Abstract

The invention relates to compounds of Formula (I) and their pharmaceutically acceptable salts, wherein the substituents are as described herein, and their use in medicine for the treatment of diseases, disorders associated with the modulation of calcium release-activated calcium (CRAC) channel. The invention also relates to pharmaceutical compositions containing such compounds in treating diseases disorders associated with calcium release-activated calcium (CRAC) channel modulators.

Claims (124)

1. A compound having the Formula (I):

wherein,

A and B are independently CR 3 or N;

ring R is Formula (i);

L is selected from —NR 8 C(O)—, —C(O)NR 8 —and —NR 8 CH 2 —;

X is CR or N where R is selected from hydrogen, halogen or substituted or unsubstituted alkyl;

R 1 , which may be same or different at each occurrence, is independently selected from halogen, cyano, hydroxyl, substituted or unsubstituted alkyl, substituted or unsubstituted cycloalkyl and haloalkyl;

R 2 , which may be same or different at each occurrence, is independently selected from halogen, cyano, nitro, hydroxyl, substituted or unsubstituted alkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted alkoxy, haloalkyl, —NR 9 R 10 , —COOR 8 and —CONR 9 R 10 ;

each of R 3 is independently selected from hydrogen, halogen and substituted or unsubstituted alkyl;

R 4 and R 5 , which may be same or different and are independently selected from hydrogen, hydroxyl, substituted or unsubstituted alkyl, haloalkyl, substituted or unsubstituted alkoxy, substituted or unsubstituted cycloalkyl and —NR 9 R 10 ; or

R 4 and R 5 together with the carbon atom to which they are attached form oxo (C═O);

one of R 6 and R 7 is alkyl, haloalkyl, hydroxyalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted alkoxy, substituted or unsubstituted alkoxyalkyl and the other is selected from hydrogen, hydroxyl, alkyl, haloalkyl, hydroxyalkyl, substituted or unsubstituted alkoxy, substituted or unsubstituted alkoxyalkyl, —NR 9 R 10 , substituted or unsubstituted cycloalkyl, —COOR 8 and —CONR 9 R 10 ; or R 6 and R 7 together with the carbon atom to which they are attached may form substituted or unsubstituted 3 to 7 membered carbocyclic ring or 4 to 7 membered heterocyclic ring;

R 8 is hydrogen or substituted or unsubstituted alkyl;

R 9 and R 10 , which may be same or different, are independently selected from hydrogen, substituted or unsubstituted alkyl, haloalkyl, and substituted or unsubstituted cycloalkyl; or R 9 and R 10 , together with the nitrogen atom to which they are attached, may form a substituted or unsubstituted, saturated or unsaturated 5 to 7 membered cyclic ring, wherein the unsaturated cyclic ring may have one or two double bonds;

R a and R b , which may be same or different and are independently selected from hydrogen, cyano, hydroxyalkyl, substituted or unsubstituted alkyl, haloalkyl, substituted or unsubstituted alkoxy, haloalkoxy, substituted or unsubstituted cycloalkyl, —NR 9 R 10 , —COOR S and —CONH 2 ;

‘n’ is an integer ranging from 1 to 3, both inclusive; and

‘p’ is an integer ranging from 0 to 2, both inclusive;

or a pharmaceutically acceptable salt thereof.

2. The compound of claim 1 having the structure of Formula (II):

wherein,

A and B are independently CR 3 or N;

L is selected from —NHC(O)—, —C(O)NH—and —NHCH 2 —;

X is CH or N;

R 1 , which may be same or different at each occurrence, is independently selected from halogen, substituted or unsubstituted alkyl, substituted or unsubstituted cycloalkyl and haloalkyl;

R 2 is selected from halogen, substituted or unsubstituted alkyl, substituted or unsubstituted cycloalkyl, haloalkyl and substituted or unsubstituted alkoxy;

R 3 is selected from hydrogen, halogen and substituted or unsubstituted alkyl;

