Abstract

Agents, compositions, methods and kits useful for the treatment and diagnosis of Staphylococcal intramammary infection are disclosed. The agents, compositions, methods and kits are derived from genes expressed during Staphylococcal intramammary infection, and more particularly genes SACOL0029, SACOL0264, SACOL0442, SACOL0718, SACOL0720, SACOL1353, SACOL1416, SACOL1611, SACOL1944, SACOL2144, SACOL2365 or SACOL2599, based on the gene nomenclature from the Staphylococcus aureus COL (SACOL) genome.

Claims (39)

1. A pharmaceutical composition comprising:

(A) at least one polypeptide that is:

(a) a polypeptide encoded by SACOL1867, the polypeptide consisting of SEQ ID NO: 100;

(b) a polypeptide comprising an amino acid sequence, wherein the amino acid sequence is at least 80% identical to the full length of the polypeptide of (a); or

(c) a polypeptide comprising an immunogenic fragment of the polypeptide of (a), the fragment consisting of at least 100 consecutive amino acids of SEQ ID NO: 100;

wherein said at least one polypeptide has the ability to elicit an immune response in a mammal; and (B) an adjuvant.

2. The pharmaceutical composition of claim 1 , comprising a combination of polypeptides.

3. The pharmaceutical composition of claim 2 , wherein said combination of polypeptides further comprises (C), wherein (C) is:

(i) (a) a polypeptide encoded by SACOL0442 comprising residues 36 to 203 of the amino acid sequence of SEQ ID NO: 37 or SEQ ID NO: 48; (b) a polypeptide comprising an amino acid sequence, the amino acid sequence being at least 95% identical to the full length of the polypeptide of (a), and comprising one or more of the following amino acid sequences: KDTINGKSNKSRNW (SEQ ID NO: 18), KDGGKYTLESHKELQ (SEQ ID NO: 19), XXTINGKSNKXRNW (SEQ ID NO: 101) and KXGGKYTLESHKELQ (SEQ ID NO: 102), wherein X is any amino acid residue; (c) a polypeptide comprising an immunogenic fragment of (a), and comprising one or more of the following amino acid sequences: KDTINGKSNKSRNW (SEQ ID NO: 18), KDGGKYTLESHKELQ (SEQ ID NO: 19), XXTINGKSNKXRNW (SEQ ID NO: 101) and KXGGKYTLESHKELQ (SEQ ID NO: 102), wherein X is any amino acid residue; or (d) any combination of (a) to (c), wherein said polypeptide has the ability to elicit an immune response in a mammal; and/or

(ii) (a) a polypeptide encoded by SACOL0720 comprising residues 310 to 508 of the amino acid sequence of SEQ ID NO: 62 or SEQ ID NO: 74; (b) polypeptide comprising an amino acid sequence, the amino acid sequence being at least 95% identical to the full length of the polypeptide of (a), and comprising one or more of the following amino acid sequences: QFGFDLKHKKDALA (SEQ ID NO: 20), TIKDQQKANQLAS (SEQ ID NO: 21), KDINKIYFMTDVDL (SEQ ID NO: 22) or DVDLGGPTFVLND (SEQ ID NO: 23); (c) a polypeptide comprising an immunogenic fragment of (a), and comprising one or more of the following amino acid sequences: QFGFDLKHKKDALA (SEQ ID NO: 20), TIKDQQKANQLAS (SEQ ID NO: 21), KDINKIYFMTDVDL (SEQ ID NO: 22) or DVDLGGPTFVLND (SEQ ID NO: 23); or (d) any combination of (a) to (c), wherein said polypeptide has the ability to elicit an immune response in a mammal.

4. The pharmaceutical composition of claim 3 , wherein

(C) is: (i)(c); and/or (ii)(c).

5. The pharmaceutical composition of claim 1 , wherein said adjuvant comprises alum, an oil, cyclic-diguanosine-5′-monophosphate (c-di-GMP), polyphosphasine or pathogen-associated molecular patterns (PAMPS).

6. The pharmaceutical composition of claim 5 , wherein said PAMPS is unmethylated dinucleotides (CpG) or microbial polysaccharides.

7. The pharmaceutical composition of claim 1 , wherein said (A) at least one polypeptide, (B) adjuvant, or both (A) and (B), are comprised in a pharmaceutical composition.

8. The pharmaceutical composition of claim 7 , wherein said pharmaceutical composition further comprises one or more pharmaceutically acceptable excipients.

