Abstract

Provided herein are compounds of formula (I) which are, inter alia, useful allosteric inhibitors of Hsp70. The compounds and methods provided are useful for the treatment of cancer, infectious and neurodegenerative diseases.

Claims (55)

1. A compound of formula:

wherein:

R 1 is hydrogen, halogen, —CX a 3 , —CN, —SR 9 , —SO 2 Cl, —SO n1 R 9 , —SO v1 NR 9 R 10 , —NHNH 2 , —ONR 9 R 10 , —NHC═(O)NHNH 2 , —NHC═(O)NR 9 R 10 , —N(O) m1 , —NR 9 R 10 , —NH—O—R 9 , —NHC(O)R 9 , —C(O)R 9 , —C(O)—OR 9 , —C(O)NR 9 R 10 , —OR 9 , substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein each substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl is substituted with at least one substituent group;

R 2 is hydrogen, halogen, —CX b 3 , —CN, —SR 11 , —SO 2 Cl, —SO n2 R 11 , —SO v2 NR 11 R 12 , —NHNH 2 , —ONR 11 R 12 , —NHC═(O)NHNH 2 , —NHC═(O)NR 11 R 12 , —N(O) m2 , —NR 11 R 12 , —NH—O—R 11 , —NHC(O)R 11 , —C(O)R 11 , —C(O)—OR 11 , —C(O)NR 11 R 12 , —OR 11 , substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein each substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl is substituted with at least one substituent group;

R 3 is hydrogen, halogen, —CX c 3 , —CN, —SR 13 , —SO 2 Cl, —SO n3 R 13 , —SO v3 NR 13 R 14 , —NHNH 2 , —ONR 13 R 14 , —NHC═(O)NHNH 2 , —NHC═(O)NR 13 R 14 , —N(O) m3 , —NR 13 R 14 , —NH—O—R 13 , —NHC(O)R 13 , —C(O)R 13 , —C(O)—OR 13 , —C(O)NR 13 R 14 , —OR 13 , substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein each substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl is substituted with at least one substituent group;

R 8 is hydrogen, halogen, —CX d 3 , —CN, —SR 15 , —SO 2 Cl, —SO n4 R 15 , —SO v4 NR 15 R 16 , —NHNH 2 , —ONR 15 R 16 , —NHC═(O)NHNH 2 , —NHC═(O)NR 15 R 16 , —N(O) m4 , —NR 15 R 16 , —NH—O—R 15 , —NHC(O)R 15 , —C(O)R 15 , —C(O)—OR 15 , —C(O)NR 15 R 16 , —OR 15 , substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein each substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl is substituted with at least one substituent group;

R 4 is unsubstituted alkyl, unsubstituted heteroalkyl, unsubstituted cycloalkyl, unsubstituted heterocycloalkyl, unsubstituted aryl, or unsubstituted heteroaryl;

R 5 is hydrogen, halogen, —CX e 3 , —CN, —SR 17 , —SO 2 Cl, —SO n5 R 17 , —SO v5 NR 17 R 18 , —NHNH 2 , —ONR 17 R 18 , —NHC═(O)NHNH 2 , —NHC═(O)NR 17 18 , —NH—O—R 17 , —NHC(O)R 17 , —C(O)R 17 , —C(O)—OR 17 , —C(O)NR 17 R 18 , —OR 17 , substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein each substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl is substituted with at least one substituent group;

R 6 is R 6A -substituted cycloalkyl, R 6A -substituted heterocycloalkyl, R 6A -substituted aryl or R 6A -substituted heteroaryl;

R 6A is independently halogen, —CX f 3 , —CN, —SR 19 , —SO 2 Cl, —SO n6 R 19 , —SO v6 NR 19 R 20 , —NHNH 2 , —ONR 19 R 20 , —NHC═(O)NHNH 2 , —NHC═(O)NR 19 R 20 , —N(O) m6 , —NR 19 R 20 , —NH—O—R 19 , —NHC(O)R 19 , —C(O)R 19 , —C(O)—OR 19 , —C(O)NR 19 R 20 , —OR 19 , unsubstituted alkyl, R 6B -substituted or unsubstituted heteroalkyl, R 6B -substituted or unsubstituted cycloalkyl, R 6B -substituted or unsubstituted heterocycloalkyl, R 6B -substituted or unsubstituted aryl, or R 6B -substituted or unsubstituted heteroaryl;

R 6B is hydrogen, halogen, ═O, ═S, —CF 3 , —CN, —CCl 3 , —COOH, —CH 2 COOH, —CONH 2 , —OH, —SH, —SO 2 Cl, —SO 3 H, —SO 4 H, —SO 2 NH 2 , —NO 2 , —NH 2 , —NHNH 2 , —ONH 2 , —NHC═(O)NHNH 2 , unsubstituted alkyl, unsubstituted heteroalkyl, unsubstituted cycloalkyl, unsubstituted heterocycloalkyl, unsubstituted aryl, or unsubstituted heteroaryl;

