Abstract

The present invention relates to a method for the treatment or prophylaxis of a PCV2 infection or for reduction of clinical symptoms caused by or associated with a PCV2 infection in animals a) having anti-PCV2 antibodies and/or b) being young piglets of 1 to 22 days of age, comprising the step of administering an effective amount of a PCV2 antigen to that animal in need of such treatment. Preferably, those animals are pigs or young piglets.

Claims (58)

1. A method for the treatment of

a) PCV2 infection; or

b) clinical symptoms caused by or associated with a PCV2 infection in piglets having anti-PCV2 antibodies, comprising the step of administering a single dose of an immunogenic composition comprising a PCV2 ORF2 antigen and a veterinary-acceptable carrier to a piglet, wherein the single dose induces an immune response against PCV2 in the piglet.

2. The method according to claim 1 , wherein said anti-PCV2 antibodies are maternal antibodies.

3. The method according to claim 1 , wherein said treatment of PCV2 infection results in an improvement in comparison to animals of a non-treated control group of the same species in a vaccine efficacy parameter selected from the group consisting of a reduction in the loss of weight gain, a shorter duration of viremia, a reduction in viral excretion, a lower virus load, and any combination thereof.

4. The method according to claim 1 , wherein the anti-PCV2 antibodies are present in an antibody titer of at least 1:100-1:320 as determined in a PCV-specific indirect immunofluorescence assay.

5. The method according to claim 4 , wherein the treated animals have said anti-PCV2 antibody titre at the time the PCV2 antigen is administered.

6. The method of claim 1 , wherein the veterinary-acceptable carrier is an adjuvant.

7. The method of claim 1 , wherein the immunogenic composition further comprises gentamicin and/or merthiolate.

8. The method of claim 1 , wherein the immune response against PCV2 results in increased PCV2 serological titers two months after administration compared to a non-treated control group.

9. The method of claim 1 , wherein the immune response to PCV2 is a protective immunological response.

10. The method according to claim 1 , wherein the treatment results in a reduction in lymphoid infection by PCV2 as determined by immunohistochemistry.

11. A method for the treatment of

a) PCV2 infection; or

b) clinical symptoms caused by or associated with a PCV2 infection in piglets having anti-PCV2 antibodies, comprising the step of administering a single dose of an immunogenic composition comprising a PCV2 ORF2 antigen and a veterinary-acceptable carrier to a piglet, wherein the administration of the single dose is effective for the vaccination of a piglet 3 weeks of age or older against PCV2 infection.

12. The method according to claim 11 , wherein said anti-PCV2 antibodies are maternal antibodies.

13. The method according to claim 11 , wherein said treatment of PCV2 infection results in an improvement in comparison to animals of a non-treated control group of the same species in a vaccine efficacy parameter selected from the group consisting of a reduction in the loss of weight gain, a shorter duration of viremia, a reduction in viral excretion, a lower virus load, and any combination thereof.

14. The method according to claim 11 , wherein the anti-PCV2 antibodies are present in an antibody titer of at least 1:100-1:320 as determined in a PCV-specific indirect immunofluorescence assay.

15. The method according to claim 14 , wherein the treated animals have said anti-PCV2 antibody titre at the time the PCV2 antigen is administered.

16. The method of claim 11 , wherein the veterinary-acceptable carrier is an adjuvant.

17. The method of claim 11 , wherein the immunogenic composition further comprises gentamicin and/or merthiolate.

18. The method of claim 11 , wherein the vaccination results in an immune response against PCV2 that includes increased PCV2 serological titers two months after administration compared to a non-treated control group.

19. The method of claim 18 , wherein the immune response to PCV2 is a protective immunological response.

20. The method according to claim 11 , wherein the treatment results in a reduction in lymphoid infection by PCV2 as determined by immunohistochemistry.

21. A method for the treatment of

a) PCV2 infection; or

b) clinical symptoms caused by or associated with a PCV2 infection in piglets having anti-PCV2 antibodies, comprising the step of administering a single dose of an immunogenic composition comprising a PCV2 ORF2 antigen and gentamicin and/or merthiolate to a piglet and, wherein the single dose induces an immune response against PCV2 in the piglet.

22. The method according to claim 21 , wherein said anti-PCV2 antibodies are maternal antibodies.

The method according to claim 21 , wherein said treatment of PCV2 infection results in an improvement in comparison to animals of a non-treated control group of the same species in a vaccine efficacy parameter selected from the group consisting of a reduction in the loss of weight gain, a shorter duration of viremia, a reduction in viral excretion, a lower virus load, and any combination thereof.