9. A kit for immunizing a mammal against a Staphylococcal intramammary infection (IMI), comprising

(A) at least one polypeptide that is:

(a) a polypeptide encoded by SACOL1867, the polypeptide consisting of SEQ ID NO: 100;

(b) a polypeptide comprising an amino acid sequence, wherein the amino acid sequence is at least 80% identical to the full length of the polypeptide of (a); or

(c) a polypeptide comprising an immunogenic fragment of the polypeptide of (a), the fragment consisting of at least 100 consecutive amino acids of SEQ ID NO: 100;

wherein said at least one polypeptide has the ability to elicit an immune response in a mammal; and (B) instructions to use the kit for immunizing a mammal against a Staphylococcal IMI.

10. The kit of claim 9 , comprising a combination of polypeptides.

11. The kit of claim 10 , wherein said combination of polypeptides further comprises (C), wherein (C) is:

(i) (a) a polypeptide encoded by SACOL0442 comprising residues 36 to 203 of the amino acid sequence of SEQ ID NO: 37 or SEQ ID NO: 48; (b) a polypeptide comprising an amino acid sequence, the amino acid sequence being at least 95% identical to the full length of the polypeptide of (a), and comprising one or more of the following amino acid sequences: KDTINGKSNKSRNW (SEQ ID NO: 18), KDGGKYTLESHKELQ (SEQ ID NO: 19), XXTINGKSNKXRNW (SEQ ID NO: 101) and KXGGKYTLESHKELQ (SEQ ID NO: 102), wherein X is any amino acid residue; (c) a polypeptide comprising an immunogenic fragment of (a), and comprising one or more of the following amino acid sequences: KDTINGKSNKSRNW (SEQ ID NO: 18), KDGGKYTLESHKELQ (SEQ ID NO: 19), XXTINGKSNKXRNW (SEQ ID NO: 101) and KXGGKYTLESHKELQ (SEQ ID NO: 102), wherein X is any amino acid residue; or (d) any combination of (a) to (c), wherein said polypeptide has the ability to elicit an immune response in a mammal; and/or

(ii) (a) a polypeptide encoded by SACOL0720 comprising residues 310 to 508 of the amino acid sequence of SEQ ID NO: 62 or SEQ ID NO: 74; (b) polypeptide comprising an amino acid sequence, the amino acid sequence being at least 95% identical to the full length of the polypeptide of (a), and comprising one or more of the following amino acid sequences: QFGFDLKHKKDALA (SEQ ID NO: 20), TIKDQQKANQLAS (SEQ ID NO: 21), KDINKIYFMTDVDL (SEQ ID NO: 22) or DVDLGGPTFVLND (SEQ ID NO: 23); (c) a polypeptide comprising an immunogenic fragment of (a), and comprising one or more of the following amino acid sequences: QFGFDLKHKKDALA (SEQ ID NO: 20), TIKDQQKANQLAS (SEQ ID NO: 21), KDINKIYFMTDVDL (SEQ ID NO: 22) or DVDLGGPTFVLND (SEQ ID NO: 23); or (d) any combination of (a) to (c), wherein said polypeptide has the ability to elicit an immune response in a mammal.

12. The kit of claim 11 , wherein (C) is (i) (c) and/or (ii) (c).

13. The kit of claim 9 , further comprising (D) an adjuvant.

14. The kit of claim 13 , wherein said adjuvant comprises alum, an oil, cyclic-diguanosine-5′-monophosphate (c-di-GMP), polyphosphasine or pathogen-associated molecular patterns (PAMPS).

15. The kit of claim 14 , wherein said PAMPS is unmethylated dinucleotides (CpG) or microbial polysaccharides.

16. The kit of claim 13 , wherein said (A) at least one polypeptide,

(D) adjuvant, or both (A) and (D), are comprised in a pharmaceutical composition.

17. The kit of claim 16 , wherein said pharmaceutical composition further comprises one or more pharmaceutically acceptable excipients.

18. The pharmaceutical composition of claim 1 , wherein said at least one polypeptide (A) is (c).

19. The kit of claim 9 , wherein said at least one polypeptide (A) is (c).

20. The pharmaceutical composition of claim 9 , wherein in (b), the amino acid sequence is at least 90% identical to the full length of (i).

21. The pharmaceutical composition of claim 1 , wherein said adjuvant comprises unmethylated dinucleotides (CpG).

22. The kit of claim 1 , wherein in (b), the amino acid sequence is at least 90% identical to the full length of (i).

23. The kit of claim 13 , wherein said adjuvant comprises unmethylated dinucleotides (CpG).