R 4 and R 5 , which may be same or different and are independently selected from hydrogen, hydroxyl, substituted or unsubstituted alkyl, substituted or unsubstituted alkoxy and —NR 9 R 10 ; or R 4 and R 5 together with the carbon atom to which they are attached form oxo (C═O);

one of R 6 and R 7 is alkyl, haloalkyl, hydroxyalkyl and the other is selected from hydrogen, hydroxyl, alkyl, haloalkyl, hydroxyalkyl, substituted or unsubstituted alkoxy, substituted or unsubstituted alkoxyalkyl, —NR 9 R 10 , substituted or unsubstituted cycloalkyl, —COOR 8 and —CONH 2 ; or R 6 and R 7 together with the carbon atom to which they are attached may form substituted or unsubstituted 3 to 6 membered carbocyclic ring or 4 to 7 membered heterocyclic ring;

R 8 is hydrogen or substituted or unsubstituted alkyl;

R 9 and R 10 are independently hydrogen or substituted or unsubstituted alkyl;

R a and R b are independently hydrogen or substituted or unsubstituted alkyl;

‘n’ is an integer ranging from 1 to 2, both inclusive; and

‘p’ is an integer ranging from 0 to 1, both inclusive;

or a pharmaceutically acceptable salt thereof.

3. The compound of claim 1 , wherein L is selected from —NHC(O)—, —C(O)NH—and —NHCH 2 —.

4. The compound of claim 1 , wherein ring R is Formula (i)

wherein R 2 is halogen or substituted or unsubstituted alkyl; ‘p’ is 0 to 1; R 4 and R 5 may be same or different and are independently selected from hydrogen, hydroxyl, substituted or unsubstituted alkyl and —NR 9 R 10 or R 4 and R 5 together with the carbon atom to which they are attached form oxo (C═O); one of R 6 and R 7 is alkyl, hydroxyalkyl and the other is selected from hydrogen, hydroxyl, alkyl, hydroxyalkyl, substituted or unsubstituted alkoxyalkyl, substituted or unsubstituted cycloalkyl, —NR 9 R 10 , —COOH, —COO—alkyl, and —CONH 2 ; or R 6 and R 7 together form substituted or unsubstituted 3 to 6 membered carbocyclic ring or 4 to 7 membered heterocyclic ring; R 9 and R 10 are selected from hydrogen or substituted or unsubstituted alkyl; and R a and R b are hydrogen.

5. The compound of claim 1 , wherein R 6 and R 7 together with carbon atom to which they are attached to form a ring which is selected from:

wherein L is selected from —NHC(O)—, —C(O)NH— and —NHCH 2 —; X is CH or N; A and B are independently CR 3 or N; R 1 is halogen, substituted or unsubstituted alkyl or haloalkyl; ‘n’ is 1 or 2; R 3 is hydrogen or substituted or unsubstituted alkyl; and ring R is Formula (i)

wherein R 2 is halogen or substituted or unsubstituted alkyl; ‘p’ is 0 or 1; R 4 and R 5 , which may be same or different and are independently selected from hydrogen, hydroxyl, —NR 9 R 10 where R 9 and R 10 are hydrogen or substituted or unsubstituted alkyl, or R 4 and R 5 together form oxo (C═O); one of R 6 and R 7 is alkyl or hydroxyalkyl and the other is selected from hydrogen, hydroxyl, alkyl, hydroxyalkyl, substituted or unsubstituted alkoxyalkyl, —C(O)OH, —C(O)Oalkyl and —CONH 2 , or R 6 and R 7 together with the carbon atom to which they are attached may form substituted or unsubstituted 3 to 4 membered carbocyclic ring or 4 to 7 membered heterocyclic ring; and R a and R b are hydrogen.

8. A pharmaceutical composition comprising one or more compounds of Formula (I) according to claim 1 and one or more pharmaceutically acceptable excipients.
