Abstract

The invention discloses diagnostic techniques based on single cell genomics, consisting of obtaining a blood sample, enriching a sub-population of cells present in the blood sample, sequestering individual cells or group of cells from the blood sample, obtaining sequencing data from the sequestered cells or group of cells, using genetic variant information to determine the provenance of the cells, and genetically analyzing the cells of the correct provenance to provide a diagnostic readout. Using the cell-based testing techniques of the invention, the number of false positives is greatly reduced when compared to cell-free DNA (cfDNA) based traditional testing techniques. The invention may be effectively employed for non-invasive prenatal (NIPT) diagnostics, oncological testing and other diagnostic procedures.

Claims (12)

1. A cell-based 1 diagnostic method, comprising the steps of:

(a) enriching a sub-population of cells of a blood sample;

(b) sequestering one or more cells of said blood sample by trapping said one or more cells under negative pressure at a junction between an inside of a well and a microfluidic channel, and thereafter lysinq said one or more cells by introducing a lysing agent in said well;

(c) sequencing said one or more cells;

(d) determining a provenance of said one or more cells based on said sequencing;

(e) determining a genetic characteristic of said one or more cells based on said sequencing and said provenance; and

(f) providing a diagnosis based on said genetic characteristic.

2. The cell-based diagnostic method of claim 1 , further utilizing at least one element selected from the group consisting of an immunomagnetic separation, a microfluidic manipulation, a cell morphology based separation and an antibody binding based separation, in said step (a) of enriching.

3. The cell-based diagnostic method of claim 2 , further utilizing at least one cell attribute selected from the group consisting of a size, a shape, a structure, a form and an elasticity, when utilizing said cell morphology based separation.

4. A method of sequestering of cells comprising the steps of:

(a) enriching a sub-population of said cells from a blood sample and feeding said sub population via a microfluidic channel into a reservoir; and

(b) sequestering one or more cells of said sub-population by trapping said one or more cells under negative pressure at a junction between an inside of a well caused by a punch-hole in said reservoir and another microfluidic channel and thereafter lysing said one or more cells by introducing a lysing agent in said well.
