Abstract

The invention relates to novel substituted pyrimidine compounds of general formula (I) in which the chemical groupings, substituents, variables and indices are as defined in the description, and to their use as medicaments, in particular as medicaments for the treatment of conditions and diseases that can be treated by inhibition of the PDE4 enzyme.

Claims (165)

1. A compound of formula (I)

wherein

A, B and C independently represent CH or N;

R 1 and R 2 independently represent

hydrogen or (C 1 -C 6 )-alkyl, whereby said (C 1 -C 6 )-alkyl is unsubstituted or substituted with at least one substituent X 1 , or

a group U, which is a 3- to 12-membered mono- or bi-cycloaliphatic ring, which is unsubstituted or substituted with at least one substituent X 2 , whereby said group U may be connected to the nitrogen atom via a C 1-6 -alkylene group, which in turn is unsubstituted or substituted with at least one substituent independently selected from the group consisting of F, Cl, CF 3 , ═O, OCF 3 and OH, or

a group V, which is a 3- to 12-membered mono-or bi-cyclic heterocycloaliphatic ring comprising at least one heteroatom selected from the group consisting of O, S and N as a ring member, and which mono- or bicyclic heterocycloaliphatic ring is unsubstituted or substituted with at least one substituent X 3 , whereby said group V may be connected to the nitrogen atom via a C 1-6 -alkylene group, which in turn may be unsubstituted or substituted with at least one substituent independently selected from the group consisting of F, Cl, CF 3 , ═O, OCF 3 and OH, or

a group W which is phenyl or 5- or 6-membered heteroaryl, wherein said phenyl or said heteroaryl is unsubstituted or substituted with at least one substituent X 4 and may be condensed with a 4-, 5-, 6- or 7-membered ring, being carbocyclic or heterocyclic, wherein said condensed ring may be saturated, partially unsaturated or aromatic and may be substituted with at least on substituent X 5 , and whereby group W may be connected to the nitrogen atom via a C 1-6 -alkylene group, which in turn may be unsubstituted or substituted with at least one substituent independently selected from the group consisting of F, Cl, CF 3 , ═O, OCF 3 and OH, or

R 1 and R 2 together with the nitrogen atom to which they are attached form a 3- to 12-membered monocyclic or bicyclic non-aromatic or aromatic ring wherein said ring may contain at least one additional heteroatom selected from the group consisting of O,S and N and wherein said ring is unsubstituted or substituted with at least one substituent X 6 ;

R 3 and R 4 ,

independently represent hydrogen, (C 1 -C 6 )-alkyl or (C 3 -C 6 )-cycloalkyl, wherein said (C 1 -C 6 )-alkyl and (C 3 -C 6 )-cycloalkyl are each unsubstituted or substituted with at least one substituent Y 1 , or

together with the carbon atom to which they are attached form a 3- to 6-membered cycloalkyl ring, which is unsubstituted or substituted with at least one substituent Y 2 ;

L represents a bond, O, S, (C 1 -C 6 )-alkylene or (C 2 -C 6 )-alkenylene, whereby the aforementioned alkylenes or alkenylenes are in each case unsubstituted or substituted with at least one substituent selected from the group consisting of F, Cl, CF 3 , ═O,OCF 3 and OH;

G represents a phenyl or 5- or 6-membered heteroaryl, wherein said phenyl or said heteroaryl may be substituted with at least one substituent Z;

X 1 , X 2 , X 3 , X 5 and X 6 , at each occurrence are independently from one another selected from the group consisting of OH, ═O,CN, nitro, halogen, (C 1 -C 6 )-alkyl, (C 1 -C 6 )-hydroxyalkyl, (C 1 -C 6 )-alkoxy, S(C 1 -C 6 )-alkyl, S(O)—(C 1 -C 6 )-alkyl, S(O) 2 —(C 1 -C 6 )-alkyl, (C 1 -C 6 )-haloalkyl, S(C 1 -C 6 )-haloalkyl, (C 1 -C 6 )-haloalkoxy, (C 1 -C 6 )-cyanoalkyl, (C 3 -C 8 )-cycloalkyl, NH 2 , NH(C 1 -C 6 )-alkyl, N((C 1 -C 6 )-alkyl) 2 , NH—CO—(C 1 -C 6 )-alkyl, NH—SO—(C 1 -C 6 )-alkyl, NH—S(O) 2 —(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-SO—(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-SO—(C 1 -C 6 )-alkyl, NHCONH 2 , NH—CO—NH—(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-CO—N((C 1 -C 6 )-alkyl) 2 , CO—(C 1 -C 6 )-alkyl, CO 2 H, CO—O—(C 1 -C 6 )-alkyl, CONH 2 , CO—NH(C 1 -C 6 )alkyl and CO—N((C 1 -C 6 )-alkyl) 2 ;

