Diagnostic devices and methods of use
The present invention relates to methods of diagnosing samples as well as various microfluidic, microcentrifuge and microfilter devices. In one embodiment, the present invention provides a method of diagnosing neurodegenerative diseases using mitochondrial and/or platelet samples. In another embodiment, the present invention provides a microfluidic device that selectively captures and analyzes a desired amount of target biological particle.
1. A method of processing a liquid sample from a subject, the method comprising the steps of:
obtaining a liquid sample from the subject;
adding a reagent to the liquid sample;
centrifuging the liquid sample to obtain platelet-rich plasma (PRP) fraction comprising unlysed platelets;
combining the PRP fraction with a staining solution;
concentrating the stained PRP fraction comprising unlysed platelets within a device comprising an optical measurement chamber; and
quantifying an amount or activity of a mitochondrial protein within the concentrated PRP fraction comprising unlysed platelets from the subject.
2. The method of claim 1 , wherein the reagent comprises an anticoagulant.
3. The method of claim 2 , wherein the anticoagulant comprises acid citrate dextrose.
4. The method of claim 1 , wherein concentrating the stained PRP fraction comprises centrifuging the stained PRP fraction within the device.
5. The method of claim 1 , wherein the mitochondrial protein comprises a mitochondrial electron transport chain enzyme.
6. The method of claim 5 , wherein the mitochondrial electron transport chain enzyme comprises cytochrome oxidase.
