Abstract

The invention, in some aspects relates to compositions and methods for altering cell activity and function and the introduction and use of light-activated ion channels.

Claims (27)

1. A method of altering ion conductivity of a membrane, the method comprising,

a) expressing in a host membrane in a cell a light-activated ion channel polypeptide comprising an amino acid sequence having at least 90% amino acid identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids of SEQ ID NO: 7, wherein the light-activated ion channel polypeptide is part of a heterologous fusion protein; and

b) contacting the light-activated ion channel polypeptide with a light that activates the light-activated ion channel and alters the ion conductivity of the membrane.

2. The method of claim 1 , wherein the activating light has a wavelength between 365 nm and 630 nm.

3. The method of claim 1 , wherein the amino acid sequence of the light-activated ion channel polypeptide comprises SEQ ID NO: 7.

4. The method of claim 1 , wherein the amino acid sequence of the light-activated ion channel polypeptide has at least 95% identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids in the sequence set forth as SEQ ID NO: 7.

5. The method of claim 1 , wherein the cell is a human cell.

6. The method of claim 1 , wherein the membrane is a cell membrane of a neuronal cell, a nervous system cell, a cardiac cell, a circulatory system cell, a visual system cell, or an auditory system cell.

7. The method of claim 1 , wherein the method has at least 99% amino acid identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids of SEQ ID NO: 7.

8. The method of claim 1 , wherein the method has at least 99% amino acid identity to amino acids 61-295 of SEQ ID NO: 7 and at least 99% identity to the remaining amino acids of SEQ ID NO: 7.

9. A method of assessing the effect of a candidate compound on ion conductivity of a membrane, the method comprising,

a) contacting a test membrane comprising a light-activated ion channel polypeptide comprising an amino acid sequence having at least 90% amino acid identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids of SEQ ID NO: 7, wherein the light activated ion channel polypeptide is part of a heterologous fusion protein with a light under conditions suitable for altering ion conductivity of the membrane, wherein the test membrane is a cell membrane;

b) contacting the test membrane with a candidate compound; and

c) identifying the presence or absence of a change in ion conductivity of the test membrane contacted with the light and the candidate compound compared to ion conductivity in a control cell membrane contacted with the light and not contacted with the candidate compound;

wherein a change in the ion conductivity in the test membrane compared to the control membrane indicates an effect of the candidate compound on the ion conductivity of the test membrane.

10. The method of claim 9 , wherein the amino acid sequence of the light-activated ion channel polypeptide has at least 95% identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids in the sequence set forth as SEQ ID NO: 7.

11. The method of claim 9 , wherein the amino acid sequence of the light-activated ion channel polypeptide has at least 99% identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids in the sequence set forth as SEQ ID NO: 7.

12. The method of claim 9 , wherein the method has at least 99% amino acid identity to amino acids 61-295 of SEQ ID NO: 7 and at least 99% identity to the remaining amino acids of SEQ ID NO: 7.

13. A light activated ion channel polypeptide comprising an amino acid sequence having at least 90% amino acid identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids of SEQ ID NO: 7, wherein the light activated ion channel polypeptide is part of a heterologous fusion protein.

14. The light activated ion channel polypeptide of claim 13 , wherein contact of the ion channel polypeptide with a light having a wavelength between 365 nm and 630 nm activates the ion channel.

15. The light activated ion channel polypeptide of claim 13 , wherein a nucleic acid sequence encoding the light-activated ion channel polypeptide comprises the nucleic acid sequence set forth as SEQ ID NO: 8.

16. The light activated ion channel polypeptide of claim 13 , wherein the amino acid sequence of the light-activated ion channel polypeptide comprises SEQ ID NO: 7.

The light-activated ion channel polypeptide of claim 13 , wherein the amino acid sequence of the light-activated ion channel polypeptide has at least 95% identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids in the sequence set forth as SEQ ID NO: 7.

18. The light activated ion channel polypeptide of claim 13 , wherein the heterologous fusion protein is in a cell and the cell is a neuronal cell, a nervous system cell, a cardiac cell, a circulatory system cell, a visual system cell, or an auditory system cell.

19. A cell comprising the light-activated ion channel polypeptide of claim 13 , wherein optionally the cell is a mammalian cell, and optionally the cell is an excitable cell.

20. The light-activated ion channel polypeptide of claim 13 , wherein the light-activated ion channel polypeptide has at least 99% amino acid identity to amino acids 61-295 of SEQ ID NO: 7 and at least 95% identity to the remaining amino acids of SEQ ID NO: 7.

21. The light-activated ion channel polypeptide of claim 13 , wherein the light-activated ion channel polypeptide has at least 99% amino acid identity to amino acids 61-295 of SEQ ID NO: 7 and at least 99% identity to the remaining amino acids of SEQ ID NO: 7.
