Abstract

The invention provides methods of inhibiting the growth or metastasis of a cancer in a mammal by inhibiting a Ral GTPase in the mammal. The invention also provides small molecule inhibitors of Ral GTPases useful in the methods of the invention and pharmaceutical compositions containing the therapeutically effective compounds of the invention, and methods of using the same.

1. A compound selected from the group consisting of:

3-(3,4-dimethoxyphenyl)-1-methyl-1H-pyrazol-5(4H)-one

3-(3,4-dimethoxyphenyl)-1-phenyl-1H-pyrazol-5(4H)-one

3-(4-methoxyphenyl)-1-methyl-1H-pyrazol-5(4H)-one

6-amino-3-methyl-1,4-diphenyl-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile

6-amino-4-(4-fluorophenyl)-3-methyl-1-phenyl-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile

6-amino-1,3-dimethyl-4-phenyl-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile

6-amino-1-methyl-3,4-diphenyl-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile

2. A pharmaceutical composition comprising at least one compound selected from the group consisting of:

3-(3,4-dimethoxyphenyl)-1-methyl-1H-pyrazol-5(4H)-one

3-(3,4-dimethoxyphenyl)-1-phenyl-1H-pyrazol-5(4H)-one

3-(4-methoxyphenyl)-1-methyl-1H-pyrazol-5(4H)-one

6-amino-3-methyl-1,4-diphenyl-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile

6-amino-4-(4-fluorophenyl)-3-methyl-1-phenyl-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile

6-amino-1,3-dimethyl-4-phenyl-1,4-dihydropyrano[2,3-c]pyrazole-5-carbonitrile

and at least one pharmaceutical excipient,

wherein the pharmaceutical composition is an oral dosage form, a parenteral dosage form, or an inhalation dosage form.

4. The pharmaceutical composition of claim 3 , wherein the oral dosage form is at least one dosage form selected from the group consisting of: capsules, tablets, pills, dragees, powders, and granules.

5. The pharmaceutical composition of claim 3 , wherein the parenteral dosage form is at least one dosage form selected from the group consisting of: liquid solutions, liquid suspensions, and solutions or suspensions of powders, granules, or tablets.

6. The pharmaceutical composition of claim 3 , wherein the inhalation dosage form is at least one dosage form selected from the group consisting of: aerosol sprays, liquid sprays, drops, and powders.

7. The pharmaceutical composition of claim 3 , wherein the pharmaceutical excipient is at least one selected from the group consisting of: sodium citrate, dicalcium phosphate, fillers, extenders, binders, humectants, disintegrating agents, solution retarding agents, absorption accelerators, wetting agents, absorbents, lubricants, coloring agents, propellants, volatile unsubstituted hydrocarbons, and combinations thereof.

8. The pharmaceutical composition of claim 7 , wherein the pharmaceutical excipient is at least one selected from the group consisting of: starches, lactose, sucrose, glucose, mannitol, silicic acid, carboxymethylcellulose, alginates, gelatin, polyvinyl pyrrolidone, sucrose, acacia, glycerol, agar-agar, calcium carbonate, potato or tapioca starch, alginic acid, silicates, silicic acid, aluminum hydroxide, sodium carbonate, paraffin, quaternary ammonium compounds, cetyl alcohol, glycerol monosterate, kaolin, bentonite clay, talc, polyamide powder, calcium stearate, magnesium stearate, solid polyethylene glycols, sodium lauryl sulfate, chlorofluorohydrocarbons, butane, propane, and mixtures thereof.

9. The pharmaceutical composition of claim 3 , further comprising at least one additive selected from the group consisting of: solubilizing agents, emulsifiers, wetting agents, dispersing agents, suspending agents, sweetening agents, flavoring agents, coloring agents, preservative agents, and mixtures thereof.
