Abstract

One aspect of the invention provides a composition of novel high penetration compositions (HPC) or a high penetration prodrug (HPP) for treatment of Parkinson's disease. The HPCs/HPPs are capable of being converted to parent active drugs or drug metabolites after crossing the biological barrier and thus can render treatments for the conditions that the parent drugs or metabolites can. Additionally, the HPPs are capable of reaching areas that parent drugs may not be able to access or to render a sufficient concentration at the target areas and therefore render novel treatments. The HPCs/HPPs can be administered to a subject through various administration routes, e.g., locally delivered to an action site of a condition with a high concentration or systematically administered to a biological subject and enter the general circulation with a faster rate.

Claims (38)

1. A compound of

HA is a pharmaceutically acceptable acid;

X is selected from O, S, and NR 3 ;

X 2 is selected from nothing, O, S, NR 3 , CHR 3 —O, CHR 3 —S, CHR 3 —O, O—CHR 3 —O, O—CHR 3 —S, S—CHR 3 O, and S—CHR 3 —S;

X 3 is selected from nothing, C═O, C═S, C(═O)—O,O, S, NR 3 , C(═O)—O—CHR 3 —O, C(═O)—O—CHR 3 —S, C(═O)—S—CHR 3 —O, and C(═O)—S—CHR 3 —S;

Y 1 is selected from R 3 C(═O), R 3 O—C(═O), and R 3 S—C(═O);

Y 2 is selected from R 3 C(═O), R 3 O—C(═O), and R 3 S—C(═O);

Y 3 is selected from R 3 , OR 3 , SR 3 , NR 3 R 4 , O—CHR 3 —OR 4 , O—CHR 3 —SR 4 , and S—CHR 3 —OR 4 ;

n and m are independently selected from 0, 1, 2, 3, 4, 5, 6, 7, and 8;

Rc is selected from nothing, CH 2 C(═O)OR 6 , substituted and unsubstituted C 1 -C 12 alkylene, substituted and unsubstituted C 3 -C 12 cycloalkylene, substituted and unsubstituted C 3 -C 12 heterocycloalkylene, substituted and unsubstituted C 1 -C 12 alkoxylene, substituted and unsubstituted C 1 -C 12 perfluoroalkylene, substituted and unsubstituted C 1 -C 12 alkylene halide, substituted and unsubstituted C 2 -C 12 alkenylene, substituted and unsubstituted C 2 -C 12 alkynylene, substituted and unsubstituted arylene, and substituted and unsubstituted heteroarylene, wherein any CH 2 in Rc may be replaced with O, S, P, or NR 6 ;

R is selected from CH 2 C(═O)OR 6 , substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted C 1 -C 12 alkoxyl, substituted and unsubstituted C 1 -C 12 perfluoroalkyl, substituted and unsubstituted C 1 -C 12 alkyl halide, substituted and unsubstituted C 2 -C 12 alkenyl, substituted and unsubstituted C 2 -C 12 alkynyl, substituted and unsubstituted aryl, and substituted and unsubstituted heteroaryl, wherein any CH 2 in R may be replaced with O, S, P, or NR 6 ;

R 3 and R 4 are independently selected from CH 2 C(═O)OR 6 , substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted C 1 -C 12 alkoxyl, substituted and unsubstituted C 1 -C 12 perfluoroalkyl, substituted and unsubstituted C 1 -C 12 alkyl halide, substituted and unsubstituted C 2 -C 12 alkenyl, substituted and unsubstituted C 2 -C 12 alkynyl, substituted and unsubstituted aryl, and substituted and unsubstituted heteroaryl, wherein any CH 2 in R 3 or R 4 may be further replaced with O, S, P, or NR 6 ;

each R 6 is independently selected from H, F, Cl, Br, I, Na + , K + , C(═O)R 5 , 2-oxo-1-imidazolidinyl, phenyl, 5-indanyl, 2,3-dihydro-1H-inden-5-yl, 4-hydroxy-1,5-naphthyridin-3-yl, substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted C 2 -C 12 alkenyl, substituted and unsubstituted C 2 -C 12 alkynyl, substituted and unsubstituted C 1 -C 12 alkyloxyl, substituted and unsubstituted C 3 -C 12 cycloalkyloxyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, —C(═O)—W, -L 1 -L 4 -L 2 -W, and W;

each R 5 is independently selected from H, C(═O)NH 2 , CH 2 CH 2 OR 6 , CH 2 CH 2 N(CH 3 ) 2 , CH 2 CH 2 N(CH 2 CH 3 ) 2 , Cl, F, Br, I, substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted C 1 -C 12 alkyloxyl, substituted and unsubstituted C 3 -C 12 cycloalkyloxyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, substituted and unsubstituted C 1 -C 12 alkylcarbonyl, substituted and unsubstituted C 1 -C 12 alkylamino, —C(═O)—W, L 1 -L 4 -L 2 -W, and W;

L 1 is selected from nothing, O, S, —O-L 5 -, —S-L 5 -, —N(L 3 )-, —N(L 3 )-CH 2 —O, —N(L 3 )-CH 2 —N(L 3 )-, —O—CH 2 —O—, —O—CH(L 3 )-O, and —S—CH(L 3 )-O—;

