Abstract

Vaccine vectors and methods for enhancing resistance to Campylobacter infection or for enhancing the immune response to Campylobacter are provided herein. The vaccine vectors include a first polynucleotide which encodes an antigenic polypeptide selected from SEQ ID NO: 7 - 9 or a fragment thereof. The vector may also include an immunostimulatory polypeptide. The methods include administering the vaccine vectors to a subject.

Claims (14)

1. A vaccine vector comprising a first polynucleotide sequence encoding an antigenic polypeptide not natively associated with the vector and a second polynucleotide sequence encoding an HMGB1 polypeptide having at least 98% sequence identity to at least one of SEQ ID NOs: 20, 27 or 28, wherein the antigenic polypeptide consists of SEQ ID NO: 7, or a fragment of SEQ ID NO: 1 comprising SEQ ID NO: 7, wherein the vaccine vector elicits an immune response to Campylobacter jejuni.

2. The vaccine vector of claim 1 , wherein the vaccine vector is a bacterium.

3. The vaccine vector of claim 2 , wherein the bacterium is from a genus selected from Salmonella, Escherichia, Bacillus or Lactobacillus.

4. The vaccine vector of claim 2 , wherein the first polynucleotide sequence and the second polynucleotide sequence are integrated into the genome of the bacterium.

5. The vaccine vector of claim 1 , wherein the vaccine vector comprises more than one copy of the first polynucleotide sequence, more than one copy of the second polynucleotide sequence or more than one copy of the first and second polynucleotide sequence.

6. The vaccine vector of claim 1 , wherein the first polynucleotide sequence is linked in frame to the second polynucleotide sequence.

7. The vaccine vector of claim 1 , wherein the antigenic polypeptide and the HMGB1 polypeptide are expressed on the surface of the vaccine vector.

8. The vaccine vector of claim 7 , wherein the first polynucleotide sequence and the second polynucleotide sequence are inserted within a third polynucleotide sequence encoding an external portion of a transmembrane protein.

9. The vector of claim 1 , further comprising a second immunostimulatory polypeptide comprising a CD154 polypeptide.

10. The vaccine vector of claim 1 , wherein the antigenic polypeptide consists of SEQ ID NO: 7.

11. A method of inducing an immune response in a subject comprising administering the vector of claim 1 to the subject in an amount effective to induce the immune response of the subject to Campylobacter jejuni.

12. The method of claim 11 , wherein the immune response includes an antibody response to Campylobacter jejuni.

13. The method of claim 12 , wherein the soluble IgA antibody response to Campylobacter jejuni is induced.

14. The method of claim 11 , wherein the vector is attenuated or killed prior to administration.