R 7 is hydrogen, halogen, —CX g 3 , —CN, —SR 21 , —SO 2 Cl, —SO n7 R 21 , —SO v7 NR 21 R 22 , —NHNH 2 , —ONR 21 R 22 , —NHC═(O)NHNH 2 , —NHC═(O)NR 21 R 22 , —N(O) m7 , —NR 21 R 22 , —NH—O—R 21 , —NHC(O)R 21 , —C(O)R 21 , —C(O)—OR 21 , —C(O)NR 21 R 22 , —OR 21 , substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein each substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl is substituted with at least one substituent group;

L 1 is a bond, —S(O)—, —S(O) 2 NH—, —NHS(O) 2 —, —C(O)O—, —OC(O)—, —C(O)—, —C(O)NH—, —NH—, —NHC(O)—, —O—, —S—, R L1 -substituted or unsubstituted alkylene, R L1 -substituted or unsubstituted heteroalkylene, R L1 -substituted or unsubstituted cycloalkylene, R L1 -substituted or unsubstituted heterocycloalkylene, R L1 -substituted or unsubstituted arylene, or R L1 -substituted or unsubstituted heteroarylene;

R L1 is hydrogen, halogen, ═O, ═S, —CF 3 , —CN, —CC 3 , —COOH, —CH 2 COOH, —CONH 2 , —OH, —SH, —SO 2 Cl, —SO 3 H, —SO 4 H, —SO 2 NH 2 , —NO 2 , —NH 2 , —NHNH 2 , —ONH 2 , —NHC═(O)NHNH 2 , unsubstituted alkyl, unsubstituted heteroalkyl, unsubstituted cycloalkyl, unsubstituted heterocycloalkyl, unsubstituted aryl, or unsubstituted heteroaryl;

R 9 , R 10 , R 11 , R 12 , R 13 , R 14 , R 15 , R 16 , R 17 , R 18 , R 19 , R 20 , R 21 , and R 22 are independently is hydrogen, halogen, ═O, ═S, —CF 3 , —CN, —CCl 3 , —COOH, —CH 2 COOH, —COCH 3 , —CONH 2 , —OH, —OC(O)CH 3 , —SH, —SO 2 Cl, —SO 3 H, —SO 4 H, —SO 2 NH 2 , —NO 2 , —NH 2 , —NHNH 2 , —ONH 2 , —NHC═(O)NHNH 2 , substituted or unsubstituted alkyl, substituted or unsubstituted heteroalkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocycloalkyl, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl, wherein each substituted alkyl, substituted heteroalkyl, substituted cycloalkyl, substituted heterocycloalkyl, substituted aryl, or substituted heteroaryl is substituted with at least one substituent group;

X a , X b , X c , X d , X e , X f and X g are independently —F, —Cl, —Br, or —I;

n 1 , n 2 , n 3 , n 4 , n 5 , n 6 and n 7 are independently an integer from 0 to 4;

m 1 , m 2 , m 3 , m 4 , m 5 , m 6 and m 7 are independently an integer from 1 to 2; and

v 1 , v 2 , v 3 , v 4 , v 5 , v 6 and v 7 are independently an integer from 1 to 2;

wherein if R 1 , R 2 , and R 3 are independently hydrogen, —Cl, —F, —OCH 3 , or —CF 3 , then —L 1 —R 6 is not —CH 2 pyridyl, -benzyl,—CH 2 -difluorophenyl, —CH 2 -cyclopropyl, —CH 2 -4-(CH 2 NHC(O)-tbutyl)phenyl, —CH 2 -5-nitrofuranyl, —CH 2 CH 2 -5-nitrofuranyl, —CH 2 -2-(5-CF 3 )furanyl, —CH 2 -fluorophenyl, —CH 2 -chlorophenyl, —CH 2 -nitrophenyl, —CH 2 -cyanophenyl, —CH(CH 3 )C(O)Ph, —CH 2 -(methyl)phenyl, —CH 2 -trifluoromethylphenyl, —CH 2 -trifluoromethoxyphenyl, —CH 2 -difluoromethoxyphenyl, —CH 2 -3-(2-CO 2 CH 3 )thienyl, —CH 2- 3-(2-bromo)thienyl, —CH 2 -3-isoxazolyl, —CH 2 -5-isoxazolyl, —CH 2 -5-(3-phenyl)isoxazolyl, —CH 2 -3-(2-bromo)pyridyl, —CH 2 -3-thienyl, —CH 2 -2-(5-CO 2 CH 2 CH 3 )furanyl, —CH 2 -4-(2-methyl)thiazolyl, —CH 2 -2-(5-CO 2 CH 3 )furanyl, —CH 2 -5-(3-methyl)isoxazolyl, or —CH 2 —CH(CH 3 )phenyl.

2. The compound of claim 1 , wherein if L 1 is —CH 2 — and R 6 is phenyl, then R 6A is not —F, —CF 3 , —OCHF 2 , —NO 2 , —OCF 3 , —CH 3 , —CN, or —Cl.

