Abstract

The present invention comprises a method, materials and apparatus for investigating asthma in humans using dust mite allergen. The dust mite allergen is prepared to be of a controlled size such that particles are respirable, effectively aerosolized, and deliver a significant amount of allergen. The dust mite allergen is applied in a controlled manner within an environmental exposure chamber to elucidate the etiological links between dust mite allergen concentration and asthma response in humans. The environmental exposure chamber is specially-designed to promote homogeneity of allergen concentration. Preferably, the dust mite allergen preparation comprises particles having an average diameter of less than 25 microns, and more preferably 5-10 microns. Correlation between aerosolized particle count and allergen concentration enables “real-time” allergen concentration estimates without the use of expensive and time-consuming assay techniques.

21 . A method of exposing an asthma patient to allergens for investigating asthma in the asthma patient, the method comprising:

preparing a dust mite allergen preparation, the dust mite allergen preparation consisting of dust mite particles having an average diameter of 5 microns to 10 microns;

introducing an amount of the dust mite allergen preparation into an environmentally controlled chamber by aerosolizing a dry powder comprising the dust mite allergen preparation inside the environmentally controlled chamber; and

placing the asthma patient into the environmentally controlled chamber, thereby exposing the asthma patient to a controlled level of the dust mite allergen preparation.

22 . The method of claim 21 , wherein the step of introducing comprises dispersing the dust mite allergen preparation substantially uniformly throughout the environmentally controlled chamber.

23 . The method of claim 22 , comprising dispersing the dust mite allergen preparation so as to achieve a concentration in the environmentally controlled chamber in the range of 50 ng/m 3 to 200 ng/m 3 .

24 . The method of claim 22 , comprising dispersing the dust mite allergen preparation so as to achieve a concentration in the environmentally controlled chamber in the range of 80 ng/m 3 to 130 ng/m 3 .

25 . The method of claim 22 , comprising operating at least one fan within the environmentally controlled chamber to provide generally turbulent flow.

26 . The method of claim 21 , comprising:

determining an airborne allergen concentration within the environmentally controlled chamber; and

correlating the airborne allergen concentration with an allergenic response in the asthma patient to the dust mite allergen preparation.

27 . The method of claim 21 , comprising:

collecting an air sample in the environmentally controlled chamber, the air sample comprising the dust mite particles of the dust mite allergen preparation;

using an enzyme-linked immuno-sorbent assay method to measure an allergen concentration for the air sample;

quantifying particle size and particle number using a particle counter in the environmentally controlled chamber; and

correlating the allergen concentration and at least one of the particle size and particle number to yield an estimate of the allergen concentration for the environmentally controlled chamber.

28 . The method of claim 27 , comprising yielding the estimate on a substantially real-time basis.

29 . The method of claim 27 , comprising quantifying particle size and particle number in a plurality of locations within the environmentally controlled chamber.

30 . The method of claim 21 , wherein, prior to the step of placing, the step of introducing comprises aerosolizing the dry powder during an equilibration period so that an allergen concentration in the environmentally controlled chamber reaches a substantially steady state.

31 . The method of claim 21 , comprising:

prior to the step of placing, aerosolizing the dry powder during an equilibration period so that an allergen concentration in the environmentally controlled chamber reaches a generally steady state; and

once the steady state is achieved,

collecting an air sample in the environmentally controlled chamber,

measuring an allergen concentration for the air sample,

quantifying particle size and particle number in the environmentally controlled chamber, and

correlating the allergen concentration and at least one of the particle size and particle number to yield an estimate of the allergen concentration for the environmentally controlled chamber.

32 . The method of claim 21 , wherein the step of introducing comprises using a fluid bed aerosol generator to aerosolize the dry powder inside the environmentally controlled chamber.

33 . The method of claim 32 , comprising placing the fluid bed aerosol generator behind an internal wall in the environmentally controlled chamber to promote laminar airflow about the fluid bed aerosol generator.

34 . The method of claim 21 , wherein the step of preparing comprises deriving the dust mite particles from at least one of Dermatophagoides pteronyssinus and Dermatophagoides farinae.

35 . The method of claim 34 , wherein the step of preparing comprises mechanically milling the dust mite particles.

36 . A method of investigating asthma in an asthma patient, the method comprising:

preparing an allergen preparation consisting of dust mite particles having an average diameter of 5 microns to 10 microns;

aerosolizing a dry powder comprising the allergen preparation inside a chamber to introduce a controlled level of the allergen preparation into the chamber;

placing the asthma patient into the chamber to expose the asthma patient to the controlled level of the allergen preparation; and

monitoring the asthma patient for an allergenic response to the controlled level of the allergen preparation.

37 . The method of claim 36 , comprising, prior to the step of placing, aerosolizing the dry powder during an equilibration period so that an allergen concentration in the environmentally controlled chamber reaches a substantially steady state.

38 . The method of claim 37 , comprising dispersing the dust mite allergen preparation substantially uniformly throughout the environmentally controlled chamber.

39 . The method of claim 37 , comprising using a fluid bed aerosol generator to aerosolize the dry powder inside the environmentally controlled chamber.

40 . A method of investigating asthma in an asthma patient, the method comprising:

preparing an allergen preparation consisting of dust mite particles selected from at least one of Dermatophagoides pteronyssinus and Dermatophagoides farinae and having an average diameter of 5 microns to 10 microns;

during an equilibration period, using an aerosol generator to aerosolize a dry powder comprising the allergen preparation inside an environmentally controlled chamber so that an allergen concentration in the environmentally controlled chamber reaches a substantially steady state;

dispersing the dust mite allergen preparation substantially uniformly throughout the environmentally controlled chamber so as to achieve a concentration in the environmentally controlled chamber in the range of 50 ng/m 3 to 200 ng/m 3 ;

after the equilibration period, placing the asthma patient into the chamber to expose the asthma patient to the allergen concentration during an exposure period; and

monitoring the asthma patient for an allergenic response to the allergen concentration during the exposure period.
