Abstract

The various technologies presented herein relate to identifying whether an individual has taken, and/or is under the influence of, a restricted drug. A density separation technique is utilized, wherein a sample (e.g., blood, saliva, urine, etc.,) which may include an analyte is exposed to a first plurality of beads having an analyte attached thereto, a second plurality of beads having a metabolite-specific antibody attached thereto, and a plurality of fluorophore-labelled analyte-specific antibodies. After incubation, any analyte in the sample (e.g., delta-9-THC) is bound to the fluorophore-labelled analyte-specific antibodies, any free fluorophore-labelled analyte-specific antibodies are attached to the analyte of the first beads, and any metabolite in the sample is bound to the second antibody. By applying centrifugal separation, the first beads move to a region which undergoes irradiation. If no fluorescence occurs, the sample includes the analyte; if fluorescence occurs, the sample does not include the analyte.

Claims (45)

1. A system comprising:

a chamber that includes a fluid, and is configured to accept a sample fluid, wherein the sample fluid comprises a delta-9-THC compound and metabolite, wherein the fluid further comprises:

a first bead having a first density and a second delta-9-THC compound attached thereto;

a second bead having a second density and a metabolite-specific antibody attached thereto, wherein the second density is less than the first density, and the metabolite binds with the metabolite-specific antibody; and

an analyte-specific antibody, wherein the analyte specific antibody is labelled with a fluorophore and the analyte specific antibody can bind with either of the delta-9-THC compound or the second delta-9-THC compound, wherein the analyte specific antibody binds with the delta-9-THC compound relative to binding with the second delta-9-THC compound attached to the first bead; and

a detector that, responsive to centrifugal force being applied to the fluid and the sample fluid, outputs an indication of a level of the delta-9-THC compound in the sample fluid.

2. The system of claim 1 , wherein the first bead further comprises a first protein attached thereto.

The system of claim 1 , wherein the chamber further comprises a second fluid, wherein the second fluid is located between the fluid and a closed end of the chamber, the second fluid having a higher density than the fluid, and upon application of the centrifugal force the first bead passes into the second fluid.

4. The system of claim 3 , wherein the second fluid comprises a colloidal suspension of silicon nanoparticles, dextran, poly(ethylene glycol), glycerol, sorbitol, iodixanol, cesium chloride, or perfluorodecalin.

The system of claim 3 , further comprising a rotatable body, wherein the first chamber is located on the rotatable body, the centrifugal force generated by rotation of the body causes the first bead to pass through to the second fluid while the second fluid has a density preventing the analyte specific antibody, when unattached to the first bead, from transferring into the second fluid.

6. The system of claim 5 , wherein the rotatable body is a disk.

7. The system of claim 5 , further comprising:

a light source located proximate to the second fluid, wherein light emitted from the light source has a wavelength to cause the fluorophore to fluoresce; and

a sensor located proximate to the second fluid, wherein the sensor is configured to:

detect fluorescence of the fluorophore when the analyte specific antibody is present in the second fluid; and

generate the indication of the presence of the analyte specific antibody.

8. The system of claim 7 , wherein the detector is configured with a threshold value, the threshold value being based upon a minimum amount of delta-9-THC compound to cause intoxication, and, in the event of:

a magnitude of the indication from the sensor being below the threshold value, the detector determines an amount of first delta-9-THC compound in the sample fluid exceeds the minimum amount of delta-9-THC compound to cause intoxication; or

a magnitude of the indication from the sensor being above the threshold value, the detector determines an amount of first delta-9-THC compound in the sample fluid is less than the minimum amount of delta-9-THC compound to cause intoxication.

9. The system of claim 1 , further comprising:

a second chamber that accepts the fluid, the system further comprises a sensor to detect presence of a third bead and a second antibody in the second chamber, wherein the second antibody is configured to bind with a second metabolite, and the centrifugal force applied to the second chamber causes the third bead to be located in a region of the second chamber separate from the second bead; and

a second sensor wherein the second sensor is configured to detect a presence of the second metabolite bound to the third antibody.

The system of claim 9 , wherein the second metabolite is a cocaine-based compound, a methamphetamine-based compound, a methamphetamine compound, an amphetamine compound, an opiate-based compound, an MDMA-based compound compound, a ketamine-based compound, a PCP-based compound, a lysergic acid diethylamide-based compound, or a psilocybin-based compound.

11. A system for detecting delta-9-THC in a fluid sample of a human, the system comprising:

a chamber for accepting the fluid sample, wherein the chamber further comprises:

a first bead having a first density and a second delta-9-THC compound attached thereto;

a second bead having a second density and a metabolite-specific antibody attached thereto, wherein the second density is less than the first density, and the metabolite binds with the metabolite-specific antibody; and

an analyte-specific antibody, wherein the analyte specific antibody is labelled with a fluorophore and the analyte specific antibody can bind with either of the delta-9-THC compound or the second delta-9-THC compound, wherein the analyte specific antibody binds with the delta-9-THC compound relative to binding with the second delta-9-THC compound attached to the first bead;

a motor for applying centrifugal force to the fluid sample in the chamber; and

a detection module for detecting, responsive to the centrifugal force being applied to the fluid sample in the chamber, whether the fluid sample comprises more than a threshold level of delta-9-THC.

12. The system of claim 11 , wherein the fluid sample is one of blood or saliva.

13. The system of claim 11 , wherein the chamber is located on a disk.

14. The system of claim 11 , wherein the first bead further comprises a first protein attached thereto.

The system of claim 11 , wherein the chamber further comprises a second fluid, wherein the second fluid is located between the fluid and a closed end of the chamber, the second fluid having a higher density than the fluid, and upon application of the centrifugal force the first bead passes into the second fluid.

16. The system of claim 15 , wherein the second fluid comprises a colloidal suspension of silicon nanoparticles, dextran, poly(ethylene glycol), glycerol, sorbitol, iodixanol, cesium chloride, or perfluorodecalin.

The system of claim 15 , further comprising a rotatable body, wherein the first chamber is located on the rotatable body, the centrifugal force generated by rotation of the body causes the first bead to pass through to the second fluid while the second fluid has a density preventing the analyte specific antibody, when unattached to the first bead, from transferring into the second fluid.

18. The system of claim 17 , wherein the rotatable body is a disk.

19. The system of claim 17 , further comprising:

a light source located proximate to the second fluid, wherein light emitted from the light source has a wavelength to cause the fluorophore to fluoresce; and

a sensor located proximate to the second fluid, wherein the sensor is configured to:

detect fluorescence of the fluorophore when the analyte specific antibody is present in the second fluid; and

generate the indication of the presence of the analyte specific antibody.

20. The system of claim 11 , further comprising:

a second chamber that accepts the fluid, the system further comprises a sensor to detect presence of a third bead and a second antibody in the second chamber, wherein the second antibody is configured to bind with a second metabolite, and the centrifugal force applied to the second chamber causes the third bead to be located in a region of the second chamber separate from the second bead; and

a second sensor wherein the second sensor is configured to detect a presence of the second metabolite bound to the third antibody.
