Abstract

The present invention provides, inter alia, compounds having the structure: wherein R 7 , R 8 and n are as disclosed herein, that selectively bind a RAS protein at two or more sites and methods for their synthesis. Compositions and kits containing the compounds, as well as methods of using the compounds and compositions for ameliorating or treating the effects of a disease associated with altered RAS signaling, such as a cancer, in a subject and methods for effecting cancer cell death are also provided herein. Methods of identifying a multivalent compound which binds selectively to a target protein also are provided herein.

Claims (22)

1. A compound having the structure:

wherein:

R 7 is aryl, wherein the aryl is optionally substituted with one or more groups consisting of halide, C 1-4 alkyl, —O—C 1-4 alkyl and a combination thereof, wherein the alkyl is optionally substituted with one or more halide;

R 8 is selected from the group consisting of H, alkyl, aryl, and C 1-4 alkyl-O-aryl wherein the alkyl is optionally substituted with halide, and the aryl is optionally substituted with one or more groups consisting of halide, C 1-4 alkyl, and a combination thereof; and

n is selected from the group consisting of an integer between 0-5,

or a hydrate or pharmaceutically acceptable salt thereof.

2. The compound of claim 1 , wherein R 7 is aryl substituted with one —O—C 1-4 alkyl, and wherein the alkyl is optionally substituted with one or more halide.

3. The compound of claim 1 , wherein R 8 is aryl substituted with one or more groups consisting of halide, C 1-4 alkyl, and a combination thereof.

4. The compound of claim 1 , wherein R 8 is H.

5. The compound of claim 1 , wherein the compound is:

or a hydrate or pharmaceutically acceptable salt thereof.

6. A pharmaceutical composition comprising a pharmaceutically acceptable carrier and a compound having the structure:

wherein:

R 7 is aryl, wherein the aryl is optionally substituted with one or more groups consisting of halide, C 1-4 alkyl, —O—C 1-4 alkyl and a combination thereof, wherein the alkyl is optionally substituted with one or more halide;

R 8 is selected from the group consisting of H, alkyl, aryl, and C 1-4 alkyl-O-aryl wherein the alkyl is optionally substituted with halide, and the aryl is optionally substituted with one or more groups consisting of halide, C 1-4 alkyl, and a combination thereof; and

n is selected from the group consisting of an integer between 0-5,

or a hydrate or pharmaceutically acceptable salt thereof.

7. The pharmaceutical composition of claim 6 , wherein R 7 is aryl substituted with one —O—C 1-4 alkyl, and wherein the alkyl is optionally substituted with one or more halide.

8. The pharmaceutical composition of claim 6 , wherein R 8 is aryl substituted with one or more groups consisting of halide, C 1-4 alkyl, and a combination thereof.

9. The pharmaceutical composition of claim 6 , wherein R 8 is H.

10. The pharmaceutical composition of claim 6 , wherein the compound is:

or a hydrate or pharmaceutically acceptable salt thereof.
