Antibodies and immunocytokines
The present invention relates to anti-PD-L1 antibodies, bispecific antibodies containing one domain with specificity to PD-L1, and to immunocytokines comprising an anti-PD-L1 antibody fused to a cytokine, such as IL-2. The present invention also provides methods of treatment, uses and pharmaceutical compositions comprising the antibodies, bispecific antibodies and immunocytokines.

An antibody or a fragment thereof, which specifically binds to human PD-L1 (hPD-L1) and comprises a heavy chain variable (VH) domain and a light chain variable (VL) domain, wherein the antibody or fragment thereof comprises a CDRH1 of amino acid sequence SEQ ID NO: 27 or SEQ ID NO: 30, a CDRH2 of amino acid sequence SEQ ID NO: 28 or SEQ ID NO: 31, a CDRH3 of amino acid sequence SEQ ID NO: 29 or SEQ ID NO: 32, a CDRL1 of amino acid sequence SEQ ID NO: 37 or SEQ ID NO: 40, a CDRL2 of amino acid sequence SEQ ID NO: 38 or SEQ ID NO: 41, and a CDRL3 of amino acid sequence SEQ ID NO: 39 or SEQ ID NO: 42.
2. The antibody or fragment thereof according to claim 1 , wherein the V H domain comprises an amino acid sequence of SEQ ID NO:33, and the V L domain comprises an amino acid sequence of SEQ ID NO:43.
3. The antibody or fragment thereof according to claim 1 , comprising first and second copies of said V H domain and/or said V L domain.
4. The antibody or fragment thereof according to claim 3 , comprising a kappa light chain.
5. The antibody or fragment thereof according to claim 1 , further comprising a second domain comprising a second antibody or antigen binding fragment thereof that specifically binds to a target antigen other than hPD-L1.

The antibody or fragment thereof according to claim 5 , wherein the first antibody or fragment thereof specifically binds to hPD-L1 and the second antibody or fragment thereof specifically binds another target antigen selected from the group consisting of: PD-1, CTLA-4, TIGIT, TIM-3, LAG-3, VISTA, BTLA, HVEM, CSF1R, CCR4, CD39, CD40, CD73, CD96, CXCR2, CD200, GARP, SIRPα, CXCL9, CXCL10, CD155, CD137, GITR, OX40, CXCR3, CD3, and ICOS.
7. A method of treating a malignant tumour in a human in need thereof, the method comprising administering to the human the antibody or fragment thereof according to claim 1 .

The method of claim 7 , wherein the malignant tumour is selected from the group consisting of: melanoma, Merkel cell carcinoma, squamous non-small cell lung cancer, non-squamous non-small cell lung cancer, renal cell cancer, bladder cancer, head and neck squamous cell carcinoma, mesothelioma, virally induced cancers, cervical cancer, nasopharyngeal cancer, soft tissue sarcomas, haematological malignancies, Hodgkin's disease, non-Hodgkin's disease, and diffuse large B-cell lymphoma.
9. An immunocytokine comprising the antibody or fragment thereof according to claim 1 and a cytokine, and optionally further comprising a linker between the antibody or fragment thereof and the cytokine.
10. The immunocytokine according to claim 9 , wherein the cytokine is IL-2.
11. A pharmaceutical composition comprising the antibody or fragment thereof according to claim 1 and a pharmaceutically acceptable carrier.
12. A pharmaceutical composition comprising the immunocytokine according to claim 9 and a pharmaceutically acceptable carrier.
13. The antibody or fragment thereof according to claim 1 , further comprising an IgG1 constant region of amino acid sequence SEQ ID NO: 340.
14. The antibody or fragment thereof according to claim 1 , further comprising an IgG1 constant region of amino acid sequence SEQ ID NO: 205.
