Liposomal preparations for non-invasive-prenatal or cancer screening
Disclosed are controls for use in identifying any of a number of genotypes and/or for use in identifying or characterizing a disease or condition. The controls may be particularly useful for diagnostic tests that utilize circulating cell-free DNA or microRNA. A control may comprise a mixture of nucleic acids. In some embodiments, a control comprises liposomes, e.g., wherein nucleic acids of the control are associated with the liposomes. For example, the controls are useful for determining whether a fetus comprises an aneuploidy. Also disclosed are methods of using the controls.
1. A control for use in determining the ploidy of a chromosome in a fetus, comprising:
a first mixture of nucleic acids comprising a first plurality of nucleotide sequences and a second plurality of nucleotide sequences, wherein each nucleotide sequence of the first plurality has sequence homology with a chromosome; each nucleotide sequence of the second plurality has sequence homology with at least one autosome, wherein the at least one autosome does not comprise the chromosome; and the ratio of the copy number for any nucleotide sequence in the first plurality to the copy number for any nucleotide sequence in the second plurality is about 3:2;
a second mixture of nucleic acids comprising a first plurality of nucleotide sequences and a second plurality of nucleotide sequences, wherein the ratio of the copy number for any nucleotide sequence in the first plurality to the copy number for any nucleotide sequence in the second plurality is about 1:1; and
liposomes;
wherein:
the first plurality of nucleotide sequences of the first mixture and the first plurality of nucleotide sequences of the second mixture consist of the same nucleotide sequences;
the second plurality of nucleotide sequences of the first mixture and the second plurality of nucleotide sequences of the second mixture consist of the same nucleotide sequences;
the first mixture of nucleic acids and the second mixture of nucleic acids are admixed in the control;
the first mixture of nucleic acids and the second mixture of nucleic acids are incorporated into the liposomes;
the first mixture of nucleic acids encodes substantially all of a first human genome, and the second mixture of nucleic acids consists of circulating, cell-free DNA obtained from a human donor; and
the average diameter of the liposomes is about 30 nm to about 300 nm.
2. The control of claim 1 , wherein each of the at least one autosome is independently selected from the group consisting of human chromosome 1, 6, and 7.
3. The control of claim 1 , wherein the chromosome is human chromosome 8, 9, 13, 18, 21, 22, or X.
4. The control of claim 1 , wherein the first mixture of nucleic acids encodes substantially all of a first human genome and the second mixture of nucleic acids encodes substantially all of a second human genome.
5. The control of claim 4 , wherein the first human genome is a cytotrophoblast cell line genome.
6. The control of claim 1 , further comprising blood serum.
7. The control of claim 1 , wherein the first plurality of nucleotide sequences of the first mixture encodes substantially all of the chromosome.
8. The control of claim 1 , wherein the second plurality of nucleotide sequences of the first mixture encodes substantially all of at least one autosome that is not the chromosome.
9. The control of claim 8 , wherein the second plurality of nucleotide sequences of the first mixture encodes substantially all of human chromosome 1, 6, or 7.
10. The control of claim 1 , wherein the concentration of nucleic acids in the first mixture relative to the total concentration of nucleic acids in the control is about 1% to about 20% by weight.
11. The control of claim 10 , wherein the concentration of nucleic acids in the first mixture relative to the total concentration of nucleic acids in the control is about 4% to about 18% by weight; and the concentration of nucleic acids in the second mixture relative to the total concentration of nucleic acids in the control is about 82% to about 96% by weight.
12. The control of claim 1 , wherein the nucleic acids of the control are less likely to aggregate than nucleic acids of a control that does not comprise liposomes.
13. The control of claim 1 , wherein the nucleic acids of the control are less likely to aggregate than a nucleic acid of a control that does not comprise liposomes when stored at a temperature between 2° C. to 42° C. for a period of about one month to about four months.
