Abstract

The present disclosure provides substituted aliphanes, cyclophanes, heteraphanes, heterophanes, hetero-heteraphanes and metallocenes, of Formula I D-M-D  (Formula I) useful as antiviral agents. In certain embodiments disclosed herein M is a group —P-A-P— where A is Certain substituted aliphanes, cyclophanes, heteraphanes, heterophanes, hetero-heteraphanes and metallocenes disclosed herein are potent and/or selective inhibitors of viral replication, particularly Hepatitis C virus replication. Pharmaceutical compositions/and combinations containing one or more substituted aliphanes, cyclophanes, heteraphanes, heterophanes, hetero-heteraphanes and metallocenes and a pharmaceutically acceptable carrier are also provided by this disclosure. Methods for treating viral infections, including Hepatitis C viral infections are provided by the disclosure.

Claims (43)

1. A compound of the formula:

2. A pharmaceutical composition comprising an effective amount of a compound of the formula:

to treat HCV in a patient diagnosed with HCV or its pharmaceutically acceptable salt, further comprising an additional active agent, in a pharmaceutically acceptable carrier.

3. The pharmaceutical composition of claim 2 , wherein the additional active agent is a protease inhibitor.

4. The pharmaceutical composition of claim 2 , wherein the additional active agent is a nucleoside analog.

5. The pharmaceutical composition of claim 2 , wherein the additional active agent is a NS5a inhibitor.

6. The pharmaceutical composition of claim 2 , wherein the additional active agent is a NS5b inhibitor.

7. The pharmaceutical composition of claim 2 , wherein the additional active agent is a NS4a inhibitor.

8. The pharmaceutical composition of claim 2 , wherein the additional active agent is a NS4b inhibitor.

9. The pharmaceutical composition of claim 2 , wherein the additional active agent is a polymerase inhibitor.

10. The pharmaceutical composition of claim 2 , wherein the additional active agent is a NS3a inhibitor.

11. The pharmaceutical composition of claim 2 , wherein the additional active agent is an immunomodulatory compound.

12. The pharmaceutical composition of claim 2 , in an oral dosage form.

13. The pharmaceutical composition of claim 3 , in an oral dosage form.

14. The pharmaceutical composition of claim 4 , in an oral dosage form.

15. The pharmaceutical composition of claim 5 , in an oral dosage form.

16. The pharmaceutical composition of claim 6 , in an oral dosage form.

32. A method to treat HCV in a patient diagnosed with HCV with a pharmaceutical composition comprising a compound of the formula:

or its pharmaceutically acceptable salt, and further comprising an additional active agent optionally in a pharmaceutically acceptable carrier.

33. The method of claim 32 , wherein the additional active agent is a protease inhibitor.

34. The method of claim 32 , wherein the additional active agent is a nucleoside analog.

35. The method of claim 32 , wherein the additional active agent is a NS5a inhibitor.

36. The method of claim 32 , wherein the additional active agent is a NS5b inhibitor.

37. The method of claim 32 , wherein the additional active agent is a NS4a inhibitor.

38. The method of claim 32 , wherein the additional active agent is a NS4b inhibitor.

39. The method of claim 32 , wherein the additional active agent is a polymerase inhibitor.

40. The method of claim 32 , wherein the additional active agent is a NS3a inhibitor.

41. The method of claim 32 , wherein the additional active agent is an immunomodulatory compound.
