Compositions and methods for treating chronic inflammation and inflammatory diseases

1. A pharmaceutical composition comprising:

a) a therapeutically-effective amount of a pharmaceutically-acceptable form of a propionic acid derivative non-steroidal anti-inflammatory drug (NSAID);

b) about 8% (v/v) to about 12% (v/v) of a pharmaceutically-acceptable polyethylene glycol (PEG) polymer of less than about 2,000 g/mol; and

c) at least 60% (v/v) of a pharmaceutically-acceptable glycerolipid,

wherein the pharmaceutical composition is formulated to have a melting point temperature of about 25° C. or higher.

The pharmaceutical composition according to claim 1 , wherein the propionic acid derivative NSAID comprises a pharmaceutically-acceptable form of an Alminoprofen, a pharmaceutically-acceptable form of a Benoxaprofen, a pharmaceutically-acceptable form of a Dexketoprofen, a pharmaceutically-acceptable form of a Fenoprofen, a pharmaceutically-acceptable form of a Flurbiprofen, a pharmaceutically-acceptable form of an Ibuprofen, a pharmaceutically-acceptable form of an Indoprofen, a pharmaceutically-acceptable form of a Ketoprofen, a pharmaceutically-acceptable form of a Loxoprofen, a pharmaceutically-acceptable form of a Naproxen, a pharmaceutically-acceptable form of an Oxaprozin, a pharmaceutically-acceptable form of a Pranoprofen, or a pharmaceutically-acceptable form of a Suprofen.

3. The pharmaceutical composition according to claim 2 , wherein the propionic acid derivative NSAID is a pharmaceutically-acceptable form of an Ibuprofen.

4. The pharmaceutical composition according to claim 1 , wherein the PEG polymer comprises PEG 100, PEG 200, PEG 300, PEG 400, PEG 500, PEG 600, PEG 700, PEG 800, PEG 900, PEG 1000, or any combination thereof.

5. The pharmaceutical composition according to claim 1 , wherein the glycerolipid comprises a monoglyceride, an acetylated monoglyceride, a diglyceride, an acetylated diglyceride, a triglyceride, an acetylated triglyceride, or any combination thereof.

6. The pharmaceutical composition according to claim 5 , wherein the amount of the monoglyceride is at least 20% (v/v).

7. The pharmaceutical composition according to claim 5 , wherein the amount of the triglyceride is at least 35% (v/v).

8. A method of treating an individual with a chronic inflammation, the method comprising the step of: administering to the individual in need thereof a pharmaceutical composition according to claim 1 , wherein administration results in a reduction in a symptom associated with the chronic inflammation, thereby treating the individual.

9. The method according to claim 8 , wherein the chronic inflammation is a tissue inflammation or a systemic inflammation.

10. The method according to claim 8 , wherein the chronic inflammation is an auto-immune disease or a non-autoimmune disease.

11. The method according to claim 8 , wherein the chronic inflammation is associated with an age related macular degeneration, an allergy, an allergic rhinitis, an Alzheimer's disease, an amyotrophic lateral sclerosis, an anemia, an appendicitis, an arthritis, an asthma, an atherosclerosis, an autoimmune disorder, a balanitis, a blepharitis, a bronchiolitis, a bronchitis, a bullous pemphigoid, a burn, a bursitis, a cancer, a cardiovascular disease, a cellulitis, a cervicitis, a cholangitis, a cholecystitis, a chorioamnionitis, a chronic neurogenic inflammation, a chronic obstructive pulmonary disease, a cirrhosis, a conjunctivitis, a cyclophosphamide-induced cystitis, a cystic fibrosis, a cystitis, a common cold, a dacryoadenitis, a dementia, a diabetes, a diabetic neuropathy, a diabetic retinopathy, a diabetic nephropathy, a diabetic ulcer, a digestive system disease, an emphysema, an encephalitis, an endometritis, an epicondylitis, an epididymitis, a fasciitis, a fibromyalgia, a fibrosis, a fibrositis, a gastroenteritis, a gastrointestinal disorder, a gingivitis, a glomerulonephritis, a glossitis, a graft-versus-host disease, a hepatitis, a Huntington's disease, a hyperlipidemic pancreatitis, an infection, an inflammatory neuropathy, an insulin resistance, an interstitial cystitis, an interstitial nephritis, an iritis, an ischemia, a keratitis, a keratoconjunctivitis, a laryngitis, a lupus nephritis, a mastitis, a mastoiditis, a meningitis, a metabolic syndrome, a migraine, a multiple sclerosis, a myelitis, a myopathy, a nephritis, a non-alcoholic steatohepatitis, an obesity, an omphalitis, an oophoritis, an orchitis, an osteochondritis, an osteopenia, an osteomyelitis, an osteoporosis, an osteitis, an otitis, a pancreatitis, a Parkinson's disease, a parotitis, a pelvic inflammatory disease, a pemphigus vularis, a peritonitis, a pharmacologically-induced inflammation, a pharyngitis, a pleuritis, a pneumonitis, a polycystic nephritis, a proctitis, a prostatitis, a pulpitis, a pyelonephritis, a renal failure, a reperfusion injury, a retinitis, a rheumatic fever, a rhinitis, a salpingitis, a sarcoidosis, a sialadenitis, a sinusitis, a skin disorder, a stenosis, a stomatitis, a stroke, a surgical complication, a synovitis, a tendonitis, tendinosis, a tenosynovitis, a Th1-mediated inflammatory disease, a tissue or organ injury, tonsillitis, a transplant rejection, a trauma, a traumatic brain injury, a transplant rejection, a trigonitis, a tuberculosis, a tumor, an urethritis, an ursitis, an uveitis, a vaginitis, a vasculitis, or a vulvitis.

