Abstract

Dominant negative (DN) variants of transcriptional enhancer factor 1-related (RTEF-1) are described. DN RTEF-1 polypeptides may be directly targeted to cells or delivered in nucleic acid expression vectors to alter cellular transcription. Methods for inhibiting VEGF production and thereby treating angiogenic disorders such as cancer are described. For example, in certain aspects, DN RTEF-1 may be used to treat angiogenic disorders of the eye such as age related macular degeneration (AMD).

Claims (22)

1. An isolated cDNA comprising a sequence encoding a polypeptide that is at least 95% identical to SEQ ID NO:2; SEQ ID NO:3 or SEQ ID NO:4.

2. The cDNA of claim 1 , wherein the isolated cDNA is comprised within a nucleic acid expression cassette.

3. The cDNA of claim 2 , wherein the nucleic acid expression cassette is positioned within a viral expression vector.

4. The cDNA of claim 3 , wherein the viral expression vector is an adenovirus, adeno-associated virus, herpes virus, SV-40 virus, retrovirus or vaccinia virus vector.

5. The cDNA of claim 4 , wherein the viral expression vector is an adeno-associated virus.

6. The cDNA of claim 4 , wherein the viral expression vector is a lentiviral expression vector.

7. The cDNA of claim 6 , wherein the lentiviral expression vector is an HIV vector.

8. The cDNA of claim 7 , wherein the expression cassette comprises a cell type specific or inducible promoter.

9. The cDNA of claim 8 , wherein the inducible promoter is a hypoxia inducible promoter.

10. The cDNA of claim 8 , wherein the inducible promoter is an angiogenesis inducible promoter.

11. The cDNA of claim 1 , further comprising a second anti-angiogenesis gene.

12. The cDNA of claim 1 , further comprising a sequence encoding a secretion signal.

13. The cDNA of claim 1 , further comprising a sequence encoding a cell internalization moiety.

14. The cDNA of claim 13 , wherein the internalization moiety comprises internalization sequences from HIV tat, HSV-1 tegument protein VP22, or Drosophila antennopedia.

15. The cDNA of claim 13 , wherein the internalization moiety comprises a poly-arginine, poly-methionine and/or poly-glycine peptide.

16. The cDNA of claim 13 , wherein the internalization moiety comprises the amino acid sequence RMRRMRRMRR (SEQ ID NO:23).

17. The cDNA of claim 1 , further comprising sequences encoding a cell secretion signal and a cell internalization moiety.

18. The cDNA of claim 17 , wherein the cell secretion signal sequences comprises the human IL-2 secretion signal sequence (SEQ ID NO:22).

19. The cDNA of claim 1 , wherein the sequence is a sequence encoding a polypeptide that is at least 95% identical to SEQ ID NO:2.

20. The cDNA of claim 1 , wherein the sequence is a sequence encoding a polypeptide that is at least 95% identical to SEQ ID NO:3.

21. The cDNA of claim 1 , wherein the sequence is a sequence encoding a polypeptide that is at least 95% identical to SEQ ID NO:4.

22. A method for producing a RTEF-1 polypeptide comprising: introducing into a cell an exogenous nucleic acid encoding a polypeptide that is at least 95% identical to SEQ ID NO:2; SEQ ID NO:3 or SEQ ID NO:4, wherein said cell expresses said exogenous nucleic acid; and isolating the RTEF-1 polypeptide produced by the cell.
