Abstract

Methods and apparatus for obtaining an image of light scattering biological tissue. A series of pulses of substantially monochromatic light of a first wavelength is generated and split into two parts, of which one part illuminates a scattering biological tissue at an intensity too low to damage the tissue, while a second part is upconverted to generate a pump beam. Sample light from the biological tissue, which may be scattered (or transmitted) light, or fluorescence, or Raman scattering, etc., is collected and directed from the scattering biological tissue, along with the pump beam, into a non-linear optical element, in a single pass or multiple passes. Parametrically amplified sample light emerging from the non-linear optical element is detected and analyzed or displayed.

Claims (37)

1. A method for obtaining an image of scattering biological tissue, the method comprising:

a. generating a series of pulses of substantially monochromatic light of a first wavelength;

b. splitting each of the pulses into a first part and a second part;

c. converting the first part of each pulse into a white light supercontinuum;

d. illuminating the scattering biological tissue with the white light supercontinuum;

e. upconverting the second part of each pulse to a generate a pump beam;

f. collecting both ballistic and multiply scattered sample light from the biological tissue;

g. forming a virtual pinhole by directing both the ballistic and multiply scattered sample light from the scattering biological tissue and the pump beam onto a non-linear optical element; and

h. selectively detecting parametrically amplified ballistic sample light emerging from the non-linear optical element.

2. A method in accordance with claim 1 , further comprising imposing a temporal offset of the pump beam relative to the ballistic and multiply scattered sample light.

3. A method in accordance with claim 2 , further comprising temporally resolving the ballistic and multiply scattered sample light.

4. A method in accordance with claim 1 , wherein the pump beam is characterized by a wavelength that is a harmonic of the substantially monochromatic light of the first wavelength.

5. A method in accordance with claim 1 , further comprising enhancing spatial resolution based on selective amplification of coherently scattered light.

6. A method in accordance with claim 1 , wherein the ballistic and multiply scattered sample light is backscattered from the scattering biological tissue.

7. A method in accordance with claim 1 , wherein the ballistic and multiply scattered sample light is Raman scattered.

8. A method in accordance with claim 7 , wherein the ballistic and multiply scattered sample light is spontaneously Raman scattered.

9. A method in accordance with claim 1 , wherein the sample light is a fluorescent emission of the scattering biological tissue.

A method in accordance with claim 1 , wherein directing both the ballistic and multiply scattered sample light from the biological tissue and the pump beam onto the non-linear optical element includes multipassing both the ballistic and multiply scattered sample light from the scattering biological tissue and the pump beam through the non-linear optical element.

11. A method in accordance with claim 10 , wherein multiple passes of the ballistic and multiply scattered sample light and pump beam are differentially delayed.

12. A method in accordance with claim 10 , wherein multipassing includes at least three traversals of the non-linear optical element.

13. A method in accordance with claim 1 , wherein generating a series of pulses includes producing pulses at a rate exceeding 200 kHz.

14. A method in accordance with claim 1 , wherein parametric amplification is achieved by type-I (ooe) phase matching in the second non-linear element.

15. An apparatus for obtaining an image of scattering biological tissue, the apparatus comprising:

a. a source of pulses of substantially monochromatic light of a first wavelength;

b. a beamsplitter for splitting each of the pulses into a first part and a second part;

c. a spectral spreader for converting the first part of each pulse into a white light supercontinuum;

d. focusing optics for illuminating the scattering biological tissue with the white light supercontinuum;

e. a first non-linear optical element for upconverting the second part of each pulse to generate a pump beam;

f. collection optics for collecting ballistic and multiply scattered sample light emergent in a backwards direction from the biological tissue; and

g. a second non-linear optical element, characterized by an aperture-limiting virtual pinhole, for receiving the ballistic and multiply scattered sample light and the pump beam and parametrically amplifying the sample light;

h. a detector for receiving parametrically amplified sample light and generating a detector signal; and

i. a processor for generating an image of the scattering biological tissue on the basis of the detector signal.

16. An apparatus in accordance with claim 15 , wherein the source of pulses is a laser.

17. An apparatus in accordance with claim 15 wherein the source of pulses is a femtosecond laser.

18. An apparatus in accordance with claim 15 wherein the source of pulses is a picosecond laser.

19. An apparatus in accordance with claim 15 , wherein at least one of the first non-linear optical element and the second non-linear optical element is beta barium borate.

20. An apparatus in accordance with claim 15 , wherein the spectral spreader is a sapphire plate.