24. The method according to claim 21 , wherein the anti-PCV2 antibodies are present in an antibody titer of at least 1:100-1:320 as determined in a PCV-specific indirect immunofluorescence assay.

25. The method according to claim 24 , wherein the treated animals have said anti-PCV2 antibody titre at the time the PCV2 antigen is administered.

26. The method of claim 21 , wherein the immunogenic composition further comprises a veterinary-acceptable carrier.

27. The method of claim 26 , wherein the veterinary-acceptable carrier is an adjuvant.

28. The method of claim 21 , wherein the immune response against PCV2 results in increased PCV2 serological titers two months after administration compared to a non-treated control group.

29. The method of claim 21 , wherein the immune response to PCV2 is a protective immunological response.

30. The method according to claim 21 , wherein the treatment results in a reduction in lymphoid infection by PCV2 as determined by immunohistochemistry.

31. A method for the treatment of

c) PCV2 infection; or

d) clinical symptoms caused by or associated with a PCV2 infection in piglets having anti-PCV2 antibodies, comprising the step of administering a single dose of an immunogenic composition comprising a PCV2 ORF2 antigen and gentamicin and/or merthiolate to a piglet, wherein the single dose is effective for the vaccination of a piglet 3 weeks of age or older against PCV2 infection.

32. The method according to claim 31 , wherein said anti-PCV2 antibodies are maternal antibodies.

33. The method according to claim 31 , wherein said treatment of PCV2 infection results in an improvement in comparison to animals of a non-treated control group of the same species in a vaccine efficacy parameter selected from the group consisting of a reduction in the loss of weight gain, a shorter duration of viremia, a reduction in viral excretion, a lower virus load, and any combination thereof.

34. The method according to claim 31 , wherein the anti-PCV2 antibodies are present in an antibody titer of at least 1:100-1:320 as determined in a PCV-specific indirect immunofluorescence assay.

35. The method according to claim 34 , wherein the treated animals have said anti-PCV2 antibody titre at the time the PCV2 antigen is administered.

36. The method according to claim 34 , wherein the immunogenic composition further comprises a veterinary-acceptable carrier.

37. The method of claim 36 , wherein the veterinary-acceptable carrier is an adjuvant.

38. The method of claim 31 , wherein the vaccination results in an immune response against PCV2 that includes increased PCV2 serological titers two months after administration compared to a non-treated control group.

39. The method of claim 38 , wherein the immune response to PCV2 is a protective immunological response.

40. The method according to claim 31 , wherein the treatment results in a reduction in lymphoid infection by PCV2 as determined by immunohistochemistry.

41. A method for the vaccination of piglets 3 weeks of age or older against PCV2 infection comprising the step of administering a single dose of an immunogenic composition comprising a PCV2 ORF2 antigen and a veterinary-acceptable carrier to a piglet, wherein the piglet has anti-PCV2 antibodies, and wherein the single dose induces an immune response against PCV2 in the piglet.

42. The method according to claim 41 , wherein said anti-PCV2 antibodies are maternal antibodies.

The method according to claim 41 , wherein said vaccination against PCV2 infection results in an improvement in comparison to piglets of a non-treated control group of piglets in a vaccine efficacy parameter selected from the group consisting of a reduction in the loss of weight gain, a shorter duration of viremia, a reduction in viral excretion, a lower virus load, and any combination thereof.

44. The method according to claim 41 , wherein the anti-PCV2 antibodies are present in an antibody titer of at least 1:100-1:320 as determined in a PCV-specific indirect immunofluorescence assay.

45. The method according to claim 44 , wherein vaccinated piglets have said anti-PCV2 antibody titre at the time the PCV2 antigen is administered.

46. The method of claim 41 , wherein the immunogenic composition further comprises gentamicin and/or merthiolate.

47. The method of claim 41 , wherein the veterinary-acceptable carrier is an adjuvant.

48. The method of claim 41 , wherein the immune response against PCV2 results in increased PCV2 serological titers two months after administration compared to a non-treated control group.

49. The method of claim 41 , wherein the immune response to PCV2 is a protective immunological response.

50. The method according to claim 41 , wherein the vaccination results in a reduction in lymphoid infection by PCV2 as determined by immunohistochemistry.