X 4 at each occurrence are independently from one another selected from the group consisting of OH, CN, nitro, halogen, (C 1 -C 6 )-alkyl, (C 1 -C 6 )-hydroxyalkyl, (C 1 -C 6 )-alkoxy, S(C 1 -C 6 )-alkyl, S(O)—(C 1 -C 6 )-alkyl, S(O) 2 —(C 1 -C 6 )-alkyl, (C 1 -C 6 )-haloalkyl, S(C 1 -C 6 )-haloalkyl, (C 1 -C 6 )-haloalkoxy, (C 1 -C 6 )-cyanoalkyl, (C 3 -C 8 )-cycloalkyl, NH 2 , NH(C 1 -C 6 )-alkyl, N((C 1 -C 6 )-alkyl) 2 , NH—CO—(C 1 -C 6 )-alkyl, NH—SO—(C 1 -C 6 )-alkyl, NH—S(O) 2 —(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-CO—(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-SO—(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-SO 2 —(C 1 -C 6 )-alkyl, NHCONH 2 , NH—CO—NH—(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-CO—N((C 1 -C 6 )-alkyl) 2 , CO 2 H, CO—O—(C 1 -C 6 )-alkyl, CONH 2 , CO—NH(C 1 -C 6 )alkyl and CO—N((C 1 -C 6 )-alkyl) 2 ;

Y 1 and Y 2 , at each occurrence are independently from one another selected from the group consisting of OH, ═O CN, nitro, halogen, (C 1 -C 6 )-alkyl, (C 1 -C 6 )-hydroxyalkyl, (C 1 -C 6 )-alkoxy, S(C 1 -C 6 )-alkyl, S(O)—(C 1 -C 6 )-alkyl, S(O) 2 —(C 1 -C 6 )-alkyl, (C 1 -C 6 )-haloalkyl, S(C 1 -C 6 )-haloalkyl, (C 1 -C 6 )-haloalkoxy, (C 1 -C 6 )-cyanoalkyl, (C 3 -C 8 )-cycloalkyl, NH 2 , NH(C 1 -C 6 )-alkyl, N((C 1 -C 6 )-alkyl) 2 , NH—CO—(C 1 -C 6 )-alkyl, NH—SO—(C 1 -C 6 )-alkyl, NH—S(O) 2 —(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-CO—(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-SO—(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-SO 2 —(C 1 -C 6 )-alkyl, NHCONH 2 , NH—CO—NH—(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-CO—N((C 1 -C 6 )-alkyl) 2 , CO 2 H, CO—O—(C 1 -C 6 )-alkyl, CONH 2 , CO—NH(C 1 -C 6 )alkyl and CO—N((C 1 -C 6 )-alkyl) 2 ;

optionally in the form of a single stereoisomer or a mixture of stereoisomers, in the form of the free compound and/or a physiologically acceptable salt and/or a physiologically acceptable solvate thereof.

2. The compound of claim 1 , wherein

A, B and C independently represent CH or N;

R 1 and R 2 independently represent

hydrogen or (C 1 -C 6 )-alkyl, whereby said (C 1 -C 6 )-alkyl is unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents X 1 , or

a group U, which is a 3- to 12-membered mono- or bi-cycloaliphatic ring, which is unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents X 2 , whereby said group U may be connected to the nitrogen atom via a C 1-3 -alkylene group, which in turn is unsubstituted or substituted with 1, 2 or 3 substituents independently selected from the group consisting of F, Cl, CF 3 , ═O, OCF 3 and OH, or

a group V, which is a 3- to 12-membered mono-or bi-cyclic heterocycloaliphatic ring comprising at 1, 2 or 3 heteroatoms selected from the group consisting of O, S and N as ring members, and which mono- or bicyclic heterocycloaliphatic ring is unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents X 3 ; whereby said group V may be connected to the nitrogen atom via a C 1-3 -alkylene group, which in turn is unsubstituted or substituted with 1, 2 or 3 substituents independently selected from the group consisting of F, Cl, CF 3 , ═O, OCF 3 and OH, or

a group W which is phenyl or 5- or 6-membered heteroaryl, wherein said phenyl or said heteroaryl is unsubstituted or substituted with 1, 2, 3, 4, or 5 substituents X 4 , and wherein said phenyl or said heteroaryl may be condensed with a 4-, 5-, 6- or 7-membered ring, being carbocyclic or heterocyclic, wherein said condensed ring may be saturated, partially unsaturated or aromatic and is unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents X 5 ; and wherein said group W may be connected to the nitrogen atom via a C 1-3 -alkylene group, which in turn may be unsubstituted or substituted with at least one substituent independently selected from the group consisting of F, Cl, CF 3 , ═O, OCF 3 and OH, or