12. The method or use according to claim 11 , wherein the arthritis is an osteoarthritis, a rheumatoid arthritis, a juvenile idiopathic arthritis, a septic arthritis, a spondyloarthropathy, a gout, a pseudogout, or Still's disease.

13. The method or use according to claim 11 , wherein the spondyloarthropathy is an ankylosing spondylitis, a reactive arthritis (Reiter's syndrome), a psoriatic arthritis, an enteropathic arthritis associated with inflammatory bowel disease, a Whipple disease or a Behcet disease.

14. The method or use according to claim 11 , wherein the myopathy is a dermatomyositis, an inclusion body myositis, a myositis, or a polymyositis.

15. The method or use according to claim 11 , wherein the vasculitis is a Buerger's disease, an arteritis, a cerebral vasculitis, a Churg-Strauss arteritis, a cryoglobulinemia, an essential cryoglobulinemic vasculitis, a giant cell arteritis, a Golfer's vasculitis, a Henoch-Schonlein purpura, a hypersensitivity vasculitis, a Kawasaki disease, a phlebitis, a microscopic polyarteritis/polyangiitis, a polyarteritis nodosa, a polymyalgia rheumatica (PMR), a pylephlebitis, a rheumatoid vasculitis, a Takayasu arteritis, a thrombophlebitis, a Wegener's granulomatosis, or a vasculitis secondary to connective tissue disorder, or vasculitis secondary to viral infection.

16. The method or use according to claim 11 , wherein the skin disorder is an acne, a dermatitis, an eczema, a statis dermatitis, a hidradenitis suppurativa, a psoriasis, a rosacea or a scleroderma.

17. The method or use according to claim 16 , wherein the eczema is an atopic eczema, a contact eczema, a xerotic eczema, a seborrhoeic dermatitis, a dyshidrosis, a discoid eczema, a venous eczema, a dermatitis herpetiformis, a neurodermatitis, or an autoeczematization.

18. The method or use according to claim 16 , wherein the psoriasis is a plaqure psoriasis, a nail psoriasis, a guttate psoriasis, a scalp psoriasis, an inverse psoriasis, a pustular psoriasis, or an erythrodermis psoriasis.

19. The method or use according to claim 11 , wherein the gastrointestinal disorder is a celiac disease, a colitis, an enteritis, an enterocolitis, a gastritis, an ileitis, an irritable bowel disease, an inflammatory bowel disease, or a spastic colon.

20. The method or use according to claim 19 , wherein the inflammatory bowel disease is a Crohn's disease or an ulcerative colitis.

21. The method or use according to claim 11 , wherein the cardiovascular disease is an acid reflux/heartburn, an aneurysm, an arterial occlusive disease, a blood vessel inflammation, a cardiac arrest, a carditis, a congestive heart failure, a diabetic cardiac conditions, a dissection, an embolism, an endocarditis, a heart disease, a heart valve dysfunction, a hypertension, an inflammatory cardiomegaly, an ischemic heart disease, a myocardial infarction, a myocarditis, a pericarditis, a peripheral arterial disease, a pseudoaneurysm, a thrombosis, a thrombphlebitis, a vascular malformation, a vascular nevus, or a varicose veins.

22. The method or use according to claim 11 , wherein the infection is a bacterial cystitis, a bacterial encephalitis, a pandemic influenza, a viral encephalitis, a viral hepatitis A, a viral hepatitis B, or a viral hepatitis C.

23. The method according to claim 8 , wherein upon administration to the individual, the pharmaceutical composition results in a bio-distribution of the propionic acid derivative NSAID different than a bio-distribution of the propionic acid derivative NSAID included in the same pharmaceutical composition, except without the glycerolipid.

24. The method according to claim 23 , wherein the amount of the propionic acid derivative NSAID of the pharmaceutical composition is delivered to a macrophage is at least 5% of the total amount of the propionic acid derivative NSAID contained in the administered pharmaceutical composition.

25. The method according to claim 8 , wherein upon administration to the individual, the pharmaceutical composition reduces gastric or intestinal irritation by at least 5% when compared to the pharmaceutical composition, except without the glycerolipid.
