Bioengineered allogeneic blood vessel
The present invention relates to methods for recellurization of blood vessels. This method is particularly useful for producing an allogeneic vein, wherein a donor vein is decellularized and then recellularized using whole blood or bone marrow stem cells. The allogeneic veins produced by the methods disclosed herein are particularly advantageous for implantation or transplantation into patients with vascular diseases.
1. A blood vessel produced by a method comprising introducing whole blood comprising endothelial cells, smooth muscle cells, and/or progenitor cells for endothelial and smooth muscle cells from a patient to the lumen of a decellularized blood vessel from a donor, wherein the donor is not the patient, wherein the patient is human, and culturing said cells on the decellularized blood vessel, thereby recellularizing the blood vessel.
2. The blood vessel of claim 1 , wherein said introducing the whole blood to the decellularized blood vessel is by injection or perfusion.
3. The blood vessel of claim 1 , said method further comprising culturing said cells by perfusion of endothelial cell medium and/or smooth muscle cell medium.
4. The blood vessel of claim 3 , wherein said perfusion of said endothelial cell medium and said smooth muscle cell medium are in alternation.
5. The blood vessel of claim 4 , wherein said perfusion of said medium in alternation is repeated at least twice.
6. The blood vessel of claim 1 , wherein said culturing the cells on the decellularized blood vessel results in proliferation and/or differentiation of the cells to endothelial cells and smooth muscle cells.
7. The blood vessel of claim 6 , wherein said endothelial cells line the inner lining or the lumen of the decellularized blood vessel and said smooth muscle cells migrate to the walls or are present over the middle and outer layers of the decellularized blood vessel.
8. The blood vessel of claim 7 , wherein said endothelial cells express VE-cadherein, AcLDL, vWF or CD31.
9. The blood vessel of claim 7 , wherein said smooth muscle cells express smooth muscle actin or vimentin.
10. The blood vessel of claim 1 , wherein said blood vessel is a vein or artery.
