Abstract

Embodiments are directed to compositions and methods of treating migraine and related neurological disorders. In certain aspects, methods and compositions are for reducing cortical spreading depression and/or suppressing the neurochemical basis for chronic and acute migraine events, and provide methods and pharmaceutical compositions related to both acute and preventive therapies for migraine events and related headaches. In particular embodiments, treatment involves IL-11, interferon-gamma, or an IGF inducer such as IGF-1 or insulin.

Claims (23)

1. A method for treating a migraine patient comprising administering to the patient a composition comprising IL-11.

2. The method of claim 1 , further comprising administering IGF-1 and/or insulin to the patient.

3. A method for treating a chronic and recurrent migraines in a patient comprising administering to the patient an effective amount of a composition comprising interferon (IFN)-γ.

4. The method of claim 3 , further comprising administering IGF-1 and/or insulin to the patient.

5. A method for treating chronic or recurrent migraines in a patient comprising administering to the patient one or more times an effective amount of a composition comprising at least one insulin growth factor receptor (IGFR) inducer, wherein the IGFR inducer is IGF-1 or insulin, wherein treating chronic or recurrent migraines in the patient does not include administering a cytokine to the patient.

6. The method of claim 5 , wherein the patient is suffering from a migraine headache.

7. The method of claim 5 , wherein the composition is administered to the patient intranasally.

8. The method of claim 5 , wherein the composition comprises between about 0.1 ng and about 2.0 g of IGF-1.

9. The method of claim 5 , wherein the patient is administered up to about 10 ml of the composition.

10. The method of claim 5 , further comprising administering the composition to the patient up to four times a day.

11. The method of claim 5 , wherein the patient is administered the composition every 4 to 8 hours.

12. The method of claim 5 , wherein the patient is administered the composition once a day.

13. The method of claim 5 , wherein the composition is administered for multiple days.

14. The method of claim 5 , wherein the composition is administered to the patient's brain cells.

15. The method of claim 5 , wherein the composition is administered to microglia, or immune cells and vascular cells in the patient's brain, or other cells in the patient's brain.

16. The method of claim 5 , further comprising administering one or more analgesics or antimigraine drugs, wherein the antimigraine drug is an ergot alkaloid, beta-blocking agent, calcium channel blocking agent, antidepressant, selective 5-HT 1 agonist, sedatives, local anesthetic, or adrenergic blocking agent.

17. The method of claim 5 , wherein microglia of the patient are treated with the IGF-1 inducer.

The method of claim 5 , wherein the method consists of administering to the patient one or more times an effective amount of a composition comprising at least one insulin growth factor receptor (IGFR) inducer, wherein the IGFR inducer is IGF-1 or insulin, and administering one or more analgesics or antimigraine drugs, wherein the antimigraine drug is an ergot alkaloid, beta-blocking agent, calcium channel blocking agent, antidepressant, selective 5-HT 1 agonist, sedatives, local anesthetic, or adrenergic blocking agent, wherein treating chronic or recurrent migraines in the patient does not include administering a cytokine to the patient.

19. The method of claim 5 , wherein the IGFR inducer is IGF-1.

20. The method of claim 19 , further comprising administering a second IGFR inducer, wherein the second IGFR inducer is insulin.

21. The method of claim 5 , wherein the patient is also administered an anti-migraine drug.

22. The method of claim 21 , wherein the composition includes the anti-migraine drug.

23. The method of claim 21 , wherein the patient is administered the anti-migraine drug within 24 hours of being administered the composition.
