Compositions and methods for modulating metabolic pathways
Compositions and methods useful for inducing an increase in fatty acid oxidation or mitochondrial biogenesis, reducing weight gain, inducing weight loss, or increasing Sirt1, Sirt3, or AMPK activity are provided herein. Such compositions can contain synergizing amounts of a sirtuin-pathway activators, including but not limited to resveratrol, in combination with beta-hydroxymethylbutyrate (HMB), keto isocaproic acid (KIC), leucine, or combinations of HMB, KIC and leucine.
1. A method of administering a composition to a subject comprising:
a. administering to the subject at least about 500 mg of leucine and/or at least about 200 mg of one or more metabolites thereof, wherein the one or more leucine metabolites are selected from the group consisting of keto-isocaproic acid (KIC), alpha-hydroxy-isocaproic acid, and HMB; and
b. administering to the subject an anti-diabetic agent comprising a biguanide and/or a thiazolidinedione, wherein the composition is substantially free of the individual amino acids alanine, glutamic acid, glycine, isoleucine, valine, and proline;
wherein when (a) is leucine, the molar ratio of (a) to (b) is greater than 2; and
wherein (a) and (b) are administered sequentially in any order or simultaneously.
2. The method of claim 1 , wherein the composition comprises a subtherapeutic amount of the anti-3 diabetic agent.
3. The method of claim 1 , wherein the anti-diabetic agent comprises at least about 25 mg to less than about 2000 mg of metformin.
4. The method of claim 1 , wherein the anti-diabetic agent comprises at least about 50 mg of resveratrol and at least about 500 mg of metformin.
5. The method of claim 1 , wherein the anti-diabetic agent comprises at least about 50 mg of metformin and less than about 500 mg of metformin.
6. The method of claim 1 , wherein the anti-diabetic agent comprises rosiglitazone in an amount effective to achieve a circulating level of at least about 1 nM rosiglitazone in a subject.
7. The method of claim 1 , wherein the composition further comprises resveratrol.
8. The method of claim 1 , wherein (a) and (b) are administered sequentially in either order.
9. The method of claim 1 , wherein each of (a) and (b) are formulated as a tablet, capsule, gel capsule, beverage, snack bar or a food composition.
10. The method of claim 1 , wherein the subject has one or more symptoms of metabolic disorders.
11. The method of claim 10 , wherein the metabolic disorder is selected from the group consisting of obesity, diabetes, hypertension, arteriosclerosis, high cholesterol, and hyperlipidemia.
12. The method of claim 1 , wherein the insulin sensitivity in the subject is increased after said administering.
13. The method of claim 1 , wherein the fat oxidation in the subject is increased or the inflammatory response in the subject is decreased.
14. The method of claim 1 , wherein the composition is substantially free of an individual non-branched amino acid.
15. The method of claim 1 , wherein the anti-diabetic agent comprises metformin and rosiglitazone.
