Abstract

The present invention relates to fibronectin based scaffold domain proteins that bind PCSK9. The invention also relates to the use of the innovative proteins in therapeutic applications to treat atherosclerosis, hypercholesterolemia and other cholesterol related diseases. The invention further relates to cells comprising such proteins, polynucleotides encoding such proteins or fragments thereof, and to vectors comprising the polynucleotides encoding the innovative protein.

Claims (16)

1. An isolated polypeptide comprising a fibronectin type III tenth ( 10 Fn3) domain wherein the 10 Fn3 has a BC, DE and FG loop, and wherein the BC loop comprises SEQ ID NO:112, the DE loop comprises SEQ ID NO:138, and the FG loop comprises SEQ ID NO:156, and wherein the polypeptide binds proprotein convertase subtilisin kexin type (PCSK9).

2. The isolated polypeptide of claim 1 , wherein the polypeptide binds PCSK9 with a K D of less than 10 nM.

3. The isolated polypeptide of claim 1 , wherein the polypeptide further comprises polyethylene glycol.

4. The isolated polypeptide of claim 1 , wherein the polypeptide further comprises sialic acid.

5. The isolated polypeptide of claim 1 , wherein the polypeptide further comprises Fc or a Fc fragment.

6. The isolated polypeptide of claim 1 , wherein the polypeptide further comprises serum albumin or a serum albumin binding protein.

7. The isolated polypeptide of claim 1 , wherein the polypeptide further comprises a serum immunoglobulin binding protein.

8. A pharmaceutical composition comprising the isolated polypeptide of claim 1 and a pharmaceutically acceptable carrier, excipient, or stabilizer.

9. An isolated polypeptide comprising a fibronectin type III tenth ( 10 Fn3) domain, wherein the polypeptide comprises SEQ ID NO:209, and wherein the polypeptide binds proprotein convertase subtilisin kexin type (PCSK9).

10. The isolated polypeptide of claim 9 , wherein the polypeptide binds PCSK9 with a K D of less than 10 nM.

11. The isolated polypeptide of claim 9 , wherein the polypeptide further comprises polyethylene glycol.

12. The isolated polypeptide of claim 9 , wherein the polypeptide further comprises sialic acid.

13. The isolated polypeptide of claim 9 , wherein the polypeptide further comprises Fc or a Fc fragment.

14. The isolated polypeptide of claim 9 , wherein the polypeptide further comprises serum albumin or a serum albumin binding protein.

15. The isolated polypeptide of claim 9 , wherein the polypeptide further comprises a serum immunoglobulin binding protein.

16. A pharmaceutical composition comprising the isolated polypeptide of claim 9 and a pharmaceutically acceptable carrier, excipient, or stabilizer.
