Abstract

Sequences of a serotype 8 adeno-associated virus and vectors and host cells containing these sequences are provided. Also described are methods of using such host cells and vectors in production of rAAV particles.

Claims (14)

1. An adeno-associated virus (AAV)8 viral vector comprising an AAV8 capsid having packaged therein a heterologous gene operably linked to regulatory sequences which direct its expression, wherein the heterologous gene encodes a factor VIII protein, β-glucuronidase, alpha 1-antitrypsin, an immunoglobulin, a chimeric antibody, a humanized antibody, or a single chain antibody, wherein the AAV8 capsid comprises at least a vp3 capsid protein having the sequence of aa 204 to 737 of SEQ ID NO: 2 or at least an AAV vp3 capsid protein having an amino acid sequence produced by expression of the nucleotide sequence 2730 to 4337 of SEQ ID NO: 1, or a nucleotide sequence at least 98% or 99% identical to nucleotides 2730 to 4337 of SEQ ID NO:1.

2. The vector according to claim 1 , further comprising one or more AAV inverted terminal repeat (ITR) sequence from an AAV heterologous to AAV8.

3. The vector according to claim 2 , wherein the one or more AAV ITR is from AAV2.

4. A composition comprising the vector according to claim 1 and a pharmaceutically acceptable carrier.

5. A host cell containing the vector according to claim 1 in culture.

6. The AAV vector according to claim 1 , wherein the AAV8 vp3 nucleic acid sequence encodes amino acids 204 to 738 of SEQ ID NO: 2.

An adeno-associated virus (AAV) viral vector comprising an AAV capsid having packaged therein a heterologous gene operably linked to regulatory sequences which direct its expression, wherein the heterologous gene encodes an immunoglobulin, wherein the AAV capsid comprises at least an AAV8 vp2 capsid protein having the sequence of aa 138 to 738 of SEQ ID NO: 2, or an AAV vp2 capsid protein having an amino acid sequence produced by expression of the nucleotide sequence 2532 to 4337 of SEQ ID NO: 1, or a nucleotide sequence at least 98% or 99% identical to nucleotides 2532 to 4337 of SEQ ID NO: 1.

8. The AAV vector according to claim 7 , wherein the AAV8 vp2 nucleic acid sequence encodes amino acids 138 to 738 of SEQ ID NO: 2.

9. A composition comprising the vector according to claim 7 and at least one pharmaceutically acceptable carrier, preservative, and/or stabilizer.

10. A host cell containing the vector according to claim 7 in culture.

An adeno-associated virus (AAV) viral vector comprising an AAV capsid having packaged therein a heterologous gene operably linked to regulatory sequences which direct its expression, wherein the heterologous gene encodes an immunoglobulin, wherein the AAV capsid comprises at least an AAV8 vp1 capsid protein having the sequence of aa 1 to 738 of SEQ ID NO: 2, or an AAV vp1 capsid protein having an amino acid sequence produced by expression of the nucleotide sequence 2121 to 4337 of SEQ ID NO: 1, or a nucleotide sequence at least 98% or 99% identical to nucleotides 2121 to 4337 of SEQ ID NO:1.

12. The AAV vector according to claim 11 , wherein the AAV8 vp1 nucleic acid sequence encodes amino acids 1 to 738 of SEQ ID NO: 2.

13. A composition comprising the vector according to claim 11 and at least one pharmaceutically acceptable carrier, preservative, and/or stabilizer.

14. A host cell containing the vector according to claim 11 in culture.
