COLLAGEN COMPOSITIONS AND USES FOR BIOMATERIAL IMPLANTS
Compositions containing purified collagen biomaterial derived from tissues, for example, insoluble amnion, soluble amnion, soluble chorion of the human placenta, are provided. The collagen compositions can be used to promote wound healing, promote tissue regeneration, prevent or reduce scarring, reduce local inflammation, minimize tissue rejection, promote graft integration. Methods for using the collagen composition as a biomaterial implant for dermal filling, skin grafting, and hair transplantation are also provided.
1 . A composition for tissue repair, or tissue augmentation comprising de-cellularized Type I collagen and Type III collagen in an amount effective to promote wound healing at site of surgery, injury, or wound relative to an untreated control, between about 30% to 100% by weight, of the total collagen present in the composition, preferably about 50% to 100%.
2 . The composition of claim 1 , wherein a part or all of the collagen is extracted from a human placenta.
3 . The composition of claim 2 , wherein the human placenta used for collagen extraction is any part of insoluble amnion, soluble amnion, soluble chorion.
4 . The composition of claim 1 , wherein the collagen is collagen-containing, de-cellularized human placenta extracellular matrix.
5 . The composition of claim 1 , wherein the collagen is sterile.
6 . The composition of claim 1 , wherein the collagen is cross-linked.
7 . The composition of claim 1 , wherein the Type III collagen is further treated with at least one alkaline solution to remove one or more intermolecular disulfide bonds.
8 . The composition of claim 7 , wherein the at least one alkaline solution is 1M NaOH.
9 . The composition of claim 1 further comprising one or more compounds selected from the group consist of antimicrobial agents, analgesic agents, local anesthetic agents, anti-inflammatory agents, immunosuppressant agents, anti-allergenic agents, enzyme cofactors, essential nutrients, growth factors, and combinations thereof.

The composition of claim 1 further comprising one or more cells selected from the group consisting of stem cells, mesenchymal stem cells, keratinocytes, fibroblasts, melanocytes, adipocytes, T lymphocytes, B lymphocytes, natural killer cells, dendritic cells, hair follicle cells, and combinations thereof.
11 . The composition of claim 1 formulated for injection into epidermis, dermis, or hypodermis.
12 . The composition of claim 1 diluted with sterile water, saline or buffer.
13 . The composition of claim 1 formulated into a gel, cream or ointment.
14 . The composition of claim 1 lyophilized into a bottle for resuspension with sterile water, saline or buffer suitable for injection.
15 . A skin graft or hair graft comprising the composition of claim 1 .
16 . The skin graft or hair graft of claim 15 wherein the composition is coated onto at least one portion of the skin graft or hair graft prior to, in conjunction with, or subsequent to implantation with the skin graft or hair graft.
17 . The skin graft or hair graft of claim 15 wherein the composition is coated onto is the at least one portion that directly contacts the site of surgery, injury, or wound.
18 . The skin graft of claim 17 wherein the skin graft is a harvested skin section from a donor site to a recipient site.
19 . The skin graft of claim 18 wherein the skin graft is autologous.
20 . The skin graft of claim 18 wherein the skin graft is a full-thickness graft or a split thickness graft.
21 . The hair graft of claim 15 wherein the hair graft is a harvested hair graft from a donor site to a recipient site.
22 . The hair graft of claim 21 wherein the hair graft comprises at least one hair follicle.
23 . The hair graft of claim 21 wherein the hair graft is autologous.
24 . The hair graft of claim 21 wherein the donor site comprises the temporal or the occipital region of scalp.
25 . The hair graft of claim 21 wherein the recipient site for hair grafts comprises scalp, the facial region, armpit or chest region or the pubic region.
26 . A method for promoting wound healing, promoting tissue regeneration, reducing scarring, reducing local inflammation, minimizing tissue rejection, promoting graft integration, comprising administering to a subject in need thereof an effective amount of the composition of claim 1 .
27 . The method of claim 26 wherein the composition is injected at a site of surgery, injury, or wound.
28 . The method of claim 26 wherein the composition is topically applied at a site of surgery, injury, or wound.
29 . The method of claim 26 wherein the subject has undergone, is undergoing, or will undergo one or more cosmetic applications to fill wrinkles, lines, folds, scars, and to enhance dermal tissue.
30 . The method of claim 26 wherein the subject is need of correcting baldness to restore head hair, body hair, eyelashes, eyebrows, and beard hair, and to fill in scars caused by accidents and surgery.
31 . The method of claim 26 wherein the subject has a site of injury due to any physical, chemical, mechanical, electrical, heat, sunlight, or radiation damages.
32 . The method of claim 26 wherein comprising repeating the administration of the composition of claim 1 until the subject achieves desired results.
