Abstract

The present invention features methods for stimulating clearance of misfolded or aggregated proteins or peptides in microglia or neurons, and treating neurodegenerative diseases associated with such pathology in brain by selectively inhibiting the expression or activity of Acyl-CoA:Cholesterol Acyltransferase 1, but not Acyl-CoA:Cholesterol Acyltransferase 2.

Claims (16)

1. A method for stimulating clearance of misfolded or aggregated proteins and peptides in microglia or neurons comprising administering to a subject in need thereof an Acyl-CoA:Cholesterol Acyltransferase 1-selective inhibitor thereby stimulating clearance of the misfolded or aggregated proteins and peptides in microglia or neurons in the subject.

2. The method of claim 1 , wherein the inhibitor has an IC 50 value for Acyl-CoA:Cholesterol Acyltransferase 1 which is at least twice the corresponding IC 50 value for Acyl-CoA:Cholesterol Acyltransferase 2.

3. The method of claim 1 , wherein the inhibitor inhibits the expression or activity of Acyl-CoA:Cholesterol Acyltransferase 1.

4. The method of claim 1 , wherein the inhibitor has an IC 50 value in the range of 1 nM to 100 μM.

5. The method of claim 1 , wherein the inhibitor is administered intranasally, intrathecally, or directly to the brain of the subject by a shunt or a catheter.

6. The method of claim 1 , wherein the inhibitor is encapsulated in an exosome or a liposome.

7. The method of claim 6 , wherein the exosome or liposome is modified with a brain-targeting moiety.

8. The method of claim 1 , wherein the proteins are amyloid beta proteins.

9. A method for treating a neurodegenerative disease associated with accumulation of misfolded or aggregated proteins or peptides in microglia or neurons of brain tissue comprising administering to a subject in need thereof an Acyl-CoA:Cholesterol Acyltransferase 1-selective inhibitor thereby decreasing the accumulation of misfolded or aggregated proteins or peptides in microglia or neurons of brain tissue in the subject, thereby treating the neurodegenerative disease.

10. The method of claim 9 , wherein the neurodegenerative disease is Alzheimer's disease, tauopathy, frontotemporal dementia, Parkinson's disease or amylotrophic lateral sclerosis.

11. The method of claim 9 , wherein the inhibitor has an IC 50 value for Acyl-CoA:Cholesterol Acyltransferase 1 which is at least twice the corresponding IC 50 value for Acyl-CoA:Cholesterol Acyltransferase 2.

12. The method of claim 9 , wherein the inhibitor inhibits the expression or activity of Acyl-CoA:Cholesterol Acyltransferase 1.

13. The method of claim 9 , wherein the inhibitor has an IC 50 value in the range of 1 nM to 100 μM.

14. The method of claim 9 , wherein the inhibitor is administered intranasally, intrathecally, or directly to the brain of the subject by a shunt or a catheter.

15. The method of claim 9 , wherein the inhibitor is encapsulated in an exosome or a liposome.

16. The method of claim 15 , wherein the exosome or liposome is modified with a targeting moiety.
