Abstract

Binding agents able to disrupt bacterial biofilms of diverse origin are described, including monoclonal antibodies suitable for administration to a selected species, and antibody mimics including aptamer nucleic acids. Methods to prevent formation of or to dissolve biofilms with these binding agents are also described. Immunogens for eliciting antibodies to disrupt biofilms are also described.

Claims (12)

1. A monoclonal antibody (mAb), which mAb is an Fv antibody, a bispecific antibody, a chimeric antibody, species-ized antibody or a complete antibody, wherein said complete antibody comprises generic constant regions heterologous to the variable regions thereof,

wherein said mAb

a) has affinity for at least one DNA binding protein II (DNABII) protein that exceeds the affinity of said DNABII protein for components of a biofilm that includes said DNABII protein and binds DNABII protein from at least three bacterial species; and

b) binds to a conformational epitope comprised in SEQ ID NO:80; and

c) wherein the variable region comprises the heavy and light chain variable region of TRL1068 (SEQ ID NO:1) and (SEQ ID NO: 2) or of TRL1330 (SEQ ID NO:21) and (SEQ ID NO: 22) or of TRL1337 (SEQ ID NO:25) and (SEQ ID NO:26).

2. A pharmaceutical or veterinary composition which comprises as an active ingredient the mAb of claim 1 along with a suitable pharmaceutical excipient.

3. The pharmaceutical or veterinary composition of claim 2 which further includes at least one antibiotic, and/or

which further includes at least one additional active ingredient which is an immunostimulant and/or an antipyrogenic and/or an analgesic.

4. A monoclonal antibody (mAb) of claim 1 , which comprises the heavy and light chain variable region of TRL1068 (SEQ ID NO:1) and (SEQ ID NO: 2).

5. A pharmaceutical or veterinary composition which comprises as an active ingredient the mAb of claim 4 along with a suitable pharmaceutical excipient.

6. The pharmaceutical or veterinary composition of claim 5 which further includes at least one antibiotic, and/or

which further includes at least one additional active ingredient which is an immunostimulant and/or an antipyrogenic and/ or an analgesic.
