Spiro-oxindole MDM2 antagonists

The compounds and compositions provided herein relate to spiro-oxindoles which function as antagonists of the interaction between p53 and MDM2, and their use as therapeutics for the treatment of cancer and other diseases.
1. A method of preparing:
the method comprising:
(a) dissolving:
in a solvent or a mixture of solvents; and
(b) allowing
to isomerize to
wherein the isomerization involves the formation of:
as an intermediate.
2. The method of claim 1 further comprising isolating
substantially free of one or more other stereoisomers.
3. The method of claim 1 , wherein the solvent or mixture of solvents is methanol/water in a 1:1 v/v ratio.
