Abstract

Disclosed are compounds having enhanced potency in the modulation of NMDA receptor activity. Such compounds are contemplated for use in the treatment of diseases and disorders, such as learning, cognitive activities, and analgesia, particularly in alleviating and/or reducing neuropathic pain. Orally available formulations and other pharmaceutically acceptable delivery forms of the compounds, including intravenous formulations, are also disclosed.

13 . A method of modulating the NMDA receptor comprising the step of administering a compound represented by:

wherein R 4 is H; R 1 is benzyl;

R 5 is

R 2 is H or CH 3 ;

R 3 is H or CH 3 , and stereoisomers, N-oxides or pharmaceutically acceptable salts thereof.

14 . The method of claim 1 , wherein R 2 and R 3 are CH 3 .

15 . The method of claim 1 , wherein R 2 is CH 3 and R 3 is H.

16 . The method of claim 1 , wherein the compound is represented by:

17 . A method of modulating the NMDA receptor comprising the step of administering a compound represented by:

The method of claim 1 , wherein said administration is provided to a patient suffering from at least one of cerebral ischemia, stroke, brain trauma, brain tumors, acute neuropathic pain, chronic neuropathic pain, sleep disorders, drug addiction, depression, certain vision disorders, ethanol withdrawal, anxiety, and memory and learning disabilities.

The method of claim 5 , wherein said administration is provided to a patient suffering from at least one of cerebral ischemia, stroke, brain trauma, brain tumors, acute neuropathic pain, chronic neuropathic pain, sleep disorders, drug addiction, depression, certain vision disorders, ethanol withdrawal, anxiety, and memory and learning disabilities
