Abstract

Disclosed are methods of attenuating activity of the PME-1 gene. siRNAs or shRNAs are used to target against PME-1, thereby reducing the PME-1 mRNA. It is disclosed that the siRNAs or shRNAs targeted against PME-1 attenuate the epithelial to mesenchymal transition, thereby inhibit endometrial cancer and triple negative breast cancer development. A kit containing siRNA or shRNA reagents for attenuating the PME-1 gene expression is also disclosed.

Claims (13)

1. A method of treating a human subject suffering from triple negative breast cancer and is in need of treatment, comprising the steps of:

a) identifying a human subject having triple negative breast cancer and in need of treatment thereof; and

b) administering to said human subject an effective amount of RNAi targeted against PME-1 selected from the group consisting of siRNA and shRNA, wherein said RNAi reduces the gene expression of PME-1 in said triple negative breast cancer and thereby inhibits tumor growth of said triple negative breast cancer, and

wherein said RNAi having a target sequence selected from the group consisting of SEQ ID NO: 17, SEQ ID NO: 2 and SEQ ID NO: 3.

2. A method of treating a human subject suffering from triple negative breast cancer and is in need of treatment, comprising the steps of:

a) identifying a human subject having triple negative breast cancer and in need of treatment thereof; and

b) administering to said human subject an effective amount of RNAi targeted against PME-1 wherein said RNAi reduces the gene expression of PME-1 in said triple negative breast cancer and thereby inhibits tumor growth of said triple negative breast cancer, and

wherein said RNAi having a target sequence selected from the group consisting of SEQ ID NO: 8 and SEQ ID NO: 9.

A method of inhibiting tumor growth of a triple negative breast cancer cell, comprising the step of exposing a triple negative breast cancer cell with RNAi selected from the group consisting of siRNA and shRNA targeted against PME-1, wherein said RNAi reduces PME-1 gene expression and thereby inhibits tumor growth of said triple negative breast cancer cell, and

wherein said RNAi having a target sequence selected from the group consisting of SEQ ID NO: 17, SEQ ID NO: 2 and SEQ ID NO: 3.

A method of inhibiting tumor growth of a triple negative breast cancer cell, comprising the step of exposing a triple negative breast cancer cell with RNAi targeted against PME-1, wherein said RNAi reduces PME-1 gene expression and thereby inhibits tumor growth of said triple negative breast cancer cell, and

wherein said RNAi having a target sequence selected from the group consisting of SEQ ID NO: 8 and SEQ ID NO: 9.

5. A composition comprising shRNA contained in an adenoviral expression vector operably linked to said shRNA and a pharmaceutical acceptable excipient, wherein said shRNA having a target sequence consisting of SEQ ID NO: 2 or SEQ ID NO: 3.