3. The compound of claim 1 , wherein if L 1 is —CH 2 — and R 6 is furanyl, then R 6A is not —CF 3 , —NO 2 , —C(O)OCH 3 or —C(O)OCH 2 CH 3 .

4. The compound of claim 1 , wherein if L 1 is —CH 2 — and R 6 is thiophene, then R 6A is not —Br, —C(O)OCH 3 , or —C(O)OCH 2 CH 3 .

5. The compound of claim 1 , wherein if L 1 is —CH 2 — and R 6 is oxazolyl, then R 6A is not methyl or phenyl.

6. The compound of claim 1 , wherein if L 1 is —CH 2 — and R 6 is pyridyl, then R 6A is not —Br.

7. The compound of claim 1 , wherein if L 1 is —CH 2 — and R 6 is thiazolyl, then R 6A is not —CH 3 or —C(O)OCH 3 .

8. The compound of claim 1 , wherein R 1 is —Br, —SR 9 , —OR 9 , —NO 2 , —CN, or substituted or unsubstituted C 1 -C 10 alkyl.

9. The compound of claim 1 , wherein R 2 is —Br, —SR 11 , —OR 11 , —NO 2 , —CN, or substituted or unsubstituted C 1 -C 10 alkyl.

10. The compound of claim 1 , wherein R 3 is —Br, —SR 13 , —OR 13 , —NO 2 , —CN, or substituted or unsubstituted C 1 -C 10 alkyl.

11. The compound of claim 1 , wherein R 8 is —Br, —CN, —SR 15 , —OR 15 , —NR 15 R 16 , or substituted or unsubstituted C 1 -C 10 alkyl.

12. The compound of claim 1 , wherein R 4 is substituted or unsubstituted C 1 -C 10 alkyl.

13. The compound of claim 1 , wherein R 5 is halogen or substituted or unsubstituted C 1 -C 10 alkyl.

14. The compound of claim 1 , wherein R 6 is R 6A -substituted heterocycloalkyl, R 6A -substituted aryl or R 6A -substituted heteroaryl.

15. The compound of claim 1 , wherein R 6 is R 6A -substituted phenyl.

16. The compound of claim 15 , wherein R 6A is —Br, —SR 19 , —SO n6 R 19 , —C(O)—OR 19 , —OR 19 , unsubstituted alkyl or substituted or unsubstituted aryl.

17. The compound of claim 16 , wherein R 19 is substituted C 1 alkyl.

18. The compound of claim 16 , wherein R 19 is trifluoromethyl.

19. The compound of claim 16 , wherein R 19 is methyl or ethyl.

20. The compound of claim 16 , wherein R 19 is hydrogen.

21. The compound of claim 1 , wherein R 6 is R 6A -substituted furanyl.

22. The compound of claim 21 , wherein R 6A is —Br, —SR 19 , —SO n6 R 19 , —C(O)—OR 19 , —OR 19 , unsubstituted alkyl or substituted or unsubstituted aryl.

23. The compound of claim 1 , wherein R 6 is R 6A -substituted oxazolyl.

24. The compound of claim 23 , wherein R 6A is trifluoromethyl, —Br, —SR 19 , —SO n6 R 19 , —C(O)—OR 19 , —OR 19 , unsubstituted alkyl or substituted or unsubstituted aryl.

25. The compound of claim 1 , wherein R 6 is R 6A -substituted pyridyl.

26. The compound of claim 25 , wherein R 6A is —Cl, —F. —CX f 3 , —SR 19 , —SO n6 R 19 , —C(O)—OR 19 , —OR 19 , unsubstituted alkyl or substituted or unsubstituted aryl.

27. The compound of claim 1 , wherein R 6 is R 6A -substituted thiazole.

28. The compound of claim 27 , wherein R 6A is —Cl or —NR 19 R 20 .

29. The compound of claim 1 , wherein R 7 is substituted or unsubstituted C 1 -C 5 alkyl.

30. The compound of claim 1 , wherein L 1 is substituted or unsubstituted C 1 -C 10 alkylene.

31. The compound of claim 1 , wherein L 1 is methylene or ethylene.

32. A pharmaceutical composition comprising a pharmaceutically acceptable excipient and a compound of claim 1 .

33. A method of treating a Hsp70-mediated disease in a patient in need of such treatment, said method comprising administering a therapeutically effective amount of a compound of claim 1 , wherein the Hsp70-mediated disease is cancer, an infectious disease or a neurodegenerative disease.

34. The method of claim 33 , wherein said cancer is acute T-cell leukemia, breast cancer, multiple myeloma, malignant melanoma, ovarian cancer, colorectal adenocarcinoma, endometrial cancer, cervical cancer or bladder cancer.

35. The method of claim 33 , wherein said neurodegenerative disease is Kennedy's disease or Alzheimer's disease.

36. The method of claim 33 , wherein said infectious disease is Dengue fever, a Hepatitis C virus (HCV) disease, or influenza.