L 2 is selected from nothing, O, S, —O-L 5 -, —S-L 5 -, —N(L 3 )-, —N(L 3 )-CH 2 —O, —N(L 3 )-CH 2 —N(L 3 )-, —O—CH 2 —O—, —O—CH(L 3 )-O, —S—CH(L 3 )-O—, —N(L 3 )-L 5 - and L 5 ;

L 4 is selected from nothing,

for each of L 1 , L 2 , and L 4 , each L 3 is independently selected from H, CH 2 C(═O)OL 6 , substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, substituted and unsubstituted C 1 -C 12 alkoxyl, substituted and unsubstituted C 1 -C 12 alkylthio, substituted and unsubstituted C 1 -C 12 alkylamino, substituted and unsubstituted C 1 -C 12 perfluoroalkyl, and substituted and unsubstituted C 1 -C 12 alkyl halide, wherein any carbon or hydrogen may be further independently replaced with O, S, or P;

for each of L 1 , L 2 , and L 4 , each L 5 is independently selected from CH 2 C(═O)OL 6 , substituted and unsubstituted C 1 -C 12 alkylene, substituted and unsubstituted C 3 -C 12 cycloalkylene, substituted and unsubstituted C 3 -C 12 heterocycloalkylene, substituted and unsubstituted arylene, substituted and unsubstituted heteroarylene, substituted and unsubstituted C 1 -C 12 alkoxylene, substituted and unsubstituted C 1 -C 12 alkylenethio, substituted and unsubstituted C 1 -C 12 alkyleneamino, substituted and unsubstituted C 1 -C 12 perfluoroalkylene, and substituted and unsubstituted C 1 -C 12 alkylene halide, wherein any carbon or hydrogen may be further independently replaced with O, S, or P;

each L 6 is independently selected from H, OH, Cl, F, Br, I, substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, and substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, substituted and unsubstituted C 1 -C 12 alkoxyl, substituted and unsubstituted C 1 -C 12 alkylthio, substituted and unsubstituted C 1 -C 12 alkylamino, substituted and unsubstituted C 1 -C 12 perfluoroalkyl, and substituted and unsubstituted C 1 -C 12 alkyl halide, wherein any carbon or hydrogen may be independently replaced with O, S, N, CH═CH, C≡C, CHL 6 , CL 6 L 7 , aryl, heteroaryl, or cyclic groups;

each L 7 is independently selected from H, OH, Cl, F, Br, I, substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, and substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted aryl, substituted and unsubstituted heteroaryl, substituted and unsubstituted C 1 -C 12 alkoxyl, substituted and unsubstituted C 1 -C 12 alkylthio, substituted and unsubstituted C 1 -C 12 alkylamino, substituted and unsubstituted C 1 -C 12 perfluoroalkyl, and substituted and unsubstituted C 1 -C 12 alkyl halide, wherein any carbon or hydrogen may be further independently replaced with O, S, N, P(O)OL 6 , CH═CH, C≡C, CHL 6 , CL 6 L 7 , aryl, heteroaryl, or cyclic groups;

R 1 is selected from H, substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted C 1 -C 12 alkyloxyl, substituted and unsubstituted C 2 -C 12 alkenyl, substituted and unsubstituted C 2 -C 12 alkynyl, substituted and unsubstituted aryl and substituted and unsubstituted heteroaryl residues;

R 2 is selected from substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted C 1 -C 12 alkyloxyl, substituted and unsubstituted C 2 -C 12 alkenyl, substituted and unsubstituted C 2 -C 12 alkynyl, substituted and unsubstituted aryl and substituted and unsubstituted heteroaryl residues;

R 11 —R 15 are independently selected from nothing, substituted and unsubstituted C 1 -C 12 alkyl, substituted and unsubstituted C 3 -C 12 cycloalkyl, substituted and unsubstituted C 3 -C 12 heterocycloalkyl, substituted and unsubstituted C 1 -C 12 alkoxyl, substituted and unsubstituted C 1 -C 12 perfluoroalkyl, substituted and unsubstituted C 1 -C 12 alkyl halide, substituted and unsubstituted C 2 -C 12 alkenyl, substituted and unsubstituted C 2 -C 12 alkynyl, substituted and unsubstituted aryl, and substituted and unsubstituted heteroaryl.

2. The compound according to claim 1 , wherein the compound is of Structure Pro-L-Dopa-2.

3. The compound according to claim 1 , wherein the compound is of Structure Pro-L-Dopa-3.

4. The compound according to claim 1 , wherein the compound is of Structure Pro-L-Dopa-4.

5. The compound according to claim 1 , wherein the compound is of Structure Pro-L-Dopa-5.

6. The compound according to claim 1 , wherein the compound is of Structure Pro-dopamine-2.

7. The compound according to claim 1 , wherein the compound is of Structure Pro-dopamine-3.

8. The compound according to claim 1 , wherein the compound is of Structure Pro-dopamine-4.

9. The compound according to claim 1 , wherein the compound is of Structure Pro-dopamine-5.

10. The compound according to claim 1 , wherein W is Structure W-1.

11. The compound according to claim 1 , wherein W is Structure W-2.

12. The compound according to claim 1 , wherein W is Structure W-4.

13. The compound according to claim 1 , wherein W is Structure W-1.
