Abstract

The invention relates to nucleic acid complexes, methods of preparation thereof, and uses thereof for delivering a nucleic acid into a cell.

Claims (28)

1. A nucleic acid complex comprising a nucleic acid, a macrocyclic compound, and a pendant polymer, wherein the pendant polymer comprises a polymer backbone and is modified with a hydrophobic group, wherein the macrocyclic compound and the pendant polymer form a host:guest polymer complex, and wherein the nucleic acid is siRNA, mRNA, tRNA, rRNA, cDNA, miRNA, ribozymes, antisense oligonucleotides, decoy oligonucleotides, plasmid DNA (pDNA), peptide nucleic acids, triplex-forming oligonucleotides (TFOs), aptamers, or genes, or a derivative or analog thereof, or a combination thereof.

2. The nucleic acid complex of claim 1 , wherein said nucleic acid is siRNA, mRNA, tRNA, rRNA, or miRNA.

3. The nucleic acid complex of claim 1 , wherein said nucleic acid is siRNA or pDNA.

4. The nucleic acid complex of claim 1 , wherein said pendant polymer comprises a poly(vinyl alcohol), polysaccharide, polyester, polyamide, poly(ethylene glycol), or poly(propylene glycol) backbone.

5. The nucleic acid complex of claim 1 , wherein said pendant polymer comprises a poly(vinyl alcohol), poly(ethylene glycol), poly(propylene glycol), polydimethylsiloxane, polyethylene, polypropylene, hyaluronic acid, or pullulan backbone.

6. The nucleic acid complex of claim 1 , wherein said pendant polymer comprises a polysaccharide backbone.

7. The nucleic acid complex of claim 1 , wherein said pendant polymer comprises a hyaluronic acid backbone.

8. The nucleic acid complex of claim 1 , wherein said pendant polymer comprises a poly(ethylene glycol) pendant group.

9. The nucleic acid complex of claim 1 , wherein said macrocyclic compound is a modified β-cyclodextrin.

10. The nucleic acid complex of claim 1 , wherein said macrocyclic compound is a modified β-cyclodextrin comprising an amino moiety.

11. The nucleic acid complex of claim 1 , wherein said macrocyclic compound is

mono-6-(amino)-6-deoxy-β-cyclodextrin,

mono-6-(N,N′-dimethylethane-1,2-diamine)-6-deoxy-β-cyclodextrin,

mono-6-(N′ -(2-aminoethyl)ethane-1,2-diamine)-6-deoxy-β-cyclodextrin,

hepta-6-(2′-aminoethyl)amino-β-cyclodextrin,

hepta-6-(2′-hydroxyethylamino)-β-cyclodextrin, or

hepta-6-(hydrazino)-β-cyclodextrin.

12. The nucleic acid complex of claim 1 , wherein said hydrophobic group is a cholesterol group, an adamantane group, a diadamantane group, or a naphthalene group, or a derivative or analog thereof.

13. The nucleic acid complex of claim 1 , wherein said hydrophobic group is adamantane, or a derivative thereof.

14. The nucleic acid complex of claim 1 , wherein said hydrophobic group is cholesterol, or a derivative thereof.

15. The nucleic acid complex of claim 1 , wherein said hydrophobic group is linked through an acetal linkage to the polymer backbone of the pendant polymer.

16. A pharmaceutical composition comprising the nucleic complex of claim 1 to produce a pharmaceutical for delivering a nucleic acid into a cell.

17. The pharmaceutical composition of claim 16 , wherein said cell is a cancer cell.

18. A method for delivering a nucleic acid into a cell, the method comprising the step of bringing a nucleic acid complex of claim 1 comprising the nucleic acid into contact with the cell.

19. A nucleic complex comprising a nucleic acid, a macrocyclic compound, and a pendant polymer, wherein the pendant polymer comprises a polymer backbone and is modified with a hydrophobic group, wherein the macrocyclic compound and the pendant polymer form a host:guest polymer complex, and wherein the hydrophobic group is an adamantane group, or a derivative or analog thereof.

20. The nucleic acid complex of claim 19 , wherein said pendant polymer comprises a hyaluronic acid backbone.

21. A nucleic complex comprising a nucleic acid, a macrocyclic compound, and a pendant polymer, wherein the pendant polymer comprises a polymer backbone and is modified with a hydrophobic group, wherein the macrocyclic compound and the pendant polymer form a host:guest polymer complex, and wherein said pendant polymer comprises a hyaluronic acid backbone.

22. The nucleic acid complex of claim 21 , wherein the hydrophobic group is an adamantane group, or a derivative or analog thereof.
