Abstract

The present invention is directed to secondary alcohol substituted triazole compounds which are useful as therapeutic agents for the treatment of central nervous system disorders associated with phosphodiesterase 10 (PDE10). The present invention also relates to the use of such compounds for treating neurological and psychiatric disorders, such as schizophrenia, psychosis or Huntington's disease, and those associated with striatal hypofunction or basal ganglia dysfunction.

Claims (73)

1. A compound of the formula I:

wherein:

A represents

R represents H or C 1-6 alkyl;

R 1 is selected from the groups consisting of (CH 2 ) n C 1-4 haloalkyl, C 1-6 alkyl, C 3-6 cycloalkyl, C 6-10 aryl, said alkyl cycloalkyl, and aryl is unsubstituted or substituted with 1 to 3 groups of R a ;

R 2 is selected from the group consisting of H, O—R, CN, O(CH 2 ) n OR, OCHR(CH 2 ) n OR, SR, SO 2 R, S(O)R, N(R) 2 , C(O)N(R) 2 , C 1-3 haloalkyl, O(CH 2 ) n C 1-3 haloalkyl, O(CH 2 ) n C 3-6 cycloalkyl, (CH 2 ) n C 5-10 heterocycle, C 1-6 alkyl, and C 3-10 cycloalkyl, said alkyl, cycloalkyl, and heterocycle is unsubstituted or substituted with 1 to 3 groups of R a ;

R a is selected from the group consisting of:

(1) halogen,

(2) hydroxyl,

(3) C 1-6 alkyl,

(4) —(CH 2 ) n O—R,

(5) (CH 2 ) n C 6-10 aryl,

(6) (CH 2 ) n C 5-10 heterocycle,

(7) OC 1-5 haloalkyl;

(8) CO 2 R;

(9) C(O)N(R) 2 ;

(10) (CH 2 ) n C(O)R;

(11) CN,

(12) (CH 2 ) n N(R) 2 ;

(13) (CH 2 ) n C 3-6 cycloalkyl,

(14) (CH 2 ) n C 1-3 haloalkyl;

n represents 0-4,

or a pharmaceutically acceptable salt thereof.

2. The compound according to claim 1 wherein A is

4. The compound according to claim 1 wherein R 1 is C 1-6 alkyl.

5. The compound according to claim 1 wherein R 1 is (CH 2 ) n C 1-4 haloalkyl.

6. The compound according to claim 1 wherein R 1 is optionally substituted C 6-10 aryl.

7. The compound according to claim 1 wherein R 1 is C 3-6 cycloalkyl.

8. The compound according to claim 1 represented by structural formula Ia:

or a pharmaceutically acceptable salt thereof, wherein R 1 is C1-6 alkyl, CF 3 , C6-10 aryl, or C3-6 cycloalkyl.

9. The compound according to claim 8 wherein R 1 is methyl, ethyl, propyl, CF 3 , phenyl, or cyclohexyl and R 2 is methyl, ethyl, propyl, CF 3 , or cyclopropyl.

10. The compound according to claim 1 represented by structural formula Iaa:

or a pharmaceutically acceptable salt thereof, wherein R 1 is C1-6 alkyl, CF 3 , C6-10 aryl, or C3-6 cycloalkyl.

11. The compound according to claim 10 wherein R 1 is methyl, ethyl, propyl, CF 3 , phenyl, or cyclohexyl and R 2 is methyl, ethyl, propyl, CF 3 , or cyclopropyl.

12. The compound according to claim 1 represented by structural formula Ib:

or a pharmaceutically acceptable salt thereof wherein R 1 is C1-6 alkyl, CF 3 , C6-10 aryl, or C3-6 cycloalkyl.

13. The compound according to claim 12 wherein R 1 is methyl, ethyl, propyl, CF 3 , phenyl, or cyclohexyl and R 2 is methyl, ethyl, propyl, CF 3 , or cyclopropyl.

14. A compound which is:

1-(3-methyl-1-(2-methyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

(S)-1-(3-methyl-1-(2-methyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

(R)-1-(3-methyl-1-(2-methyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl) ethanol;

1-(3-methyl-1-(6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)-2-(trifluoromethyl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

1-(1-(2-methoxy-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-3-methyl-1H-1,2,4-triazol-5-yl)ethanol;

1-(1-(2-cyclopropyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-3-methyl-1H-1,2,4-triazol-5-yl)ethanol;

2,2,2-trifluoro-1-(3-methyl-1-(2-methyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

(S)-2,2,2-trifluoro-1-(3-methyl-1-(2-methyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

(R)-2,2,2-trifluoro-1-(3-methyl-1-(2-methyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

2,2-difluoro-1-(3-methyl-1-(2-methyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

2-fluoro-1-(3-methyl-1-(2-methyl-6-(3-(1-methyl-1H-benzo[d]imidazol-2-yl)azetidin-1-yl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

(S)-1-(3-methyl-1-(6-((1R,2R)-2-(1-methyl-1H-benzo[d]imidazol-2-yl)cyclopropyl)-2-(trifluoromethyl)pyrimidin-4-yl)-1H-1,2,4-triazol-5-yl)ethanol;

1-(1-(2-cyclopropyl-6-((1R,2R)-2-(1-methyl-1H-benzo[d]imidazol-2-yl)cyclopropyl)pyrimidin-4-yl)-3-methyl-1H-1,2,4-triazol-5-yl)ethanol

(S)-1-(1-(2-cyclopropyl-6-((1R,2R)-2-(1-methyl-1H-benzo[d]imidazol-2-yl)cyclopropyl)pyrimidin-4-yl)-3-methyl-1H-1,2,4-triazol-5-yl)ethanol;

or a pharmaceutically acceptable salt thereof.

15. A pharmaceutical composition which comprises a pharmaceutically acceptable carrier and a compound of claim 1 or a pharmaceutically acceptable salt thereof.

16. A method for treating anxiety in a mammalian patient in need thereof which comprises administering to the patient a therapeutically effective amount of a compound of claim 1 or a pharmaceutically acceptable salt thereof.
