Abstract

This disclosure describes, in one aspect, a method for identifying β-glucan binding to immune cells of a subject. Generally, the method includes obtaining a blood sample from the subject, the blood sample comprising immune cells, adding soluble β-glucan to at least a portion of the blood sample and incubating the mixture under conditions allowing the soluble β-glucan to bind to the immune cells, and detecting soluble β-glucan bound to the immune cells. In another aspect, this disclosure describes a method that generally includes identifying the subject as a low binder of β-glucan, and co-administering to the subject a soluble β-glucan and an antibody preparation capable of converting the subject from a low binder to a high binder.

Claims (14)

1. A method for identifying soluble yeast β-glucan binding to immune cells of a subject to determine if the subject has low binder status, the method comprising:

obtaining a blood sample from the subject, the blood sample comprising immune cells comprising polymorphonuclear leukocytes (PMNs) or monocytes;

adding soluble yeast β-glucan to at least a portion of the blood sample and incubating the mixture under conditions allowing the soluble yeast β-glucan to bind to the immune cells;

detecting soluble yeast β-glucan bound to at least a portion of the PMNs or monocytes;

measuring the portion of PMNs or monocytes that bind soluble β-glucan as a percentage fraction of total PMNs or monocytes; and

identifying the subject as having high binder status when at least 10% of PMNs or monocytes in the sample bind soluble β-glucan after incubation in conditions allowing the soluble yeast β-glucan to bind to immune cells in the blood sample or having low binder status when the subject fails to exhibit high binder status.

2. The method of claim 1 , wherein the soluble yeast β-glucan comprises a β-1,3/1,6 glucan.

3. The method of claim 1 , wherein the soluble yeast β-glucan comprises β(1,6)-[poly-(1,3)-D-glucopyranosyl]-poly-β(1,3)-D-glucopyranose.

4. The method of claim 1 , wherein detecting soluble yeast β-glucan bound to the PMNs or monocytes comprises contacting the sample with a monoclonal antibody that specifically binds to the β-glucan.

5. The method of claim 1 , further comprising:

obtaining a second blood sample from the subject, the second blood sample comprising immune cells;

adding soluble yeast β-glucan to at least a portion of the second blood sample and incubating the mixture under conditions allowing the soluble yeast β-glucan to bind to the immune cells; and

detecting soluble yeast β-glucan bound to the immune cells.

6. The method of claim 1 , wherein the immune cells comprise neutrophils or monocytes.
