Sense oligonucleotide capable of controlling the expression of INOS and composition comprising the same
The invention relates to sense oligonucleotide having a sequence complementary to a single-stranded RNA (antisense transcript) having a sequence complementary to mRNA of iNOS gene in order to control expression of iNOS (inducible nitric oxide synthase). The sense oligonucleotide of the present invention can control expression of iNOS and is useful for biological defense and treatment and prevention of diseases related to excessive production of NO, such as cancerogenesis, inflammatory disease, endotoxin shock by bacterial infection and the like.

A sense oligonucleotide, wherein said sense oligonucleotide is a single-stranded DNA oligonucleotide which comprises a sequence complementary to human iNOS (inducible nitric oxide synthase) mRNA antisense transcript, wherein said sequence complementary to human iNOS mRNA antisense transcript is complementary to at least 16 contiguous nucleotides of SEQ ID NO: 11,
and wherein said sense oligonucleotide contains one or more non-naturally occurring modifications which stabilize the oligonucleotide against decomposition by a nucleolytic enzyme.
2. The sense oligonucleotide according to claim 1 , wherein said sequence complementary to human iNOS mRNA antisense transcript is complementary to a thermodynamically unstable loop structure in said human iNOS mRNA antisense transcript.
3. The sense oligonucleotide according to claim 1 , wherein said oligonucleotide is 16-335 bases in length.
4. The sense oligonucleotide according to claim 1 , wherein the stabilization is performed by at least one modification selected from the group consisting of phosphorothioate, LNA, PNA, ENA, and morpholino.
5. A composition comprising the sense oligonucleotide according to claim 1 , wherein said composition has an activity of suppressing production of NO by promoting decomposition of iNOS mRNA.
6. A method of controlling production of the amount of iNOS, comprising introducing the sense oligonucleotide according to claim 1 into a cell.
7. The sense oligonucleotide according to claim 1 , wherein said sense oligonucleotide consists of a nucleotide sequence complementary to a non-stem region in a stem-loop structure of human iNOS (inducible nitric oxide synthase) mRNA antisense transcript.
