Abstract

A method for producing a purified beta-1,3-glucan includes growing a biomass from genus Euglena organism and dewatering the grown biomass. The biomass is lysed to form a lysed biomass, which is washed and dewatered to produce a beta-1,3-glucan. The beta-1,3-glucan is dried to produce the purified beta-1,3-glucan.

Claims (45)

1 . A method for producing a purified beta-1,3-glucan, comprising:

growing a biomass from genus Euglena organisms;

dewatering the grown biomass;

lysing the biomass to form a lysed biomass;

washing and dewatering the lysed biomass to produce a beta-1,3-glucan; and

drying the beta-1,3-glucan to produce the purified beta-1,3-glucan.

2 . The method according to claim 1 comprising washing and dewatering the lysed biomass a plurality of times.

3 . The method according to claim 1 wherein the dewatering comprises centrifuging or gravity decanting the biomass.

4 . The method according to claim 3 wherein the centrifuging is selected from the group consisting of decanter, stacked-disk, conical plate, pusher, and peeler centrifuging.

5 . The method according to claim 1 wherein the lysing is selected from the group consisting of mechanical, pH and temperature driven, surfactant, and enzymatic lysing.

6 . The method according to claim 5 wherein the mechanical lysing comprises homogenizing or bead milling the biomass.

7 . The method according to claim 6 comprising homogenizing the biomass at a pressure greater than 500 barg.

8 . The method according to claim 6 comprising homogenizing the biomass at a pressure range of 500 to 1,900 barg.

9 . The method according to claim 6 comprising homogenizing the biomass at a pressure range of 750 to 1,000 barg.

10 . The method according to claim 5 comprising lysing the biomass at a pH greater than 7.0 and at a temperature greater than 5 degrees centigrade.

11 . The method according to claim 10 further comprising treating the biomass with a base and lysing the biomass at a pH greater than 7.0 and at a temperature greater than 45 degrees centigrade.

12 . The method according to claim 10 further comprising treating the biomass with a base and lysing the biomass at a pH between 7.0 to 12.5 at a temperature between than 45 to 100 degrees centigrade.

13 . The method according to claim 5 wherein the lysing comprises treating the biomass with a surfactant derived from fatty acids and comprising at least one of coconut oil, palm oil, palm kernel oil, and pilu oil.

14 . The method according to claim 5 wherein the lysing comprises treating the biomass with an acyl-amino surfactant derived from fatty acids and comprising at least one of coconut oil, palm oil, palm kernel oil, and pilu oil.

15 . The method according to claim 5 further comprising lysing or treating a post-lysis broth to make the cellular debris more amenable to washing by treating the biomass with one or more of lysozyme, protease, lipase or a combination.

16 . The method according to claim 1 comprising washing the lysed biomass by treating the lysed biomass with one or more of water, acids, bases, ethanol, or a combination.

17 . The method according to claim 1 wherein the drying is selected from the group consisting of spray drying, ribbon drying, tray drying, freeze drying, drum drying, vacuum ribbon drying, refractance window drying and vacuum drum drying.

18 . The method according to claim 1 comprising dewatering the grown biomass to a concentration between 50 to 350 grams per liter (g/L).

19 . A method for producing a purified beta-1,3-glucan, comprising:

growing a biomass from genus Euglena organisms;

lysing the biomass to form a lysed biomass;

washing and dewatering the lysed biomass to produce a beta-1,3-glucan; and

drying the beta-1,3-glucan to produce the purified beta-1,3-glucan.

20 . The method according to claim 19 comprising washing and dewatering the lysed biomass a plurality of times.

21 . The method according to claim 19 wherein the dewatering comprises centrifuging or gravity decanting the lysed biomass.

22 . The method according to claim 21 wherein the centrifuging is selected from the group consisting of decanter, stacked-disk, conical plate, pusher, and peeler centrifuging.

23 . The method according to claim 19 wherein the lysing is selected from the group consisting of mechanical, pH and temperature driven, surfactant, and enzymatic lysing.

24 . The method according to claim 23 wherein the mechanical lysing comprises homogenizing or bead milling.

25 . The method according to claim 24 comprising homogenizing the biomass at a pressure greater than 500 barg.

26 . The method according to claim 24 comprising homogenizing the biomass at a pressure range of 500 to 1,900 barg.

27 . The method according to claim 24 comprising homogenizing the biomass at a pressure range of 750 to 1,000 barg.

28 . The method according to claim 23 comprising lysing the biomass at a pH greater than 7.0 and at a temperature greater than 5 degrees centigrade.

29 . The method according to claim 28 further comprising treating the biomass with a base and lysing the biomass at a pH greater than 7.0 and at a temperature greater than 45 degrees centigrade.

30 . The method according to claim 28 further comprising treating the biomass with a base and lysing the biomass at a pH between 7.0 to 12.5 at a temperature between than 45 to 100 degrees centigrade.

31 . The method according to claim 23 wherein the lysing comprises treating the biomass with a surfactant derived from fatty acids and comprising at least one of coconut oil, palm oil, palm kernel oil, and pilu oil.

32 . The method according to claim 23 wherein the lysing comprises treating the biomass with an acyl-amino surfactant derived from fatty acids and comprising at least one of coconut oil, palm oil, palm kernel oil, and pilu oil.

33 . The method according to claim 23 further comprising lysing or treating a post-lysis broth to make the cellular debris more amenable to washing by treating the biomass with one or more of lysozyme, protease, lipase or a combination.

34 . The method according to claim 19 comprising washing the lysed biomass by treating the lysed biomass with one or more of water, acids, bases, ethanol, or a combination.

35 . The method according to claim 19 wherein the drying is selected from the group consisting of spray drying, ribbon drying, tray drying, freeze drying, drum drying, vacuum ribbon drying, refractance window drying and vacuum drum drying.

36 . The method according to claim 19 wherein the biomass is grown or concentrated to a concentration between 3 to 350 grams per liter (g/L).
