Abstract

Cellulose from lignocellulosic biomass which also contains protein, such as grasses and straws, can be enzymatically converted to valuable products such as glucose at lower enzyme consumption by: (a) extracting the biomass with water at 15-75° C. to produce an extracted biomass and a protein-containing aqueous extract; (b) mechanically, chemically and/or thermally pretreating cellulose-containing-biomass, in particular by organosolv treatment; and (c) contacting biomass pretreated in step (b) with: (i) an enzyme capable of hydrolysing cellulose; and (ii) the protein-containing aqueous extract obtained in step (a).

Claims (20)

1 . A process for enzymatic hydrolysis of cellulose, comprising:

(a) extracting biomass containing both protein and cellulose with an aqueous liquid to produce an extracted biomass and an aqueous extract;

(b) mechanically, chemically and/or thermally pretreating cellulose-containing biomass; and

(c) contacting biomass pretreated in step (b) with:

(i) an enzyme capable of hydrolysing cellulose; and

(ii) the aqueous extract obtained in step (a), optionally after concentration.

2 . The process according to claim 1 , wherein the extracted biomass obtained in step (a) is used as the cellulose-containing-biomass in step (b).

3 . The process according to claim 2 , wherein the extracted biomass obtained in step (a) is subjected prior to step (b) to extraction with an organic liquid comprising at least 20 wt % of an organic solvent.

4 . The process according to claim 1 , wherein in step (a) the biomass is extracted with water having a temperature of 10-75° C.

5 . The process according to claim 1 , wherein in step (a) the biomass is extracted with 2-12 l of water per kg of biomass.

6 . The process according to claim 1 , wherein the aqueous extract obtained in step (a) has a protein content of 0.01-50 g/l.

7 . The process according to claim 1 , wherein an amount of aqueous extract is used in step (c) which contains between 0.001 and 0.1 g of protein per g pretreated biomass on dry weight basis.

8 . The process according to claim 1 , wherein step (b) comprises heating at a temperature above 75° C.

9 . The process according to claim 1 , wherein step (b) comprises treating with a water-miscible organic solvent, optionally in the presence of an acid.

10 . The process according to claim 1 , wherein step (b) comprises a steam explosion step or an aquathermolysis step.

11 . The process according to claim 1 , wherein in step (c) the enzyme capable of hydrolysing cellulose comprises a mixture of cellulases.

12 . The process according to claim 4 , wherein in step (a) the biomass is extracted with water having a temperature of 15-60° C.

13 . The process according to claim 6 , wherein the aqueous extract obtained in step (a) has a protein content of 0.1-20 g/l.

14 . The process according to claim 8 , wherein step (b) comprises heating at a temperature between 120 and 240° C.

15 . The process according claim 9 , wherein the water-miscible organic solvent is selected from the group consisting of lower alcohols and lower ketones.
