Therapeutic applications of Smad7
The invention provides methods and compositions for the treatment of inflammatory and/or tissue damage conditions. In particular, the use of Smad7 compositions delivered locally or systematically to a site of inflammation and/or tissue damage is described. Other specific embodiments concern treatment or prevention of side effects caused by radiation and/or chemotherapy, including but not limited to mucositis.
1. A method for treating one or more of skin or oral tissue damage, skin inflammation or oral ulcers in a subject in need thereof comprising:
administering to the subject in need thereof a therapeutically effective amount of a fusion protein comprising Smad7 (mothers against decapentaplegic homolog 7) fused to a protein transduction domain, wherein the protein translocates to the nucleus, and wherein the one or more of tissue damage or skin inflammation is associated with ulceration.
2. The method of claim 1 , wherein the protein comprising Smad7 accelerates epithelial cell proliferation and/or migration.
3. The method of claim 1 , wherein the subject is undergoing chemotherapy, radiotherapy, cytokine therapy, immunotherapy, hormone therapy, toxin therapy, or targeted therapy.
4. The method of claim 1 , wherein the one or more of skin or oral tissue damage or skin inflammation is the result of radiation toxicity or cancer therapy.
5. The method of claim 1 , wherein the subject has an autoimmune disease or disorder.
6. The method of claim 1 , wherein the protein comprising Smad7 is administered systemically or locally to an affected region.
7. The method of claim 1 , wherein the protein comprising Smad7 is administered topically.
8. The method of claim 1 , wherein the protein comprising Smad7 is provided in a formulation on a patch, in a gelatinous composition, in a microsphere, in a microbead or combination thereof.
9. The method of claim 1 , wherein the protein transduction domain is Tat.
10. A method for treating inflammatory or skin or oral tissue damage conditions related to one or more therapeutic treatment in a subject in need thereof comprising:
administering to the subject in need thereof a therapeutically effective amount of a fusion protein comprising Smad7 (mothers against decapentaplegic homolog 7) fused to a protein transduction domain, wherein the protein translocates to the nucleus, and wherein the therapeutic treatment is one or more of chemotherapy, radiotherapy, cytokine therapy, immunotherapy, hormone therapy, toxin therapy, or targeted therapy, and wherein the one or more of inflammatory or skin or oral tissue damage conditions is associated with ulceration.
11. The method of claim 10 , wherein the protein comprising Smad7 accelerates epithelial cell proliferation and/or migration.
12. The method of claim 10 , wherein the subject has cancer.
13. The method of claim 10 , wherein the subject has an autoimmune disorder.
14. The method of claim 10 , wherein the protein comprising Smad7 is administered systemically or locally to an affected region.
15. The method of claim 10 , wherein the protein comprising Smad7 is administered topically.
16. The method of claim 10 , wherein the protein comprising Smad7 is provided in a formulation on a patch, in a gelatinous composition, in a microsphere, in a microbead or combination thereof.
17. The method of claim 10 , wherein the protein transduction domain is Tat.
