Methods and systems for chemoautotrophic production of organic compounds
The present disclosure identifies pathways, mechanisms, systems and methods to confer chemoautotrophic production of carbon-based products of interest, such as sugars, alcohols, chemicals, amino acids, polymers, fatty acids and their derivatives, hydrocarbons, isoprenoids, and intermediates thereof, in organisms such that these organisms efficiently convert inorganic carbon to organic carbon-based products of interest using inorganic energy, such as formate, and in particular the use of organisms for the commercial production of various carbon-based products of interest.
1. An engineered cell comprising:
an energy conversion pathway comprising a formate dehydrogenase having an amino acid sequence of any one of SEQ ID NOs: 5-8, or an amino acid sequence encoded by any one of SEQ ID NOs: 1-4;
a carbon fixation pathway, wherein the carbon fixation pathway is a ribulose monophosphate cycle comprising a hexulose-6-phosphate synthase having Enzyme Classification No. E.C. 4.1.2.43; and
a lysine biosynthetic pathway comprising aspartate aminotransferase.

The engineered cell of claim 1 , wherein the carbon fixation pathway converts an inorganic carbon to a carbon-based central metabolite; wherein the inorganic carbon is formate, formic acid, formaldehyde, carbon dioxide, methane or methanol; and wherein the carbon-based central metabolite is fructose-6-phosphate.

The engineered cell of claim 1 , wherein the cell produces one or more of: L-aspartate, L-4-aspartyl phosphate, L-aspartate 4-semialdehyde, L-homoserine, (2S,4 S)-4-Hydroxy-2,3,4,5-tetrahydrodipicolinate, L-2,3,4,5-tetrahydrodipicolinate, N-acetyl-L-2-amino-6-oxopimelate, L-2-amino-6-oxopimelate, N-succinyl-L-2-amino-6-oxopimelate, N-acetyl-LL-2,6-diaminopimelate, LL-2,6-diaminopimelate, N-succinyl-LL-2,6-diaminopimelate, meso-2,6-diaminopimelate, and lysine.

The engineered cell of claim 1 , wherein the host cell is selected from the group consisting of Escherichia coli, Klebsiella oxytoca, Anaerobiospirillum succiniciproducens, Acetobacter aceti, Actinobacillus succinogenes, Mannheimia succiniciproducens, Mesoplasma florum, Rhizobium etli, Bacillus subtilis, Corynebacterium glutamicum, Gluconobacter oxydans, Zymomonas mobilis, Lactococcus lactis, Lactobacillus plantarum, Cupriavidus necator (formerly Ralstonia eutropha ), Streptomyces coelicolor, Clostridium ljungdahlii, Clostridium thermocellum, Clostridium acetobutylicum, Pseudomonas fluorescens, Pseudomonas putida, Saccharomyces cerevisiae, Schizosaccharomyces pombe, Kluyveromyces lactis, Kluyveromyces marxianus, Aspergillus terreus, Aspergillus niger, Penicillium chrysogenum and Pichia pastoris.
5. The engineered cell of claim 1 , wherein the host cell is Escherichia coli.
6. A method of producing a carbon-based product, comprising culturing the engineered cell of claim 1 .
