Abstract

Receptor protein kinases (RPTKs) transmit extracellular signals across the plasma membrane to cytosolic proteins, stimulating formation of complexes that regulate key cellular functions. Over half of the known tyrosine kinases are implicated in human cancers and are therefore highly promising drug targets. A large-scale loss-of-function analysis of tyrosine kinases using RNA interference in the clinically relevant Erb-B2 positive, BT474 breast cancer cell line showed that Bruton's tyrosine kinase (BTK), a cytosolic, non-receptor tyrosine kinase that has been extensively studied for its role in B cell development, is required, in altered form, for BT474 breast cancer survival. This alternative form contains an amino-terminal extension that is also present in tumorigenic breast cells at significantly higher levels than in normal breast cells. Thus, embodiments of the invention find application in the field of cancer therapy.

Claims (21)

1. A method of treating cancer, comprising:

a) providing;

i) a subject, wherein said subject has a breast cancer cell population not located in bone marrow, wherein said breast cancer cells are capable of migrating to said bone marrow as metastatic breast cancer cells;

ii) Ibrutinib; and

b) treating said subject with said Ibrutinib wherein migration of said metastatic breast cancer cells to said bone marrow is reduced.

2. The method of claim 1 , wherein said treating comprises inhibiting growth of a secondary bone marrow metastatic breast cancer cell tumor.

3. A method of treating cancer, comprising:

a) providing;

i) a subject, wherein said subject has a prostate cancer cell population not located in bone marrow, wherein said cancer cells are capable of migrating to said bone marrow as metastatic prostate cancer cells;

ii) Ibrutinib; and

b) treating said subject with said Ibrutinib wherein migration of said metastatic prostate cancer cells to said bone marrow is reduced.

4. The method of claim 3 , wherein said treating comprises inhibiting growth of a secondary bone marrow metastatic prostate cancer cell tumor.

5. The method of claim 1 , wherein said breast cancer cell population is a primary breast cancer cell population.

6. The method of claim 1 , wherein said breast cancer cell population comprises Her-2 negative cells.

7. The method of claim 1 , wherein said breast cancer cell population comprises cells that do not overexpress Her-2.

8. The method of claim 1 , wherein said breast cancer cell population comprises Her-2 positive cells.

9. The method of claim 1 , wherein said breast cancer cell population comprises cells over-expressing Her-2.

10. The method of claim 3 , wherein said prostate cancer cell population is a primary prostate cancer cell population.

11. The method of claim 3 , wherein said prostate cancer cell population is a PTEN negative prostate cancer cell population.

12. The method of claim 1 , wherein said bone marrow is from a leg bone or a spinal vertebra bone.

13. The method of claim 3 , wherein said bone marrow is from a leg bone or a spinal vertebra bone.
