myocardial infarction, organ ischemia, traumatic brain injury, liver disease, cancer chemo/radiation therapy-induced necrosis, acute necrotizing pancreatitis, and a neurodegenerative disease,

said method comprising the step of administering the compound selected from the group consisting of

or any pharmaceutically acceptable salt thereof, or stereoisomer thereof, to said subject in a dosage sufficient to decrease said necroptosis or said necrosis.

10. The method of claim 9 , wherein said condition is a neurodegenerative disease.

11. The method of claim 9 , wherein said condition is stroke, liver disease, myocardial infarction, or retinal ischemia.