R 1 and R 2 together with the nitrogen atom to which they are attached form a 3- to 12-membered monocyclic or bicyclic non-aromatic or aromatic ring wherein said ring may contain 1, 2 or 3 additional heteroatoms selected from the group consisting of O, S and N and wherein said ring is unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents X 6 ;

R 3 and R 4 ,

independently of one another, each represent hydrogen, (C 1 -C 6 )-alkyl or (C 3 -C 6 )-cycloalkyl, wherein said (C 1 -C 6 )-alkyl and (C 3 -C 6 )-cycloalkyl are each unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents Y 1 , or

together with the carbon atom to which they are attached form a 3- to 6-membered cycloalkyl ring, which is unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents Y 2 ;

L represents a bond, O,S, (C 1 -C 6 )-alkylene or (C 2 -C 6 )-alkenylene, whereby the aforementioned alkylenes or alkenylenes are in each case unsubstituted or substituted with 1, 2 or 3 substituents independently selected from the group consisting of F, Cl, CF 3 , ═O, OCF 3 and OH;

G represents a phenyl or 5- or 6-membered heteroaryl, wherein said phenyl or said heteroaryl is unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents Z;

X 1 , X 2 , X 3 , X 5 and X 6 , at each occurrence are independently from one another selected from the group consisting of OH, ═O, CN, F, Cl, Br, CHF 2 , CH 2 F, CF 3 , OCF 3 , SCF 3 , (C 1 -C 6 )-alkyl, (C 1 -C 6 )-hydroxyalkyl, (C 1 -C 6 )-alkoxy, S(C 1 -C 6 )-alkyl, S(O)—(C 1 -C 6 )-alkyl, S(O) 2 —(C 1 -C 6 )-alkyl, (C 1 -C 6 )-cyanoalkyl, (C 3 -C 6 )-cycloalkyl, NH 2 , NH(C 1 -C 6 )-alkyl, N((C 1 -C 6 )-alkyl) 2 , NH—CO—(C 1 -C 6 )-alkyl, NH—SO—(C 1 -C 6 )-alkyl, NH—S(O) 2 —(C 1 -C 6 )-alkyl, CO 2 H, CO—(C 1 -C 6 )-alkyl, CO—O—(C 1 -C 6 )-alkyl, CONH 2 , CO—NH(C 1 -C 6 )alkyl and CO—N((C 1 -C 6 )-alkyl) 2 ;

X 4 at each occurrence is independently selected from the group consisting of OH, CN, F, Cl, Br, CHF 2 , CH 2 F, CF 3 , OCF 3 , SCF 3 , (C 1 -C 6 )-alkyl, (C 1 -C 6 )-hydroxyalkyl, (C 1 -C 6 )-alkoxy, S(C 1 -C 6 )-alkyl, S(O)—(C 1 -C 6 )-alkyl, S(O) 2 —(C 1 -C 6 )-alkyl, (C 1 -C 6 )-cyanoalkyl, (C 3 -C 6 )-cycloalkyl, NH 2 , NH(C 1 -C 6 )-alkyl, N((C 1 -C 6 )-alkyl) 2 , NH—CO—(C 1 -C 6 )-alkyl, NH—SO—(C 1 -C 6 )-alkyl, NH—S(O) 2 —(C 1 -C 6 )-alkyl, CO 2 H, CO—O—(C 1 -C 6 )-alkyl, NHCONH 2 , NH—CO—NH—(C 1 -C 6 )-alkyl, N((C 1 -C 6 )-alkyl)-CO—N((C 1 -C 6 )-alkyl) 2 , CO 2 H, CO—O—(C 1 -C 6 )-alkyl, CONH 2 , CONH 2 , CO—NH(C 1 -C 6 )alkyl and CO—N((C 1 -C 6 )-alkyl) 2 ;

