Abstract

A method for the storage of red blood cells provides for mixing an additive solution with packed red blood cells to create a suspension of red blood cells. The method further provides for reducing the oxygen in the suspension of red blood cells; and storing the suspension of red blood cells under oxygen-depleted storage.

Claims (27)

11 . A method of enhancing the ATP level in stored red blood cells comprising the steps of:

mixing an additive solution having a pH ranging from 5.5 to 6.5 with packed red blood cells creating a suspension of red blood cells;

reducing the oxygen in said suspension of red blood cells to approximately 10% or less of the level in whole blood to prepare an oxygen reduced suspension of red blood cells; and

storing said oxygen reduced suspension of red blood cells under oxygen-depleted storage conditions,

wherein said enhanced ATP level after 3 weeks of storage is at least 33% more than that of a suspension of red blood cells stored in the presence of oxygen.

12 . The method according to claim 11 , further comprising obtaining said packed red blood cells from whole blood.

The method according to claim 11 , wherein said additive solution is OFAS3 comprising the following concentrations of about 0.5 to 4.0 mmole/liter of adenine, about 50 to 150 mmole/liter of dextrose, about 20 to 70 mmole/liter of mannitol, about 0 to 100 mmole/liter of NaCl, and about 2 to 20 mmole/liter of NaH 2 PO 4 .

14 . The method according to claim 11 , wherein said additive solution has an adjusted pH of about 6.5.

15 . The method according to claim 11 , wherein suspension of red blood cells under oxygen-depleted storage are stored at a temperature of 4° C. Page 4

16 . The method according to claim 11 , further comprising one or more additions of a metabolic supplement to said suspension of stored red blood cells.

17 . The method according to claim 11 , wherein said metabolic supplement comprises pyruvate, inosine, and adenine.

18 . The method according to claim 17 , wherein said metabolic supplement further comprises a compound selected from the group consisting of dibasic sodium phosphate, monobasic sodium phosphate, and combinations thereof.

19 . The method according to claim 11 , wherein the additive solution is AS-3 or EAS61.

20 . A method of enhancing the 2,3-DPG level in a stored red blood cells comprising the steps of:

mixing an additive solution having a pH ranging from 5.5 to 6.5 with packed red blood cells creating a suspension of red blood cells;

reducing the oxygen in said suspension of red blood cells to approximately 10% or less of the level in whole blood to prepare an oxygen reduced suspension of red blood cells; and

storing said oxygen reduced suspension of red blood cells under oxygen-depleted storage conditions,

wherein said enhanced 2,3-DPG level is greater than that of a suspension of red blood cells stored in the presence of oxygen.

21 . The method according to claim 20 , further comprising obtaining said packed red blood cells from whole blood.

The method according to claim 20 , wherein said additive solution is OFAS3 comprising the following concentrations of about 0.5 to 4.0 mmole/liter of adenine, about 50 to 150 mmole/liter of dextrose, about 20 to 70 mmole/liter of mannitol, about 0 to 100 mmole/liter of NaCl, and about 2 to 20 mmole/liter of NaH 2 PO 4 .

23 . The method according to claim 20 , wherein said suspension of stored red blood cells are stored at a storage temperature of 4° C.

24 . The method according to claim 20 , further comprising one or more additions of a metabolic supplement to said suspension of stored red blood cells.

25 . The method according to claim 20 , wherein said metabolic supplement comprises pyruvate, inosine, and adenine.

26 . The method according to claim 25 , wherein said metabolic supplement further comprises a compound selected from the group consisting of dibasic sodium phosphate, monobasic sodium phosphate, and combinations thereof.

27 . The method according to claim 20 , wherein the additive solution is AS-3 or EAS61.

28 . The method according to claim 20 , wherein said enhanced level of 2,3 DPG in said oxygen reduced suspension of red blood cells under oxygen-depleted storage conditions is greater than 5 μmol 2,3 DPG/gram of hemoglobin.
