Abstract

The present invention relates to compositions and methods comprising genes and peptides associated with cyclic peptides and cyclic peptide production in mushrooms. In particular, the present invention relates to using genes and proteins from Galerina species encoding peptides specifically relating to amatoxins in addition to proteins involved with processing cyclic peptide toxins. In a preferred embodiment, the present invention also relates to methods for making small peptides and small cyclic peptides including peptides similar to amanitin. Further, the present inventions relate to providing kits for making small peptides.

Claims (25)

1. A method of making a peptide from a recombinant prepropeptide sequence, comprising:

(a) providing a fungal prolyl oligopeptidase comprising an amino acid sequence selected from SEQ ID NO: 236, 237, 348, 716, and 722, and a proline-containing prepropeptide; and

(b) contacting the fungal prolyl oligopeptidase with the prepropeptide to make the peptide.

2. The method of claim 1 , wherein the peptide is at least six amino acids in length.

3. The method of claim 1 , wherein the peptide is up to fifteen amino acids in length.

4. The method of claim 1 , wherein the peptide is a cyclic peptide.

5. The method of claim 1 , wherein the peptide is a bicyclic peptide.

6. The method of claim 1 , wherein the peptide comprises sequence SEQ ID NO: 50.

7. The method of claim 1 , wherein the peptide is biologically active.

8. A method of making a peptide from a recombinant prepropeptide sequence, comprising:

(a) providing a fungal cell comprising a nucleic acid encoding a fungal prolyl oligopeptidase comprising an amino acid sequence selected from SEQ ID NO: 236, 237, 348, 716, and 722, and a proline-containing prepropeptide; and

(b) contacting the fungal cell with the prepropeptide to make the peptide.

9. The method of claim 8 , further comprising growing the fungal cell under conditions for expressing the fungal prolyl oligopeptidase and thereby making the peptide.

10. The method of claim 8 , wherein the peptide is at least six amino acids in length.

11. The method of claim 8 , wherein the peptide is up to fifteen amino acids in length.

12. The method of claim 8 , wherein the peptide is a cyclic peptide.

13. The method of claim 8 , wherein the peptide is a bicyclic peptide.

14. The method of claim 8 , wherein the peptide comprises sequence SEQ ID NO: 50.

15. The method of claim 8 , wherein the peptide is biologically active.

16. A cDNA encoding a fungal prolyl oligopeptidase comprising an amino acid sequence selected from SEQ ID NO:236, 237, 348, 716, and 722.

17. An expression cassette comprising a nucleic acid encoding a fungal prolyl oligopeptidase comprising an amino acid sequence selected from SEQ NO:236, 237, 348, 716, and 722.

18. An expression vector comprising a nucleic acid encoding a fungal prolyl oligopeptidase comprising an amino acid sequence selected from SEQ NO:236, 237, 348, 716, and 722.

19. A fungal prolyl oligopeptidase comprising an amino acid sequence with one amino acid change in a sequence selected from amino acid SEQ ID NO:236, 237, 348, 716, and 722.

20. The fungal prolyl oligopeptidase of claim 19 , where the amino acid change is a conservative amino acid substitution.

21. A composition comprising an a proline-containing prepropeptide and a fungal prolyl oligopeptidase comprising an amino acid sequence with one amino acid change in a sequence selected from amino acid SEQ ID NO: 236, 237, 348, 716, and 722.