Y 1 and Y 2 , at each occurrence, are independently from one another selected from the group consisting of OH, ═O, CN, F, Cl, Br, CHF 2 , CH 2 F, CF 3 , OCF 3 , SCF 3 , (C 1 -C 6 )-alkyl, (C 1 -C 6 )-hydroxyalkyl, (C 1 -C 6 )-alkoxy, S(C 1 -C 6 )-alkyl, S(O)—(C 1 -C 6 )-alkyl, S(O) 2 -(C 1 -C 6 )-alkyl, (C 1 -C 6 )-cyanoalkyl, (C 3 -C 6 )-cycloalkyl, NH 2 , NH(C 1 -C 6 )-alkyl, N((C 1 -C 6 )-alkyl) 2 , NH—CO—(C 1 -C 6 )-alkyl, NH—SO—(C 1 -C 6 )-alkyl, NH—S(O) 2 —(C 1 -C 6 )-alkyl, CO 2 H, CO—O—(C 1 -C 6 )-alkyl, CONH 2 , CO—NH(C 1 -C 6 )alkyl and CO—N((C 1 -C 6 )-alkyl) 2 ;

Z at each occurrence is independently selected from the group consisting F, Cl, Br, CHF 2 , CH 2 F, CF 3 , OCF 3 , SCF 3 , OH, CN, SH, nitro, (C 1 -C 6 )-alkyl, (C 2 -C 6 )-alkenyl, (C 2 -C 6 )-alkinyl, (C 1 -C 6 )-hydroxyalkyl, (C 1 -C 6 )-cyanoalkyl, (C 1 -C 6 )-alkoxy, (C 1 -C 6 )-thioalkyl, (C 1 -C 6 )-alkylen-S—(C 1 -C 6 )-alkyl, (C 3 -C 6 )-cycloalkyl, hydroxyl-(C 3 -C 6 )-cycloalkyl, (C 3 -C 8 )-heterocycloalkyl, NH 2 , NH(C 1 -C 6 )-alkyl, N((C 1 -C 6 )-alkyl) 2 , NH—CO—(C 1 -C 6 )-alkyl, NH—CO—O—(C 1 -C 6 )-alkyl, NH—S(O) 2 OH, NH—S(O) 2 (C 1 -C 6 )-alkyl, NH—S(O) 2 O(C 1 -C 6 )-alkyl, NH—S(O) 2 NH 2 , NH—S(O) 2 NH(C 1 -C 6 )-alkyl, NH—S(O) 2 N((C 1 -C 6 )-alkyl) 2 , NH((C 1 -C 6 )-alkylen)-S(O) 2 OH, NH((C 1 -C 6 )-alkylen)-S(O) 2 (C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-S(O) 2 O(C 1 -C 6 )-alkyl, NH((C 1 -C 6 )-alkylen)-S(O) 2 NH 2 , NH((C 1 -C 6 )-alkylen)-S(O) 2 NH(C 1 -C 6 )-alkyl, CO 2 H, CO(C 1 -C 6 )-alkyl, CO—O(C 1 -C 6 )-alkyl, O—CO(C 1 -C 6 )-alkyl, CONH 2 , CO—NH(C 1 -C 6 )-alkyl, CO—N((C 1 -C 6 )-alkyl) 2 , O—CO—NH(C 1 -C 6 )-alkyl, O—CO—N((C 1 -C 6 )-alkyl) 2 , O—S(O) 2 —(C 1 -C 6 )-alkyl, O—S(O) 2 OH, O—S(O) 2 —(C 1 -C 6 )-alkoxy, O—S(O) 2 NH 2 , O—S(O) 2 —NH(C 1 -C 6 )-alkyl, O—S(O) 2 —N((C 1 -C 6 )-alkyl) 2 , S(O)(C 1 -C 6 )-alkyl, S(O) 2 (C 1 -C 6 )-alkyl, CH 2 S(O)(C 1 -C 6 )-alkyl, CH 2 S(O) 2 (C 1 -C 6 )-alkyl, S(O) 2 OH, S(O) 2 O(C 1 -C 6 )-alkyl, S(O) 2 NH 2 , S(O) 2 NH(C 1 -C 6 )-alkyl, and S(O) 2 N((C 1 -C 6 )-alkyl) 2 ;

optionally in the form of a single stereoisomer or a mixture of stereoisomers, in the form of the free compound and/or a physiologically acceptable salt and/or a physiologically acceptable solvate thereof.

3. The compound of claim 1 , wherein

A, B and C independently represent CH or N;

R 1 and R 2 independently represent

hydrogen or (C 1 -C 6 )-alkyl, whereby said (C 1 -C 6 )-alkyl is unsubstituted or substituted with 1, 2, 3, 4 or 5 substituents X 1 ; whereby at each occurrence X 1 is independently selected from the group consisting of F, Cl, Br, CN, (C 1 -C 6 )-alkoxy, OH, CONH 2 , CONH(C 1 -C 6 )-alkyl, CON((C 1 -C 6 )-alkyl) 2 , NH 2 , NH(C 1 -C 6 )-alkyl and N((C 1 -C 6 )-alkyl) 2 , or
